Cytonix Slides and Other Analysis Methods

• Study conducted at the London Royal Veterinary College.

• Compare 6 methods for determining sperm concentration:

• Flow-Cytometry (Flow)• Hemocytometry (Hem)• Image-Analysis (Image)• Cytonix Chambers (MicroX)• Plate-Readers (Plate)• Spectrophotometry (Spec).

• The results show that Cytonix chambers showed good correlation (>0.8) with other cell counting methods

Sperm Mobility• Study conducted at the Wayne State University,

Michigan

• Sperm mobility and type of sperm movement can be assessed reliably in Cytonix counting chambers due to a unique coating in the devices.

Repeatability of Sperm Counts• Repeatability of sperm counting methods was studied by evaluating

the coefficients of Variation (CV) across 5 repeats:

• Flow-Cytometry (2.3%)• Spectrophotometer (4.1%)• Hemocytometer (7.8%)• Cytonix-Counters (10.7%)• Image-Analysis (14.9%)• Fluorescent Plate Reader (20.0%).

• Although Cytonix-Counters did not have the lowest CV, they have the advantage of allowing sperm motility assessment and sperm counting at the same time

Boar Sperm Pre-freeze, Post-Thaw and Post-Thaw Progressive Motility

• University of Illinois thesis studied boar sperm motility prior to freezing and after thaw using a Spermvision system and Cytonix 20 µm depth counting chambers

• The results showed average measures of ~73.1 x 109 cells per ejaculate, ~82% Pre-freeze Motility, ~55% Post-Thaw Motility and ~47% Post-Thaw Progressive Motility. The methods and results from this work showed good acceptable pregnancy rates and litter sizes from frozen-thawed boar sperm.
