I. INTRODUCTION
Thank you for purchasing the 30 mm ID Preparative Guard Cartridge and Guard Holder assembly. This preparative guard assembly is designed to protect 30 mm ID Preparative Optimum Bed Density (OBD™) Columns and is suitable for both HPLC and SFC.
II. SPECIFICATIONS

Solvent Compatibility Suitable for common HPLC (reversed-phase and normal-phase) and SFC mobile phases. Minor swelling of O-rings in THF and chloroform may require more frequent replacement if leaks observed. Solvent compatibility of the packing is the same as that of the preparative column.

III. T IGHTENING PROCEDURE
1. After inserting guard cartridge, tighten holder by hand until O-rings make contact with the guard cartridge.
2. While holding the 2-1/16” flats, use a 3/4” wrench to tighten holder cap until O-rings are fully seated, between 5-9 tick marks.
IV. START-UP RECOMMENDATIONS
a. Background (Avoid Pressure Pikes)It is possible that a SUDDEN high flow rate start at 50-60 mL/min may contribute to initial disruption of the prep guard cartridge
bed leading to the peak splitting and shortened lifetimes.
It is recommended to start the flow rate up MORE SLOWLY (over 5 or 10 minutes) to avoid this issue.
b. Start-Up1. Purge the pumping system of any buffer-containing mobile
phases and connect the inlet end of the column to the injector outlet.
2. Flush column with 100% organic mobile phase (methanol or acetonitrile) by setting the pump flow rate to 0.1 mL/min and increase the flow rate to 50-60 mL/min over 5 minutes.
3. When the mobile phase is flowing freely from the column outlet, stop the flow and attach the column outlet to the detector. This prevents entry of air into the detection system and gives more rapid baseline equilibration.
4. Gradually increase the flow rate as described in step 2.
5. Once a steady backpressure and baseline have been achieved, equilibrate the column with a minimum of 10-column volumes of the mobile phase.
6. For added assurance, perform an efficiency test on the prep guard cartridge before using it. Waters recommends using a suitable solute mixture, as found in the “Performance Test Chromatogram”, to analyze the prep guard cartridge upon receipt. Determine the number of theoretical plates (N) and use this value for periodic comparisons.
V. INSTALLING AND REPLACING THE GUARD CARTRIDGE AND CARTRIDGE HOLDER
Install the cartridge holder onto the HPLC system in front of the column using the rigid connector. The tubing in the rigid connector is 0.020” or 0.5 mm ID. For all other connections, use the same tubing.
Installing and replacing the guard cartridge:
1. Remove the cartridge holder from the system.
2. Unscrew the holder cap from the holder body.
3. Remove the old guard cartridge, if present.
4. Visually inspect the O-rings for damage or debris. Replace, if necessary (see Section V).
5. Install the new guard cartridge. The direction of the flow arrow on the guard cartridge and holder body should point towards the detector.
6. Re-assemble the holder and tighten according to instructions above.
7. Attach inlet side of assembled cartridge holder to the tubing from HPLC injector.
8. Flush with 10 volumes of mobile phase at a low flow rate.
9. Attach outlet side of the cartridge holder to the column inlet using rigid connector.
10. Check for leaks.

VI. WHEN TO REPLACE THE GUARD CARTRIDGE
Replace guard cartridges before contaminants affect your separation. For best results, replace the guard cartridge when:
1. The quality of your separation deteriorates.
2. The initial backpressure increases significantly. (more than 20 percent).
3. Contaminant peaks appear in the chromatogram.

VII. REPLACING THE O-RINGS
The O-rings should be replaced if leaks from the cartridge holder are observed. You will need replacement part number 186007012 (O-rings - 30 mm ID Prep Guard Holder, 2/pk), a pointed device (such as a needle or dental pick), and a clear workspace where small parts are easily retrieved if dropped.
To replace the O-rings:
1. Remove the cartridge holder from the system.
2. Unscrew the holder cap from the holder body.
3. Remove the guard cartridge.
4. Using a sharp needle, pry the O-rings from their grooves in the holder cap and body. Be careful not to scratch the groove, this may lead to leaks. It may be helpful to insert the needle partway into the O-ring in order to remove it.
5. Examine the grooves and new O-rings. Remove any particulate matter with a cotton tip applicator.
6. Install the new O-rings into the grooves. To ensure O-rings are installed properly in the groove, insert guard cartridge into holder body or holder cap and rotate 1/4 turn, applying light hand pressure against the cartridge.
7. Re-assemble holder with cartridge and tighten according to directions above.

IX. GUARD CARTIDGE STORAGE
a. HPLC Guard CartridgesTo maintain the integrity of the bonded phase as well as the guard cartridge hardware and frits, flush the guard cartridge with at least eight column volumes of 100% acetonitrile after every use. Never store the guard cartridge in methanol or aqueous methanol. If the mobile phase contains a buffer salt, first flush the guard cartridge with at least ten column volumes of 18 MegaOhm water followed by at least eight column volumes of 100% acetonitrile. Please refer to Section II for the column volume.
b. SFC Guard CartridgesPurging the SFC prep guard cartridge in pure CO2 is recommended prior to storage. This will eliminate or minimize retention changes of the guard cartridge during storage. Storing the guard cartridge in methanol is not recommended.

XI. WARRANT Y/SERVICE INFORMATION
Waters Corporation warrants its guard cartridges and guard holders in accordance with the following terms and conditions:
Waters will replace without cost, any guard cartridge or guard holder that fails to perform satisfactorily, if notified within 90 days from your receipt. To return guard cartridges and guard holders, the user must have a Return Authorization Number granted by the Waters Technical Service Department.
Approval is subject to the following exclusions:
1. Physical damage to the guard cartridge or guard holder because of misuse or abuse.
2. Damage to the guard cartridge or guard holder because of use with incompatible solvents or use at incorrect temperatures or pressures.
3. Failure or decreased performance as a consequence of normal use.
