    1.1. Cc phng php chuyn gen

    Cho n nay c hn 150 loi thc vt khc nhau, trong rt nhiuloi cy trng c chuyn gen thnh cng.     1.1.1. Chuyn gen bng vi khun Agrobacterium tumefaciens

    Qu trnh ny phc tp nn khng thch hp cho vic ng dng trongcng ngh gen v c ba nguyn nhn sau:

    - Vic to m do nhng gen tng hp cytokinin v auxin khng th tisinh t t bo thnh nhng cy hon chnh v khe mnh.

    - S tng hp opine l khng mong mun v cy tiu tn nng lngkhng cn thit.

    - DNA l khng th a vo Ti-plasmid cng nh T-DNA. Nhngplasmid ln hn 200 kb l qu ln, kh thao tc trong phng th nghim.

    Ngi ta thnh cng trong vic lm bin i Ti-plasmid v T-DNA nhng phytohormone ny khng c to nn. Trong nhng bc tiptheo gen tng hp opine c ct ra v a vo nhng gen chn lc(xem mc 1.2), v d gen khng kanamycin. Ngy nay, ngi ta s dngnhng h thng c gi l vector hai ngun (binary vector), y chcnng ca Ti-plasmid c thc hin hai plasmid.

    1.2. H thng chn lc v ch th

    Ni chung, tt c cc h thng bin np t l cy tip nhn DNA bnvng thng l rt thp. V vy, ngi ta phi c phng php phn bitrt t cy bin np t mt lng ln cy khng bin np. Thc t xc nhnhng cy c bin np ngi ta s dng h thng gen chn lc. c bit

    a thch l h thng chn lc tri, c ngha l ch nhng cy bin i gen cth ti sinh v pht trin. Ngc li, mt h thng ch thn gin nh hthng ch th GUS, th c nhng cy khng bin np cng ti sinh.     Trong nhiu trng hp ngi ta s dng khng sinh kanamycin cho

    chn lc tri. Tuy nhin, nng cao n c i vi phn ln thc vt.in hnh cho khng sinh nhm ny l mt aminoalcohol gn vi gcng cha nhm amin (Hnh 1.12). Bn cnh kanamycin thu c tStreptomyces kanamyceticus cn c gentamycin (t Micromonosporapurpurea), neomycin (t Streptomyces fradiae) v streptomycin (tStreptomyces griseus) cng thuc nhm ny.

    Ngi ta s dng cc gen khng, v d gen neomycin-phosphotransferase (nptII) m ha cho phosphotransferase, sn phm gen

    2.1. Tng tnh khng v thch nghi vi mi trng

    V cbn phn bit loi thuc dit c chn lc vi loi khng chnlc. liu lng nht nh loi khng chn lc c tc dng i vi thc vtc c im sinh l v hnh thi nht nh. V d: thuc nhm ny gm c:atrazin, bromoxynil, 2,4-D. Tc dng ca thuc dit c chn lc:

    Atrazin: Lm ngng s vn chuyn in t trong h thng quang haII lc lp (cn thit i vi quang hp ca thc vt). Ng khng mn cmvi atrazin.

    Bromoxynil:nh ch s vn chuyn in t trong h thng quangha II ca lp th, lm cht cy hai l mm.

    Glyphosate (Round upR): Lm ngng hot ng enzyme EPSP-

    synthase v qua km hm s tng hp cc amino acid thm.

    Phosphinothricin (PPT) cn gi l Basta: PPT l ng phn dng Lca sn phm tng hp glufosinate. PPT c cu trc tng t glutamic acid,gn khng thun nghch vi enzyme glutamatsynthase, gy c do tch lyNH3.

    2,4-D: Auxin tng hp ny gy hi cho s pht trin ca cy hai lmm, phn ln cy mt l mm khng mn cm.

    Mt s loi thuc dit c chn lc tn ti lu trong t, v vy n lmnhim ngun nc.

    Virus thng c coi l tc nhn gy bnh nguy him. Tuy nhin,quan nim ny khng hon ton chnh xc, v khi phn tch tng phn hgen thc vt cho thy, trong cy trng v cy hoang di tn ti s lng lncc loi virus, trong chng phn ln l khng gy hi. Ngoi ra, virus thc

    vt khng phi l virus ngi v ng vt. Ch c mt s loi virus gythit hi ln cho ma mng. V d loi virus to ra nhiu r nhc cing trong mt s vng ln c gy thit hi n 75%.

    2.2.1. Carbohydrate v acid bo
