ZET

Alkol dehidrogenaz (ADH) ve aldehid dehidrogenaz (ALDH) enzimleri etanol metabolizmasnda rol oynayan bafllca enzimlerdir.Etanolden ADH oksidasyonu ile oluflan asetaldehid, toksik ve reaktif bir metabolittir. Alkolik karacier hastalnn ve alkolle iliflkili t-mrlerin patogenezinde asetaldehidin rol olduu bildirilmifltir (1). ADH1C (eski adyla ADH3) genetik polimorfizm gsteren bir en-zimdir. Toplumumuzda salkl gnlllerde ADH1C alel sklklar daha nceki bir alflma ile saptanmfltr (2) .

Ama: Alkolle iliflkili bafl-boyun tmr (epidermoid kanser) olgularnda ADH1C genotip ve alel frekanslarn saptamak idi. leri a-lflmalarda bu polimorfizmin bafl boyun tmr (epidermoid kanser) patogenezindeki olas rolnn aydnlatlmas planlanmaktadr.

Hastalar ve Yntem: Bafl boyun tmr (Epidermoid Ca) olgularndan kan rnekleri topland (n= 50). Lkositlerden DNA izolas-yonu yapld. Yabanl (ADH1C *1) ve mutant (ADH1C *2) alellerin sklklarn saptamak amacyla PCR sonrasnda endonkleaz (SspI)restriksiyonu yntemi kullanld (3).

Bulgular: Genotip sklklar ADH1C *1/*1, ADH1C *1/*2 ve ADH1C *2/*2 iin srasyla %58 (Gven aral (GA)= 50-65), % 38(GA= 32-44) ve %4 (GA= 2-6) idi. ADH1C *1 ve ADH1C *2 alel frekanslar srasyla % 77 (GA= 0.71-0.83) ve % 23 (GA= 0.17-0.29) ola-rak saptand.

Yorum: ADH1C alel frekanslarnn bafl-boyun tmr (Epidermoid Ca) olgularnda saptanmasnn ardndan halen hasta grubu ilebenzer epidemiyolojik zeliklere sahip kontrol grubu bulmaya alfllmaktadr. Kontrol grubunda benzer flekilde genotipleme ifllemi ya-placak, daha sonra bulgular hasta grubuna ait bulgular ile karfllafltrlacaktr.

Bu alflma Pamukkale niversitesi Tp Fakltesi Arafltrma Birimi Baflkanlnca (2007TPF001) desteklenmifltir.

ABSTRACT

Alcohol dehydrogenase (ADH) and aldehyde dehydrogenase (ALDH) are primary enzymes which play role in ethanol metabolism.Acetaldehyde produced from ethanol by ADH oxidation, is a toxic and reactive metabolite. The role of acetaldehyde in the pathoge-nesis of alcoholic liver disease and alcohol-related tumors had been reported (1). In our population ADH1C (formerly called ADH3)allele frequencies in healthy volunteers had been detected with previous study (2).

Objective: To detect ADH1C genotype and allele frequencies in cases of alcohol-related head and neck tumors (Epidermoid can-cer). In further studies we planned the clarification of the possible role of this polymorphism in the pathogenesis of head and neck tu-mors (Epidermoid Ca).

Patients and Methods: Blood samples were obtained from patients (n= 50) with alcohol-related head and neck tumors (Epider-moid cancer). The genomic DNA was isolated from peripheral blood leukocytes. For the detection of ADH1C *1 (wild type) andADH1C *2 (mutant) alleles, the restriction endonuclease (SspI) method was used (3).

Results: Genotype frequencies of ADH1C *1/*1, ADH1C *1/*2 and ADH1C *2/*2 were 58 % (Confidence interval (CI) = 50-65) , 38% (CI = 32-44) and 4 % (CI = 2-6), respectively. ADH1C *1 and ADH1C *2 allele frequencies were 0.77 (CI = 0.71-0.83) and0.23 (CI = 0.17-0.29), respectively.

Conclusion: After the detection of ADH1C allele frequencies, now we try to find the control subjects with similar epidemiologi-cal properties. The genetic linkage with the clinical data will be analyzed with large number of patient samples in the future researchplan, and then results will be compared with patients groups.

ZET

Ama: Beyin hemisferlerinden biri baskn olarak kullanlmaktadr (1). Hemisferler arasnda anatomik ve fizyolojik asimetri mevcut-tur. Acaba bu asimetri morfolojik olarak ele nasl yansmaktadr? Bu soruya cevap aramak ve solak ile salak kiflilerdeki el asimetrisi-ni ortaya karmak amacyla bu alflma planlanmfltr.

Gere ve Yntem: alflmamzda 164 niversite rencisi zerinde morfolojik lmler yaplmfltr. niversitemiz klinik etik kurulun-dan onay alnmfl olup alflmaya tam solak ve tam salak olan renciler dahil edilmifltir. Seksen yedi (55 salak, 32 solak) kz ve 77 (56salak, 21 solak) erkek rencinin, morfolojik olarak elin palmar uzunluu, palmar genifllii, 1-5. parmak uzunluklar ve parmak boum-lar (distal, orta ve proksimal) yzeyel anatomik yaplar gzetilere, 0.01/0.0005 mm okuma hassasiyetli dijital caliper ile lld (2,3).Ayrca el ve ayak dijital fomlleri kaydedildi. lmler SPSS program ile istatistiksel olarak deerlendirildi p< 0.05 anlaml kabul edildi.

Bulgular: Solak kzlarn salaklara gre sol el palmar genifllii anlaml olarak byk, sol el 2. parmak distal boum anlaml olarak uzun,sol el 5. parmak boyu anlaml olarak uzun tespit edildi (p< 0.05). El dijital formlnde 3, 4, 2, 5, 1 flekline en az salak kzlarda %54.54,en fazla salak erkeklerde ve %78.57 orannda rastland. El dijital formlnde 3, 2, 4, 5, 1 flekline en fazla salak kzlarda %45.46, en azsalak erkeklerde ve %21.43 orannda rastlanmfltr. Solak erkeklerde ayak dijital formlnde ikinci parman dierlerinden uzun olmasnadaha sk rastland. Ayak dijital formlnde 2, 1, 3, 4, 5 flekline en fazla solak erkeklerde %47.70, en az solak kzlarda %34.40 oranndarastlanmfltr. Solak kzlarn iki tanesinde 1, 3, 2, 4, 5 fleklinde dierlerinde rastlanmayan ayak dijital forml grlmfltr.

Yorum: alflmamz solak kzlarda elde morfolojik asimetri grldn ortaya koymufltur. Ayak dijital formlnde de solak kz-larda, dierlerinde rastlanmayan bir forml tespit edilmifltir. El cerrahisi ynnden morfolojik lmler deer tafldndan, gerek cer-rahi gerekse protezlerin hazrlanmas asndan solak kzlardaki asimetri gz nne alnmaldr.

ABSTRACT

Objective: One of the brain hemispheres is dominantly used. There is anatomic and physiological asymmetry between the he-mispheres. How is this asymmetry morphologically reflected in the hand? The present study was planned to find an answer to thisquestion and to reveal the hand asymmetry in right- and left-handed individuals.

Materials and Methods: In our study, morphological measurements were performed on 164 university students. After havingthe ethical consent from the Ethical Committee of our university, completely left-handed and completely right-handed students we-re included in the study. In 87 female (55 right-handed, 32 left-handed) and 77 male (56 right-handed, 21 left-handed) students, pal-mar length and palmar width of the hand, as well as 1st to 5th finger lengths and finger digits (distal, medial and proximal) were me-asured morphologically using a digital caliper with a reading sensitivity of 0.01/0.0005 mm by taking into consideration superficialanatomic structures. Moreover, digital formula for hands and feet were recorded. The data were fed into the SPSS program, whichwas used as the statistical method for the study and p value of < 0.05 is used for significance.

Results: Left hand palmar width was significantly greater, left hand 2nd finger distal digit was significantly longer, and left hand5th finger length was significantly longer among left-handed females when compared to the right-handed (p< 0.05). The hand di-gital formula of 3, 4, 2, 5, 1 was least frequently observed among right-handed females (54.54%) and most frequently observedamong right-handed males with a rate of 78.57%. The hand digital formula of 3, 2, 4, 5, 1 was most frequently observed amongright-handed females (45.46), and least frequently observed among right-handed males with a rate of 21.43%. The foot digital for-mula in which the second finger is longer than the others was more frequent among left-handed males. The foot digital formula of2, 1, 3, 4, 5 was most common among left-handed males (47.70%) and least common among left-handed females (34.40%). In twoof the left-handed females, an uncommon foot digital formula of 1, 3, 2, 4, 5 was found.

Conclusion: Our study revealed that morphological hand asymmetry was observed among left-handed females. As for the footdigital formula, an uncommon formula was also detected among left-handed females. Since morphological measurements are criti-cally essential for hand surgery, the asymmetry in left-handed females should be taken into consideration both for surgical and forprosthesis preparation purposes.
