    Abstract

    Agrobacterium- mediated transformation was used to produce transgenic walnut (Juglans regia L.)

    embryos. in this research Agrobacterium strains LBA 4404 , C58 , EHA101 carrying vectors PBI121 with gus

    and nptII genes under CaMV35S and NOS promoters respectively, and P6u-ubi-FVTI included Fldand hpt

    genes under the same promoters were employed. Somatic embryos were used as explant in transformation trials.

    Explants were co-cultivated with recombinant plasmid bearing Agrobacterium and Inoculated embryos were

    evaluated on selective media. Positive gus assay analysis with the alive embryos inoculated by LBA 4404

    included PBI121 on selective media indicated the success of our transformation approach. Inoculated embryos

    by LBA 4404 included P6u-ubi-FVTI carrying fldgene had higher growth related to their controls and did not

    show browning and necrosis symptoms on selective media after 1 month. This is the first report on the

    probability of walnut transformation byA. grobactreum-mediated in Iran that can be used for introduceing other

    interested genes.
