Methods for the Determination of Legionella in Warm

Water Systems

Overview

On the 1 February 2017, it became mandatory for all Queensland

Health Hospitals with inpatients, Queensland Health operated

residential aged care facilities and licensed private health facilities

to have a Water Risk Management Plan. These plans are to include

a schedule for testing water for Legionella and other identified

hazards, and require that confirmed Legionella detections be

reported to the Department of Health within one business day of

detection. As per the amended Public Health Regulation 2005, a prescribed

test for Legionella

• quantifies the number of Legionella colony forming units

in the sample tested: and

• is carried out by an accredited laboratory and is

identified in the scope of the laboratory’s accreditation

MALDI-TOF MS Legionella Bio typing

ALS can provide rapid MALDI-TOF MS confirmation on both

standard level and low level Legionella culture methods.

Traditional methods of detecting Legionella in a water sample take

up to 10 days for the standard method and 12 days for the low-

level method. This involves using a cultural method incubated for

7 (standard) to 10 (low level) days, followed by biochemical

confirmation, which typically takes a further 2 to 3 days to produce

a confirmed detected result.

MALDI-TOF MS (Matrix assisted laser desorption ionization-time of

flight mass spectrometry) eliminates the 2-3 day confirmation

time, giving a confirmed Legionella result within minutes. This

means that confirmed Legionella results are available in 7 days for

the standard method and 10 days for the low-level method.

MALDI-TOF MS uses Mass Spectrometry to measure a unique

molecular fingerprint of an organism. Specifically, it identifies

highly abundant biomolecules that are found in all microorganisms.

The characteristic patterns of these molecules are matched against

an extensive open database comprising thousands of individual

strains of micro-organisms to identify the organism to the species

level.

ALS in the United Kingdom was one of the first Environmental

laboratories in the world to validate MALDI-TOF technology for

Legionella and has characterised one of the world’s largest culture

collections for Legionella Species. This innovation led to ALS

Environmental being awarded the UK Combating Legionella

Supplier of the Year award in 2016.

ALS recently completed a citywide project to provide Icon

Water, a water and wastewater public utility, with a new

District Metering Area (DMA) monitoring network. In total 42

locations throughout Canberra make up the DMA Network,

comprising 6 flow monitoring stations and 38 pressure

monitoring stations (Figure 1). The flow monitoring stations collect consumption trends in

the water supply drawn from key reservoirs across the ACT,

whilst 38 strategically placed monitoring locations show

pressure of the water mains, indicating supply pressure to

residential areas on a suburb by suburb basis. The pressure

data collected is used by Icon Water from an operational

perspective to manage leakage control, identify burst water-

mains, determine when supply reservoir head pressures are

low and provide accurate figures to ensure they are meeting

the minimum pressure requirements for the water supply

network.As the project was to upgrade an outdated DMA monitoring

network with leading-edge monitoring technology and

telemetry, ALS was faced with a number of challenges not

least of which was due to the size of the existing station

housings; requiring a custom compact retrofit to be designed

and installed.A new station enclosure removed the entire monitoring,

charging and telemetry systems from moisture ingress that

was present in the previous housings. The new updated

system utilises the Campbell CR200X data logger which

scans for changes onsite every 10 seconds and then stores

any data that sits outside set parameters. The majority of the

stations are powered by 240VAC mains with a battery backup

however, for some of the remote sites a solar powered option

was developed. Advances in cellular telemetry systems have enabled ALS

to develop ways of retrieving the data in real time through

the 3G modem via ALS Secure RADIUS Server. Data is

then automatically processed and stored in the ALS data

management software Hydstra (Figure 2), then sent to Icon

Water’s server for their application.

to resolve supply disruption but also identify distribution problems before disruption arises.

Scheduled internal audits are performed on all quality management system elements. System audits are qualitative evaluations of all components of the laboratory systems. They determine if the measurement systems are being used appropriately. Such audits typically involve a comparison of the activities given in the QA Manual with those actually scheduled or performed and are conducted by a Quality Assurance Manager at each facility. Audit findings are used by ALS to improve process performance.

With projects that ALS supports reaching every corner of this continent and often outer islands and seas, logistics is very important to ALS. Sample numbers from clients are approaching a million per annum with many having multiple containers. This makes for a lot of bottles.

What is equally important to ALS is that we constantly innovate to improve sampling efficiency for our clients. ALS miniaturisation projects have led the industry for the last decade with significant reduction in volumes.

Key outcomes for clients include: � Significant manual handling risk reduction � Improved sampling efficiency on low flow wells � Improved logistics and reduced freight costs � Removal of 50L, 44L then 36L eskies and no further

purchases of 32L eskies from supply to reduce further strain/sprain risks.

Collectively these changes have cut the weight of samples collected, handled and freighted by ALS clients by ~280,000kgs across Australia in 2012 and every year after.ALS is nearing the end of this project for waters however remains focused on the safety and efficiency of field operations and will support our clients through great science and great innovation. If we can facilitate our clients to sample a ground water monitoring event (GME) using a ‘six pack’ esky versus lugging a 26L esky we will do it. Understanding laboratory quality, working well with laboratories and maximising the communication was challenging in a decade of high industry growth. Understanding all those preservation, headspace and filtering requirements and different rules for bottles was also challenging. What was also observed was that many field techniques had significant (sometime negative) effect on sample quality and therefore results.

After discussions with several auditors, major consultants and industry major clients a possible opportunity was identified in training field staff and new graduates in the machinations of laboratories and how to optimise quality and minimise potential resampling events or data quality qualifiers.

ALS made a commitment to develop a course with senior 'expert' presenters.

In the last two years ALS has had over 200 industry graduates, field staff and a few experienced industry professionals attend these courses. Attendees have originated from Consultant, Oil Company Majors, Mining Industry, Coal Seam Gas, and General industry sectors.

ALS is committed to continuing and expanding this training to raise quality and knowledge in the industry and support our valued clients. For further information please contact local ALS management.

Among the feedback questions ALS asks participants are:

“Would you recommend this to colleagues?” and

“Would you recommend this be included in graduate and refresher training programmes?”

Feedback to date on both of these questions is typically in the range of 95-99% "Yes!". Using a rating of 1 (poor) to 10 excellent, ratings for “worthwhile experience”, “met my training needs” and “overall value” typically average 8-9.5.

Phase two of this programme is just commencing and includes the new NEPM plus Asbestos and canister sampling topics.

Recent successes of the R&D programmeAlgal ToxinsALS has developed a full range of Algal toxin analyses including Cylindrospermopsin, Anatoxin, Microcystin, Saxitoxin and Nodularin groups/chemicals.

Part per trillion Organo-Chlorine Pesticide (OCP plus PCB) analysisALS has developed incredibly low testing capabilities to meet ANZECC 99% protection in fresh and marine matrices, setting new benchmarks in environmental testing LORs.

Part per trillion PAH analysis in waterALS has developed a very precise and accurate trace PAH method with Benzo(a)pyrene LOR at 1ppt. This provides a testing limit well below the benchmark limit of 10ppt benefiting drinking water protection.

Super Ultra Trace Pesticides and HerbicidesThis extensive range of approximately 100 organic analytes by LC/MS/MS provides clients with some of the lowest LORs (detection limits) available in Australia with many in the part per trillion (ppt) range.

Sampling and Analysis of Soil Gas Using CanistersInterest in Soil Gas has increased significantly as reflected in the Vapour and Soil Gas Investigations section of the 2013 National Environment Protection Measure (NEPM). ALS offers comprehensive accredited analyses to meet this guidance.

PPCPs, Endocrine Disrupting Compounds & SteroidsRecent global research has identified Pharmaceutical and Personal Care Products (PPCPs), Endocrine Disrupting Compounds (EDCs) and Steroids as contaminants of emerging concern (CECs) to humans, animals and the environment. These are analysed to ppt levels to meet 2008 Australian Guidelines for Water Recycling.

PFOS/PFOA & Extended AFFF'sALS provides accurate PFOS results in a variety of matrices from Water and Soil to pristine drinking water. ALS has several R&D projects occurring in this area to develop new capabilities and support PFC assessment and remediation. ALS also has leading capabilities in separation of eleven PFOS Isomers including accurate total PFOS quantification. New PFAS testing will include 28 parameters following a review of global requirements and testing trends.

Diesel Particulate Matter & Respirable Crystaline SilicaThese tests have been developed to support ALS clients in the Occupational hygiene sector. Full NATA accreditation has been gained on DPM – a key test for underground mining and and increasingly of concern in metropolitan vehicle tunnels.

TRH in Soil gas to meet NEPM including C16ALS has developed TRH analysis in Soil gas including quantification of heavier hydrocarbons to C16 in accordance with NEPM.

Analysis Of Infectious Enteric VirusesEnteric viruses such as adenoviruses and enteroviruses are a and may pose a significant risk to public health. The Australian Guidelines for Water Recycling recommend the use of adenoviruses and enteroviruses as “representatives of viral pathogens” for the validation monitoring of wastewater treatment processes.

Extractable Petroleum HydrocarbonsALS has developed an EPH test method in waters that improved accuracy and precision plus reduces false positive interferences from natural organic matter. This is designed to assist practitioners in resolving low level residual traces of natural or weathered organics.
