Octet System Overview: Label-Free, Real-Time Analysis for Bioprocessing ForteBio’s Octet System for label-free, real-time analysis T he FortéBio Octet System has been designed to address the need to accelerate the production and characterization of antibody and protein therapeutics in a simple label-free format. By measuring in real-time directly in a microplate, Octet’s analytical capabilities provide greater value in applications where existing methods such as HPLC, ELISA, and surface plasmon resonance have limitations in throughput, performance, workflow, and ease of use. KEY APPLICATIONS Quantitation of antibodies and proteins: • Early cell culture screening • Process development • Manufacturing • Protein purification Kinetic and affinity analysis • k obs , k a , k d , K D • Rapid screening for specificity • Rapid ranking of dissociation rates BIOSENSOR SURFACE CHEMISTRIES Optical biosensors are at the core of BioLayer Interferometry on the Octet system. By relying strictly on the physical properties of molecules binding to the surface, there is no interference from the sample media. A variety of surface chemistries are manufactured to produce signals that are accurate and reliable without needing background subtraction. Available biosensor surface chemistries include • Antihuman IgG biosensors • Protein A biosensors • Streptavidin biosensors • Amine-reactive biosensors KEY FEATURES OF THE OCTET Label-Free Detection: BioLayer Interferometry does not require the use of detection labels to measure the binding interaction at the biosensor surface. Real-Time Analysis: BioLayer Interferometry continuously measures the protein binding at the biosensor surface throughout the interaction. Raw data is displayed in real time and the rapid analysis fits with process workflow. Minimal Interference: Because BioLayer Interferometry detects binding only at the sensor surface, there is minimal interference from biological sample media. Proteins can be assayed in cell culture media or crude lysates without interference. Automated: Octet is automated to perform multistep experimental protocols and complete data analysis. The system runs up to eight samples in parallel, and up to 96 samples in unattended operation.
