Light-Guided Flow Cell Technology

Small bore high-capacity columns, as used in ACQUITY UPLC, produce small volume peaks. To avoid bandpreading and maintain concentration, the detector’s flow cell volume must be correspondingly small. Additionally, high light levels are needed to preserve a high signal-to-noise ratio.

The ACQUITY UPLC TUV Detector’s uniquely designed light-guided flow cell was created with optimum path length and high light throughput in mind. The cell uses Teflon AF, an internally reflective surface, to improve light transmission efficiency by eliminating internal absorption. This design complements and maintains the significant advantages gained by the use of the ACQUITY UPLC fluidic technology.

Light rays entering the liquid core of the flow cell are internally reflected when they meet the Teflon AF boundary. These rays are transmitted through the cell without loss, except for absorption by the sample.

Speed

High-throughput laboratories have a constant need to generate more information and process more samples, while maintaining the quality of the data. In today’s laboratories, Ultra Performance LC systems can generate peaks less than 0.5 seconds wide and analysis times as little as half a minute long.

The ACQUITY UPLC TUV Detector has been holistically designed with the ACQUTY UPLC System to offer a wide variety of sampling rates, from 1 to 80 Hz, that fit the requirements of any ultra-fast, ultra performance application, while still conforming to the foundations of sensitivity and resolution. Effective integration algorithms for ultra-fast UPLC separations, as well as independent optimization of time constants, allow for accurate and reproducible integration.

Detector data acquisition rates must be fast enough to enable detection of narrow UPLC peaks. Accurate integration at slower rates would be impossible.

Over the years, the pursuit of increased sensitivity has been driven by the need to detect compounds at increasingly lower levels. The ACQUITY UPLC TUV Detector has been designed, from the ground up, to provide the highest performance in UV/Visible detection for UPLC applications.

The ACQUITY UPLC TUV detector offers both the superior sensitivity and the linear dynamic range needed for the detection of even the lowest-level impurities allowing for the simultaneous quantitation of major and minor compounds in both routine and complex, ultra-fast UPLC separations.

Linearity

The ACQUITY UPLC TUV has a linear dynamic range that extends more than 3.5 orders of magnitude allowing for the identification and quantitation of both low level impurities (< 0.01%) and their parent compounds over a wide concentration range.

Additionally, the unique design of ACQUITY UPLC light-guided flow cells delivers the same level of quality data regardless of flow cell volume and pathlength.

Console and Software

The ACQUITY UPLC TUV Detector is equipped with a customizable instrument console, for both Empower™ and MassLynx™ software that enables users to easily stay in stay control of the detector. Interactions that were historically done through an instrument keypad—instrument setup, status monitoring, and diagnostics—are now easily accessible through an intuitive, easy-to-learn software interface.

The console uses a simple navigational approach, combined with instrument default parameters, defined range settings, tool tips, and online help, that makes system navigation and instrument usability a snap. Furthermore, at any time the control panel can display the operating status of the module providing assurance that the system is working properly.

The console of the ACQUITY UPLC TUV gives users accessibility to all primary detector functions through a single, intuitive software interface.
