GENERAL INFORMATION

The APVMA (Australian Pesticides and Veterinary Medicines Authority) Gazette is published fortnightly and contains details of

the registration of agricultural and veterinary chemicals products and other approvals granted by the APVMA, notices as requi red

by the Agricultural and Veterinary Chemicals Code (the Agvet Code) and related legislation and a range of regulatory material

issued by the APVMA.

Pursuant to section 8J(1) of the Agvet Code, the APVMA has decided that it is unnecessary to publish details of applications

made for the purpose of notifying minor variations to registration details. The APVMA will however report notifications activity in

quarterly statistical reports.

The following instructions set out how a person can deal with the cancelled labels.

SUPPLY

A person may supply or cause to be supplied product bearing a cancelled label manufactured prior to 15 September 2014

at wholesale and retail level, until the 15 September 2015.

After 15 September 2015 it will be an offence against the Agvet Codes to have possession or custody of product bearing

a cancelled label with the intention to supply, or to supply the product bearing a cancelled label.

USE

A person may continue to use the product bearing a cancelled label according to its label instructions until 15 September

2015.

Any person who possesses, has custody of, uses, or otherwise deals with the listed products bearing a cancelled label in

accordance with the above instructions is taken to have been issued with a permit under the Agvet Codes to so possess,

have custody of, use or otherwise deal with the product after the label has been cancelled until 15 September 2015.

The supply and use of the product bearing cancelled labels must be in accordance with the conditions of registration or

approval, including any conditions relating to the shelf life or expiry date.

It is an offence to possess, have custody of, use, or deal with the products bearing cancelled labels listed in the table in a

manner that contravenes the above instructions.

Agricultural and Veterinary Chemicals Code Act 1994 Section 47C Notice end of registrations of chemical products

This notice is published under section 47C of the Agricultural and Veterinary Chemicals Code scheduled to the

Agricultural and Veterinary Chemicals Code Act 1994 (Agvet Code). The APVMA provides notice that:

1. The registration of the products listed at Attachment A ended and were not renewed for the financial year

2014–2015 (the Product(s)).

2 The registrations for the Product(s) ended on 30 June 2014.

INSTRUCTIONS FOR DEALING WITH THE CHEMICAL PRODUCTS

The APVMA issues the following instructions for the possession, custody, supply and use of existing stocks of the

Product(s) listed in this notice and which were manufactured in, or imported into, Australia before 30 June 2014:

1. The label attached to the container of the product must be identical (except for particulars relating to the batch

number, date of manufacture or expiry date of the product) to an approved label for that product that was in force

on 30 June 2014; and

2. The products are to be used in accordance with the instructions on the approved label for that product.

Note: The label(s) for these products in force on 30 June 2014 continue in force while these instructions apply to the

product.

INSTRUCTIONS FOR POSSESSION, HAVING CUSTODY OF, USING OR OTHERWISE DEALING WITH THE

PRODUCTS

If after publication of this notice a person, possesses, has custody of, uses or otherwise deals with the Product(s) in

accordance with the instructions in this notice, the person is taken to have been issued with a permit to possess, have

custody of, use or otherwise deal with the Product(s) in accordance with those instructions until:

(a) 1 year after the day on which the registration ended (being 1 July 2015); or

(b) The APVMA, by notice published in the Gazette, declares that this subsection does not apply in respect of the

Product(s);

whichever first occurs.

LATE APPLICATIONS FOR RENEWAL OF REGISTRATION

A late application for renewal of registration for products whose registration has lapsed as at 30 June 2014 will only be

accepted up until 15 December 2014.

Products whose registration lapsed before 30 June 2014 and late applications for products whose registration lapsed as

at 30 June 2014 that are received after 15 December 2014 will need to submit a new application for product registration.

FAILURE TO COMPLY WITH NOTICE

All products listed in this notice are now subject to APVMA Compliance. Where instances of possession, custody of, use

or otherwise dealing with the Product(s) with the intention of supply, or to supply the Product(s) outside the scope of the

notice are identified, regulatory action will be considered.

It is an offence under the Agvet Code for a person to supply, or cause or permit to be supplied or possess for supply, any

Product(s) listed in this notice after 30 June 2014 other than in accordance with this notice. It will be an offence to supply

the Product(s) or to possess or have custody of the Product(s) with the intent of supplying it after 30 June 2015 or an

earlier date notified by the APVMA by notice in the gazette.

The Australian Pesticides and Veterinary Medicines Authority (APVMA) has before it an application from Bayer Australia

Ltd (Animal Health) for the approval of a new active constituent, pradofloxacin. The APVMA also has before it

applications from the same applicant for the registration of four new products containing the new active constituent. The

products are: Veraflox 120 mg Tablets for Dogs Antimicrobial, Veraflox 60 mg Tablets for Dogs Antimicrobial, Veraflox 15

mg Tablets for Dogs and Cats Antimicrobial, Veraflox 25 mg/mL Oral Suspension for Cats Antimicrobial (‘the products’).

SUMMARY OF THE APVMA’S EVALUATION OF PRADOFLOXACIN

Pradofloxacin is a new active constituent and there is no compendial specification available.

The Veterinary Medicines Program of the APVMA has evaluated the chemistry aspects of pradofloxacin active

constituent (manufacturing process, quality control procedures, impurities, stability, specifications, batch analysis,

analytical methods, and packaging) and found them to be acceptable.

An external toxicology reviewer has completed a toxicology evaluation of pradofloxacin. The Reviewer found that

pradofloxacin is a low acute oral toxicant and a low dermal toxicant. Although it can predispose to slight photoirritation of

the skin, it is not a skin irritant or a skin sensitiser. The eye irritation potential is considered to be slight. Pradofloxacin is

mutagenic, clastogenic and genotoxic. All positive genotoxic effects in mammalian systems were associated with

exposure levels of at least 13 μg/mL. Effects were not observed at exposure levels below 10 μg/mL, which correlates

with a No Observed Effect Concentration of 10 μg/mL for inhibition of topoisomerase IIα. A high dose of pradofloxacin

(7000 ppm) in a carcinogenicity study resulted in hyperplasia of the gall bladder and an increase in the incidence of gall

bladder adenoma. Multigenerational reproduction studies showed a slight reduction in fertility, while foetal abnormalities

and retarded foetal development were observed in developmental studies.

An Acceptable Daily Intake and an Acute Reference Dose were not established as the products are intended for use in

non-food-producing animals.

The Delegate to the Secretary of the Department of Health for scheduling has decided pradofloxacin to be appropriate

for inclusion in Schedule 4 of the Standard for the Uniform Scheduling of Medicines and Poisons (SUSMP) with no cut-

offs or exceptions, along with an implementation date of 01 February 2014.

The External Toxicology Reviewer has indicated there are no objections on toxicological grounds to the approval of the

active constituent pradofloxacin.

The APVMA has accepted the findings and recommendations of its advisers. The APVMA is satisfied that the proposed

use of pradofloxacin would not be an undue toxicology hazard to the safety of people exposed to it during its handling

and use.

REQUIREMENTS OF SECTION 14(3)(e) AND (f) OF THE AGRICULTURAL AND VETERINARY CHEMICALS CODE

(THE ‘AGVET CODE’), SCHEDULED TO THE AGRICULTURAL AND VETERINARY CHEMICALS CODE ACT 1994

1. The APVMA has evaluated the applications and in its assessment in relation to human and environmental safety under

section 14(3)(e)(i) and 14(3)(e)(ii) of the Agvet Code, it proposes to determine that:

(i) The APVMA is satisfied that the proposed use of the products would not be an undue hazard to the safety of people

exposed to them during their handling and use (section 14(3)(e)(i)).

An External Toxicology Reviewer has conducted a risk assessment and found that the submitted data support the

safe use of the products from a toxicological perspective.

A toxicity estimation of the associated products was conducted, based on information on the products’ ingredients

and their concentration in the formulations. It was concluded that the products have low acute oral and dermal

toxicity, slight eye irritation potential and no skin irritation and sensitisation properties.

Human exposure to pradofloxacin is possible through hand contact with the products when owners administer the

products to dogs and cats. Accidental skin contact with the suspension formulation may also occur in the event of

splashing or a spillage and subsequent clean-up procedures. Risks associated with the acute hazards in adults can

be mitigated by safety directions—Harmful if swallowed. May irritate the eyes. Avoid contact with eyes. Wash

hands after use. Those risks associated with oral exposure in young children as a result of hand to mouth transfer

or by accidental direct ingestion will be mitigated by blister packs for tablet products and child-resistant closures for

the oral suspension.

The APVMA has considered these findings and has accepted the Reviewer’s recommendations.

An External Reviewer assessed a qualitative risk assessment provided by the Applicant. The Reviewer considered

the risk of antimicrobial resistance to be medium for Salmonella and Campylobacter, high for methicillin-resistant

Staphylococcus aureus (MRSA) and medium for methicillin resistant Staphylococcus pseudintermedius (MRSP).

Multi-resistant extraintestinal pathogenic Escherichia coli (ExPEC) associated with human urinary tract infections

presented a medium hazard, with an unknown exposure (the role of dogs in the carriage of ExPEC strains is

unresolved), but a medium to high impact.

The Reviewer concluded that the use of pradofloxacin in accordance with the proposed label instructions could

lead to the emergence of fluoroquinolone-resistant zoonotic organisms in treated dogs and cats, could select for

multi-drug resistant organisms and consequently could compromise the health of humans in contact with treated

dogs and cats through the acquisition of fluoroquinolone and multi-drug resistant enteric organisms, MRSA and

MRSP.

The Reviewer recommended that the products be regarded as reserve agents and their use restricted. The

National Health and Medical Research Council also advised the APVMA to consider appropriate restrictions that

might apply to the use of fluoroquinolones in small animal species, and added that best practice suggests that

fluoroquinolones should be regarded as reserve agents for use in veterinary applications when resistance to other

antimicrobial agents has been proven.

The APVMA has accepted the advice and recommendations from its advisors. The APVMA proposes to be

satisfied that the risk to human health through the acquisition of fluoroquinolone and multi-drug resistant pathogens

from dogs and cats treated with the products could be mitigated when the products are prescribed only for the

indications in the target animals specified on the labels and based on sensitivity testing, and the products are

prescribed when the response to other treatments is poor and there are no suitable alternatives.

(ii) The APVMA is satisfied that the proposed use of the products will not be an undue hazard to the safety of people

using anything containing their residues (section 14(3)(e)(i)).

The products are intended for use in non-food-producing animals only. Pradofloxacin is unlikely to enter the food

chain and therefore the determinations of an Acceptable Daily Intake, an Acute Reference Dose and Maximum

Residue Limits are not considered necessary.

(iii) The APVMA is satisfied that the proposed use of the products containing the active constituent pradofloxacin is not

likely to be harmful to human beings if used according to the product label directions (section 14(3)(e)(i)).

The Delegate to the Secretary of the Department of Health for scheduling has decided pradofloxacin is appropriate

for inclusion in Schedule 4 of the Standard for the Uniform Scheduling of Medicines and Poisons with no cut-offs.

The signal heading appropriate to Schedule 4, the first aid instructions and safety directions recommended by the

External Toxicology Reviewer will appear on the product labels.

2. The APVMA has evaluated the applications and in their assessment in relation to environmental safety under section

14(3)(e)(iii) of the Agvet Code, it proposes to determine that:

(i) The APVMA is satisfied that the proposed use of the products containing the active constituent pradofloxacin would

not be likely to have an unintended effect that is harmful to animals, plants or things or the environment.

The Department of the Environment has conducted a VICH Phase 1 assessment and has concluded that the

environmental risk is likely to be acceptable under the proposed use pattern and estimated levels of use of the

products. The APVMA has accepted this advice. The product labels will contain a suitable disposal statement.

The APVMA has assessed the safety of the proposed products to the target animals based on the data the

Applicant had provided. Safety studies with the tablet products demonstrated that dogs tolerated daily oral doses of

15 mg/kg bw for 91 days. Safety studies with the tablets and the oral suspension demonstrated that cats tolerated

daily oral doses of 15 mg/kg bw for 21 days. Pradofloxacin induced articular cartilage lesions in young dogs and

kittens under 12 months old (18 months for giant breeds of dogs) and is contraindicated in these age groups.

Studies were not provided to establish the safety of pradofloxacin in breeding dogs and cats. therefore, the

products are contraindicated for use in this sub-set of animals.

The APVMA is satisfied that the proposed products would not have an unintended effect that is harmful to animals.

The contraindications, precautions and side effects will be included on the product labels.

3. The APVMA has evaluated the applications and in their assessment in relation to whether the trade criterion has been

met in accordance with section 14(3)(e)(iv) of the Agvet Code, it proposes to determine that:

(i) The APVMA is satisfied that the proposed use of the products would not unduly prejudice trade and commerce

between Australia and places outside Australia as the product is not for use in animals producing any major

Australian export commodities.

The products are intended for use in non-food-producing animals only. Therefore, it is unlikely that humans would

be exposed to residues of pradofloxacin in their diet.

4. The APVMA has evaluated the applications and in their assessment in relation to whether the efficacy criterion has been

met in accordance with section 14(3)(f) of the Agvet Code, it proposes to determine that:

(i) The APVMA is satisfied that data from trials supporting the efficacy of the products adequately demonstrate that if

used according to the product label directions, the products are effective for their proposed uses.

Dose determination studies have established the recommended label dose of 3 mg/kg bw/day for the tablets and

5 mg/kg bw/day for the oral suspension. The dissolution profiles of the tablet products are similar. Batch analysis

data have confirmed that the products can meet the release specification for dissolution.

Pharmacokinetic studies have shown that pradofloxacin was absorbed rapidly (Tmax of 2 hours) after oral

administration, reaching peak serum concentrations (Cmax 0.15 μg/mL) in dogs that were greater than the minimum

inhibition concentrations (MICs) of the target pathogens. Peak concentrations of 0.12 μg/mL were reached within

30 minutes in cats dosed with tablets at 3 mg/kg bw and 0.21 μg/mL within 1 hour when dosed with the oral

suspension at 5 mg/kg bw. MIC90 values for the target pathogens ranged from 0.03 - 0.25 μg/mL. A high volume of

distribution (>2L/kg bw) indicated good tissue penetration. The bioavailability was high in both target host species.

Pradofloxacin was eliminated from serum with a terminal half-life of approximately 7 and 9 hours in dogs and cats,

respectively.

Clinical trials and MIC studies both have demonstrated that pradofloxacin was efficacious for the indications that

are claimed and was effective against susceptible strains of the following target pathogens: Staphylococcus

intermedius group (including Staphylococcus pseudintermedius), Escherichia coli, Staphylococcus aureus,

Streptococcus canis, Porphyromonas gingivalis, Prevotella intermedia, Staphylococcus felis and Pasteurella

multocida. These studies justified treatment durations of 7 days for wound infections, and infections of the urinary

tract, gingiva and periodontal tissues, 14 days for superficial pyoderma and 21 days for deep pyoderma.

The label bears adequate instructions for the use of the product.

should relate only to matters that are required by the APVMA to be taken into consideration in determining whether the

legislative criteria set out in section 14(3)(e) of the Agvet Code have been met. Submissions should state the grounds on

Afoxolaner is a new active constituent and there is no compendial specification available.

The Pharmaceutical Chemistry Section of the APVMA has evaluated the chemistry and manufacturing aspects of

afoxolaner and is satisfied that all the data requirements (including the physico-chemical properties, spectral

identification, manufacturing and quality control aspects, impurity formation, active constituent specification, stability,

batch analysis data, analytical methods and packaging information) necessary for the approval of this new active

constituent have been met.

The Office of Chemical Safety (OCS) in the Department of Health and Ageing has conducted a toxicological assessment

of afoxolaner and advised that the proposed use of this active constituent would not be an undue hazard to human

health. An Acceptable Daily Intake and Acute Reference Dose were not established as the products are intended for use

in non-food-producing animals.

The Delegate to the Secretary of the Department of Health for scheduling has included afoxolaner in Schedule 5 of the

Continued Suspension of Certain Products Containing Dimethoate and their Associated Label Approvals

The APVMA has continued the suspension of the registrations and label approvals for thirteen agricultural chemical

products containing dimethoate, as identified in the table below. The decisions to extend the suspensions of the

registrations and label approvals were made under subsections 41(1) and 41(2) of the Agricultural and Veterinary

Chemicals Code Act 1994 (Agvet Code).

The suspension of product registrations and label approvals are now in effect until 5 October 2015.

Active constituent approvals are not affected by the APVMA’s decision to continue this suspension.

REASONS FOR CONTINUING THE SUSPENSION

The APVMA suspended all products containing dimethoate and their associated labels on 6 October 2011. The dietary

exposure concerns that formed the basis of this original suspension of label approvals and product registrations have not

altered for the products listed in Table 1. Therefore continued suspension is necessary to ensure that the use of products

containing dimethoate does not result in any undue hazard to the safety of people or have an effect that is harmful to

human beings.

All other products containing dimethoate are not suspended because their approved labels contain instructions that are

consistent with the instructions for use of suspended products. These products may be used according to the instructions

on their approved labels.

INSTRUCTIONS

In accordance with subsection 45B(3) of the Agvet Code, a person may only possess, have custody of or deal with a

suspended product (as identified in Table 1) in accordance with the instructions contained in this notice of suspension.

During the period of suspension, a person may not possess, have custody of, use or deal with any of the suspended

products or products bearing suspended labels other than in accordance with:

the instructions set out at the end of this Notice, and

a permit issued by the APVMA.

In October 2011, permits specifying modified instructions for use, were issued under section 114 of the Agvet Code (PER

13155 for agricultural products and PER 13156 for home garden products). The duration of these permits has been

extended to 5 October 2015. The instructions for use have been reproduced at the end of this Notice.

WARNING

The registration and label approvals of the products listed above are not in force during the period of suspension.

From 6 October 2011 a person may only possess, have custody of, use or otherwise deal with the products identified

above in accordance with a permit issued by the APVMA and the instructions set out at the end of this Notice.

The APVMA may cancel any of these permits and issue replacement permits at any time during the suspension period.

Before using, supplying or otherwise dealing with products containing dimethoate, suppliers and users need to be

satisfied that the instructions set out at the end of this Notice continue to apply, and that their activities are authorised by

a permit. Current permits are available at www.apvma.gov.au/permits/search.php.

Failure to comply with the instructions set out at the end of this Notice attracts a penalty under section 45C(2) of the

Agvet Code.

INSTRUCTIONS FOR POSSESSING, HAVING CUSTODY OF, USING OR DEALING WITH SUSPENDED PRODUCTS

CONTAINING DIMETHOATE

Possession and custody

A person may possess, have custody of, use or otherwise deal with the suspended or cancelled dimethoate products

only in accordance with these instructions or permits PER 13155 for agricultural products or PER 13156 for home garden

products, issued by the APVMA.

Supply

All suppliers must at the time of the supply of a product covered by these instructions provide to the person taking

responsibility for the supplied product:

a copy of the relevant permit (currently PER 13155 for agricultural products or PER 13156 for home garden

products) in full setting out the conditions and instructions for use, and

supply product with a copy of the instructions contained in those permits securely affixed to each container of

product.

Use

PERSONS who wish to use suspended products containing dimethoate (as listed above) must read, or have read to

them, the instructions included in the relevant APVMA permit. Users who have had the instructions read to them must

confirm to the reader that they understand the instructions.

READ THESE INSTRUCTIONS before using or otherwise handling the product.

When using or otherwise handling the product, follow the instructions of the current label except as follows:

PROHIBITED CROP USES: HOME GARDEN PRODUCTS

The current (suspended) label may include instructions for use on fruit trees and vegetables. The use of dimethoate on

these crops is no longer approved and the following restraint applies.

DO NOT apply to food producing plants in the home garden

PROHIBITED CROP USES: AGRICULTURAL PRODUCTS

The existing (suspended) label may include instructions for use on the crops listed below. The use of dimethoate on

these crops is no longer approved and the following restraints apply.

DO NOT USE as a post-harvest treatment for capsicums or tomatoes.

DO NOT USE as a post-harvest quarantine treatment for capsicums or tomatoes.

DO NOT USE on tomatoes grown in covered or protected situations such as glasshouses, green houses or plastic

tunnels.

DO NOT USE on cherry, grape or mini tomatoes

DO NOT USE as a foliar, post harvest or quarantine treatment on:

tropical or subtropical edible peel fruit [babacos, carambolas (five corner), figs and edible peel varieties of guavas,

kiwifruit and persimmons].

onion, parsnips, radish, rhubarb, sweetcorn, tomatoes for processing, tomatoes (large field grown for fresh

consumption) prior to fruit set, turnip and zucchini

These directions for use must be used in conjunction with existing label directions and the restraint statements. Where

the instructions in this notice are inconsistent with the label instructions, the instructions in this notice must be followed.
