Medium MRS broth dibuat dengan mengencerkan 52,2 gram bubuk Merck MRS broth
dalam 1 liter aquades, kemudian medium diaduk hingga terlarut dengan merata. Setelah
itu medium disterilkan menggunakan autoclave pada suhu 121oC selama 15 menit.
