The CD20 antigen is a nonglycosylated membrane-embedded protein which exhibitsanhydrophobic regionspanning four times themembrane.The longN-andC-terminalendsofthemoleculearelocatedwithinthecytoplasm.Thelevelofphosphorylationofthecytoplasmictail isresponsiblefortheheterogeneityofthemolecularweight.CD20mayalsoexistonthecellsurfaceasahomooligomericcomplexformingwithothermoleculesamultimericreceptorcomplex.TheCD20moleculeispresentonallnormalBlymphocytesfromperipheralblood,lymphnode,spleen,tonsilandbonemarrowbutisabsentfromplasmacells.TheCD20antigenmaybeweaklyexpressedonasubsetof restingT lymphocytes.However,CD20 isnotexpressedonotherleucocytesubsetsincludingNKcells,monocytesandgranulocytes.
