Anti biotics di ssolved in ethanol need not be sterilized.  Do not use expired antibiotics to make stock soluti ons, as t hey may be labile and may cause inconsist ent results.  Add antibiotics to liquid medium i mmediately prior t o use from pr operly stor ed steril e stock solutions.  Lower concentrations are suitable f or the selection of cells contai ning low copy number plasmids, higher concentrations for high copy number plasmids.  Agar medium should be cooled to 50 o C or lower before the addition of heat-sensitive antibiotics and nutrients.  Plates should be stored upside down at 4 o C in the dark (foil wrapped) for no longer than three months due to degradation of antibiotics.  Magnesium is an antagonist to tetracycl ine; use media wi thout magnesium salts when working wi th bacteria that are tetracycline resistant.
