Whataremitochondriaandmitochondrialdiseases?Oftendescribedasthepowerhousesofcells,mitochondriaareessentialcellularcomponentsthatproduceenergy.Virtuallyeverycellinourbodiescontainmitochondria,whichhavetheirownDNA:mitochondrialDNA(mtDNA),whichisuniqueandseparatefromnuclearDNA(nDNA).MutationsineithermtDNAornDNAcandisruptmitochondrialfunctionsleadingtodiversediseasesthatoftenaffectmultipleorgans.

UnderstandingmtDNAmutationsBecausemtDNAsarepresentinhundredsorthousandsofcopiespercell,theproportion(heteroplasmy)ofmutantmtDNAcanvarywidelyandcandictatetheseverityofapatient’smitochondrialdisease.AnotherspecialpropertyofmtDNAisitsmaternalinheritance;onlymotherspassontheirmtDNAtotheirchildren.

Oocytes(eggsshownascircles)containingamixedpopulationofmutant(red)andnormal(green)mitochondriatransmitmtDNAafterfertilizationtodaughtercells.

OurgoalToinvestigatethesafetyandeffectivenessofOocyteMitochondrialReplacement(OMR)asmeanstopreventtransmissionofmtDNAtooffspring.

Whatisinvolved?WomenwhocarrymtDNAmutationscanparticipateinthisstudy.TheywillundergotheeggharvestingstagesofInVitroFertilization(IVF),awell-establishedtreatmentformanyformsofinfertility.Thenucleargenomeofanunaffecteddonor’soocytewillberemoved.ThenthenuclearDNAfromtheoocytewithamtDNAmutationwillbetransferredintotheenucleatedoocyteofthedonor.Thisstudywillresultinembryosthatarecryopreservedandwillnotresultinpregnancy.Embryotransferintotheuterus(womb)maybepartofafuturestudythatwillrequireapprovalbytheFoodandDrugAdministration(FDA).
