INTRODUCTION

APO C-III:

The apolipoprotein C-III its a very importantcomponent of lipoproteins.

It´s synthesized by the liver, but the intestine alsosynthesizes small amounts.

Genes ApoA-I and ApoA-IV , chromosome 11q23.3.

GENERAL PURPOSE

The aim of this study is to determine the correlation

between the polymorphism of apo C-III and chronic

alcohol consumption, high lipid levels in serum.

MATERIALES Y MÉTODOS

Sujetos:

1030 personas.

Edades entre los 15 y los 89, y edad promedio de

43 años.

16 aldeas diferentes del condado de napo (RPC)

MEDICIÓN BIOLÓGICA

Extracción de sangre.

Medición de lípidos.

Extracción del DNA, separación del DNA

genómico de leucocitos( fenol cloroformo).

PCR y FRLP.

Separación por electroforesis.

Reacción en cadena de la polimerasa.

Kary mullis, años 80.

Amplificación directa de un gen o un fragmento de

DNA.

Incremento geométrico del numero de copias.

Se da a partir de concentraciones muy bajas de

DNA.

RFLP

Polimorfismos en la longitud de los fragmentos de

restricción.

No se basa en un cambio fenotípico.

Es la detección de aquellas variaciones de la

secuencia del DNA que tienen como consecuencia

un cambio en el sitio de restricción.

Esto hace que los fragmentos de restricción que se

obtienen son distintos para cada alelo.
