The image on the cover shows the new logo that the Physiology Department developed in May-June 2016. This replaces our old logo, which the design was outdated. The new logo, in the KU colors, shows the acronym of our Department (MIP, Molecular and Integrative Physiology) and captures the global study of the physiology of the human body as represented by the modified letter “P”. Also, the logo gives the idea of a magnifying lens exploring cells. The logo was designed by A. Blanco. YEAR IN REVIEW 2015-2016

The Department of Molecular and Integrative Physiology has continued under the leadership of Dr. V. Gustavo Blanco, who served as Interim Chair. During this past year, the Department has continued to hold up well, despite the continuing difficult funding environment at the Federal and State levels. There have been some changes with respect to the Department’s size. As expected, two Faculty members have retired. In addition, two transferred to the Surgery Department and another one obtained a position in another University. Research programs in the various areas have continued vibrant and the Department has maintained a consistent level of publications. Also, the Department has had another important year with respect to teaching. In the medical curriculum, the Department has maintained its long lasting presence in the Cardiopulmonary and Renal-Endocrine modules. Dr. John Wood and Dr. Michael Wolfe, who lead these modules, have played an instrumental role in leading these sections of the curriculum. In addition, several of our Faculty has been recognized with important teaching awards for their efforts in medical education. Our Faculty has also continued playing a major role in the core IGPBS curriculum and they have been teaching different Advanced Physiology courses. The outstanding teaching reviews from the students show the commitment of our Faculty to high quality teaching, something that we are always very proud of. Regarding service, our Department has continued contributing by serving in numerous key committees at our University, which are essential to the mission of our School. At the national and international level, our Faculty has: 1) served on different review panels from the National Institutes of Health and other funding agencies, 2) participated on editorial boards for different journals, and 3) have disseminated the work performed in our Department through presentations at other Universities as invited speakers, and at national and international meetings. We are very grateful to our faculty, who with their talent and hard work, have continued maintaining the academic success of our Department during the 2015-2016 period.

It is also important to highlight the excellent support that the Department received from our office staff, despite a reduction in personnel in this area. Special thanks go to our Senior Administrator, Shari Standiferd for her continuous dedication and hard work.

We would like to specifically recognize our benefactor and continuous supporter, Jim Osborn, as well as his wife Marion, and his daughter Kathleen. His generous contributions over the years have significantly contributed to our success. The Kathleen M. Osborn Endowed Chair, the Marion M. Osborn Professorship, and the lectureship in honor of Kathleen M. Osborn that Jim established have made a great difference to our Department. All members of our Department immensely appreciate the exceptional support of Jim Osborn and his family.

HIGHLIGHTS FROM THE 2015-2016 ACADEMIC YEAR

RESEARCH FUNDING: Most of our research programs are supported by major external funding. This is something that we are proud of, especially taking into consideration the extremely difficult current funding environment. We owe this to the dedication of our Faculty, who has continued crafting high quality research grant proposals. Based on data provided by KUMC Enterprise Analytics, total NIH funding was $3,765,688, which is lower than that of last year ($4,223,748). The latest data available shows that our Department was ranked 42nd at the national level for NIH research funding among 82 medical schools receiving NIH funding. While this is a good position for our Department, we would like to improve this ranking. Based on data obtained from QlikView, during fiscal year 2016, grant requests from the Department indicated total costs of $34,667,506 (which included a $26,143,005 in direct costs and $8,524,501 in indirect costs). From these, $30,549,185 corresponded to grant proposals submitted to the National Institutes of Health.

EDUCATION: Continuing a long lasting tradition, the Physiology Department has had another outstanding year with respect to teaching. As mentioned previously, Dr. John Wood and Dr. Michael Wolfe led two key modules of the medical curriculum, the Cardiopulmonary and Renal-Endocrine modules. The Cardiopulmonary module received from the medical students an award for being the best module, presented to

the Department at the “Grande Affair” celebration on March 21, 2015. At this same event, two of our Faculty members, Dr. John Wood and Dr. Gustavo Blanco were recognized with ”Student Voice Awards” for their excellence in teaching. These awards are a testimony of the long lasting commitment of our Faculty members to education.

TENURE TRACK APPOINTMENTS: No new primary tenure track appointments were made this year. We have however new appointments from Stowers Institute of Medical Research. These include: Randal Halfmann, Ph.D., started as Assistant Professor on April 1, 2016. His research focuses on determining the mechanisms by which proteins assemble to form polymers or aggregates. Nicolas Rohner, Ph.D., started as Assistant Professor on December 1, 2015. His work involves the study of genetic basis of adaptation to environmental changes, focusing on metabolism. RESEARCH TRACK APPOINTMENTS: E. Matthew Morris, Ph.D., was appointed as Research Assistant Professor, beginning on July 1, 2016. Dr. Morris investigates the role of liver energy metabolism in the development of obesity, type-2 diabetes, and cardiovascular disease. JOINT AND ADJUNCT APPOINTMENTS: The following researchers received joint appointments at their current ranks, after they moved from their primary appointments in Physiology. Shawn Frost, Ph.D., Research Assistant Professor. Transferred to Rehabilitation Medicine on July 1, 2015. Shrikant Anant, Ph.D., Professor, started on January 3, 2016, after transferring to the Surgery Department. Shahid Umar, Ph.D., Associate Professor, started on January 3, 2016 after transferring to the Surgery Department. FACULTY PROMOTIONS: Dr. Vargheese Chennathukuzhi was promoted to Associate Professor with tenure starting July 1, 2015. We are very pleased of Dr. Chennathukuzhi’s accomplishments. Following his recruitment to the Department, he has developed a well-funded research program and he has built a strong research group. His research is directed to understanding the molecular mechanism by which uterine leiomyomas are produced. His research seeks to find small molecule inhibitors of pathways involved in uterine fibroid growth.

FACULTY/STAFF DEPARTURES: There have been several departures from our Department. Reasons for departure include, retirements, transfers to other departments, and one Faculty accepted a position at another University. Among the Faculty who retired are: David Albertini, Ph.D. finished his phased retirement period and fully retired on June 30, 2016. He was recruited to KUMC on June 6, 2004. His research was focused on human and animal reproduction with special emphasis on the translation of basic science into the treatment of human infertility. He served as the Director of the Center for Reproductive Sciences in our Institution. We thank Dr. Albertini for his 12 years of service in our Department and wish him the best in his retirement. Norberto C. Gonzalez, M.D., retired on January 3, 2016. He joined the Physiology Department on September 8, 1970. Since then, his research was directed to understand the molecular mechanisms of adaptation of intact organisms to alveolar hypoxia, a condition that commonly occurs in altitude, or in some types of pulmonary disease. Dr. Gonzalez had a stellar career at KUMC. He is not only a renowned investigator nationally and internationally, but also he is an outstanding teacher, who has contributed immensely to the educational mission of our Department. We wish him the best retirement after his 44 ½ years of dedicated service. The following members of the Department made transitions to other KUMC departments: Shrikant Anant, Ph.D., Professor, transferred to the Surgery Department on January 3, 2016. Dr. Anant continues working in the area of cancer and he maintains a joint appointment with our Department. Shahid Umar, Ph.D., Associate Professor, also transferred to the Surgery Department on January 3, 2016. He is also working in the area of cancer, with emphasis in colon cancer and maintains a joint appointment with our Department. Prabhu Ramamoorthy, Ph.D., Research Assistant Professor, transferred to Surgery on January 3, 2016. Dharmalingam Subramaniam, Ph.D., Research Assistant Professor, transferred to Surgery on January 3, 2016. Satish Ramalingam, Ph.D., Research Assistant Professor, departed on August 25, 2015 and returned to India. Shawn Frost, Ph.D., Research Assistant Professor, transferred to Rehabilitation Medicine on July 1, 2015 and maintains a joint appointment with our Department. Among our Stowers Affiliate Faculty, Rong Li, Ph.D., Professor, departed on July, 2015 to start at Johns Hopkins University in Baltimore, MD. One of our Faculty, T. Rajendra Kumar, Ph.D., Professor, was recruited by the University of Colorado Medical Center. He left on April 30, 2016. He served as the

Director of the Center for Reproductive Sciences at KUMC. He now obtains a position as Professor & Associate Vice-Chair of Research in the Department of Obstetrics & Gynecology at the University of Colorado Medical Center in Aurora, Colorado. FACULTY AWARDS/ACCOMPLISHMENTS: Numerous members of our Department have been serving on review panels from NIH and other agencies. They have also served on the editorial board of different journals and presented invited lectures at national and international meetings. Dr. Lane Christenson has been involved with the organization of the research program for the Society for the Study of Reproduction annual meeting. GRADUATE PROGRAM AND PHYSIOLOGY SOCIETY: The student led Physiology Society continued functioning well this year. A new President was elected and a new committee was chosen. The leadership of the Physiology Society is currently the following: Joshua Curry, President Asona Lui, Vice-President Mina Farahbakhsh, Social Chair Younshim Park, Stowers Chair The graduate students in the department had another active year. In 2015-2016 three new students were recruited to the department. These include students who are working with our affiliate members at Stowers Research Institute. Currently, our Department has a total of 28 enrolled doctoral students. Several of our students were able to obtain external fellowships to support their training. Funding came from different sources, including the SELF Fellowship program, the KUMC Biomedical Training Program, NSF, and NIH. The new graduate students that joined the Department in the fall of 2015 included the following individuals (their respective mentors are listed in parenthesis): Zahraa Alali (Dr. Warren Nothnick) Younshim Park (Dr. Kausik Si) Afreen Sayed (Dr. Shrikant Anant) The Cancer Biology program was formally approved by the Kansas Board of Regents and students from our Department moved to Cancer Biology in 2016. These include: Eric Young (Dr. Danny Welch), transferred on January 2016 Afreen Sayed (Dr. Shrikant Anant), transferred on June 2016 Wahid Mulla and Akshay Narkar (Dr. Rong Li) transferred with their mentor to Johns Hopkins in August 2015. Five students completed their PhD degrees during the fall of 2015 and the spring of 2016. Congratulations to all of them and their mentors. These are listed below:

Summer 2015 Fall 2015 Spring 2016 No Graduates Mohammed Khan – PhD Swathi Iyer - PhD Robert Rogers - PhD Danny Miller – PhD Nairita Roy - PhD Mohammed Khan, Ph.D. (August 19, 2015) received his degree with Dr. Si. The title of his dissertation was “Role of Drosophila ORB2 (CPEB) in Synaptic Protein Synthesis”. Dr. Khan is now a Postdoctoral Fellow with Dr. Valina Dawson at Johns Hopkins University in Baltimore, MD. Robert Rogers, Ph.D. (September 1, 2015) received his degree with Dr. Geiger. The title of his dissertation was “Heat Shock Proteins in Metabolic Disease – Skeletal Muscle, Liver and Adipose Tissue Cross Talk”. Dr. Rogers is now a Postdoctoral Fellow in Dr. Hiroshi Nishimune’s research lab in the Anatomy and Cell Biology department of KUMC. Nairita Roy, Ph.D. (November 10, 2015) received her degree with Dr. Swerdlow. The title of her dissertation was “Investigating the Presence of Mitochondrial DNA Methylation, Its Alteration and Association with Mitochondrial Transcription and Mitochondrial Function”. Dr. Roy is now working as a Postdoctoral Fellow at the University of Pittsburgh, PA. Swathi Iyer, Ph.D. (March 24, 2016) received her degree with Dr. Iwakuma. The title of her dissertation was “Roles of TMIGD3 and A3AR in the Suppression of Osteosarcoma Malignancy”. Dr. Iyer is currently working as a Postdoctoral Fellow in Dr. Tomoo Iwakuma’s research lab in Cancer Biology at KUMC. Danny Miller, Ph.D. (April 4, 2016) received his degree with Dr. Scott Hawley from Stowers. The title of his dissertation was “A Genomic Analysis of Meiosis in Drosophila Melanogaster”. Dr. Miller is working on his MD degree at KU Medical Center. Prepared by:

Andrei B. Belousov, Ph.D., Associate Professor We are interested in the mechanisms of regulation of neuronal gap junctions (electrical synapses) during development and neuronal injury. We also study the role of neuronal gap junctions in death of neurons in the developing nervous system and in the secondary, glutamate-dependent neuronal death following ischemic stroke, traumatic brain injury and epilepsy. As a result of these studies, a novel model of the mechanisms of glutamate-dependent neuronal death has been proposed, where neuronal gap junctions play a critical role.

V. Gustavo Blanco, M.D., Ph.D., Professor & Interim Chair Our laboratory studies the role of ion-transport proteins of the plasma membrane in

cell function. Research is focused on the Na, K-ATPase, a plasma membrane enzyme

system that uses the energy from ATP to establish and maintain the high internal K+

and low internal Na+ concentrations characteristic of most animal cells. Specific

research projects include:

1). Mechanism of function, regulation and role of the testes-specific isoform of the

Na,K-ATPase (Na,K-ATPase α4) in sperm function and male fertility. We discovered

Na,K-ATPase α4, characterized its expression and determined its function as

essential for sperm motility and male fertility. Because of the importance of α4 in

sperm physiology, we are pursuing experiments directed to utilize Na,K-ATPase α4 as

a marker for male fertility; and as a target for male contraception. We are trying to

develop specific inhibitors of the Na,K-ATPase α4 that can block sperm motility for

their use in male contraception. Overall, this project is important to understand sperm

physiology, male fertility and contraception.

2). Role of the Na,K-ATPase in autosomal dominant polycystic kidney disease

(ADPKD). In this project, we are investigating the role of the Na,K-ATPase in cyst

generation and progression in ADPKD kidneys. We have found that in ADPKD, the

Na,K-ATPase exhibits an abnormally increased sensitivity to ouabain, a hormone

released by the adrenal glands. We discovered ouabain as a circulating factor that

promotes ADPKD cystic phenotype. We are currently studying the intracellular

pathways involved in ouabain effects and the mechanisms of action of ouabain in

ADPKD cells, with the idea to use this information to pharmacologically interfere with

the pro-cystogenic actions of ouabain to treat ADPKD.

Neurophysiological techniques are used to investigate the functional contribution of neurons in the cerebral cortex and brainstem to the control of voluntary movement. The spike (action potential) activity of single neurons is recorded in awake monkeys trained to perform various movement tasks. Computerized analysis techniques are used to reveal the functional contribution of a neuron or localized groups of neurons to movement. In another project, SIV infection in monkeys is used as model of neuro-AIDS. This model is used to investigate interactions between SIV infection and drugs of abuse using neurobehavioral, neurophysiological, and neuroanatomical methods. Vargheese M. Chennathukuzhi, Ph.D., Associate Professor My research interests include uterine fibroids, fertility and contraception. Uterine fibroids are the most common tumors of the female reproductive tract and are the leading indication for hysterectomies in the US. Sadly, there is no approved drug to treat uterine fibroids chronically. Our laboratory is trying to understand the regulation and function of GPR10, a G protein-coupled receptor, aberrantly expressed in uterine fibroids. We have generated transgenic mice overexpressing GPR10 in the myometrium in order to understand its role in the pathogenesis of fibroids. Transgenic-hGPR10 mice develop uterine fibroids that are phenotypically identical to the human disease. We plan to use this preclinical model for the development of small molecule modulators of GPR10 as treatment for fibroids. In addition, we have identified the molecular machinery that regulates aberrant expression of GPR10 in uterine fibroids. We recently developed a conditional knockout model for Rest, the transcriptional repressor of GPR10. Rest cKO mice are infertile and develop uterine fibroid phenotype. We have also identified PRICKLE1 as the upstream regulator of REST stability and function in the uterus. Our laboratory is trying to validate the role of environmental estrogens in the suppression of Prickle1 gene and REST protein expression using rodent models. Our laboratory is also interested in molecules that regulate sperm function and male fertility. We are studying functional regulation of the sperm specific sodium-proton exchanger (sNHE) in expression systems that utilize chimeric channels. Lane K. Christenson, Ph.D., Associate Professor Fertility, infertility and ovarian cancer are major health concerns for women. Research in my laboratory focuses on understanding the terminal events involved in follicular development and ovulation. The studies have direct implications in development of a fertile healthy oocyte, contraception and disease conditions such as polycystic ovarian syndrome and ovarian cancer. Current studies focus on understanding the role extracellular vesicles (exosomes and microvesicles) play in ovarian function (oocyte maturation, granulosa cell function) and in ovarian cancer. The laboratory is also examining how post-transcriptional regulatory processes such as RNA editing impacts oocyte developmental competence. These studies use the latest state of the art technologies, including next-gen-sequencing, nanoparticle analysis in addition to standard molecular and biochemical techniques. The laboratory also uses a comparative approach using human, bovine and murine cells and animal studies in both cows and mice to gain further insights into the reproductive biology of the ovary. Paige C. Geiger, Ph.D., Associate Professor Diabetes affects 23.6 million Americans (7.8% of the population); and by 2030, the global numbers are projected to increase above 400 million. A progressive decline in the body's ability to detect and respond to stress could result in the development and/or acceleration of a number of metabolic disease states, including insulin resistance and obesity. Heat shock proteins (HSPs) are best known as molecular chaperones, but they also play key anti-inflammatory and cell signaling roles. Patients with type 2 diabetes demonstrate reduced expression of heat shock protein 72 (HSP72) and a blunted exercise-induced heat stress response. Previous studies from our laboratory and others demonstrate that the induction of HSPs via heat treatment, transgenic overexpression, or pharmacological agents is protective against high fat diet-induced skeletal muscle insulin resistance. Planned studies should provide compelling new information about the relationship between heat shock proteins, aerobic capacity and metabolic function. As a result, new strategies could be developed to modulate HSP-dependent pathways as a therapeutic approach to treat insulin resistance. Meetings Attended:

Leslie L. Heckert, Ph.D., Marion M. Osborn Professor for Reproductive Sciences Research interests have consistently focused on the transcriptional and cell-signaling processes for proper function and development of the gonads, with the goal of extending our mechanistic understanding of the biology of fertility, in hopes to facilitate clinical improvements for contraceptive options and diagnosis and treatment of infertility. More recently, I have developed a significant interest in establishing a collaborative program with Dr. Jill Jacobson, M.D. at Children’s Mercy Hospital. The intent is to develop a program that integrates both clinical and basic research to improve our understanding of the mechanisms underlying disorders of sex development and gender dysphoria. Melissa A. Larson, Ph.D. Research Assistant Professor As the Technical Director of the Transgenic and Gene-Targeting Institutional Facility, my role is largely defined by service. I am responsible for providing services in the generation of gene-modified mice, sperm and embryo cryopreservation, assisted reproductive technologies, and embryonic stem cell propagation and targeting. We provide these services for investigators at KU Med and KU-Lawrence, as well as the entire Kansas City metro area. In addition, I am interested in exploring and implementing new technologies that allow creation of unique mouse research models. Presentations:

The major focus of my research is to characterize structural, functional, physiological changes linked to disease activity and progression using advanced in vivo magnetic resonance imaging techniques. Warren B. Nothnick, PhD, HCLD, Professor Endometriosis is a disease common in women of reproductive age characterized by pain and infertility. Despite its high prevalence, those factors which contribute to the development and progression of the disease remain unknown. The focus of my research is to examine the role of microRNAs, small RNAs which post-transcriptionally regulate gene expression, in the pathogenesis of the disease as well as examine their utility as prognostic markers and therapeutic targets. Stanford, Ph.D., Associate Professor My research is focused on preclinical models of normal aging and age-related diseases and conditions that affect motor function, such as Parkinson’s disease (PD), Amyotrophic Lateral Sclerosis (ALS) and neonatal hyperbilirubinemia (kernicterus). We are currently examining the effects of isometric strength training on neuromuscular denervation in aged rats and in the SOD1-G93A rat model of ALS. Our most recent studies involve characterizing behavioral and neurological abnormalities in the Gunn rat model of kernicterus. Meetings Attended:

Joseph S. Tash, Ph.D., Professor My research interests, funded by NIH, focus on the discovery and development of reversible non-hormonal male contraceptives. In addition, my NASA funded research is focused on elucidating the impact of long term space flight on reproductive health in males and females. During the last year I was awarded a 6 month Sabbatical by the university, which was supported by a Burroughs-Welcome Fund Collaborative Travel grant. I devoted the Sabbatical to gain experience in female reproductive biology at the Oregon National Primate Research Center in collaboration with the investigators in the U54 Center: Contraception by Blockade of Periovulatory Events in Primates. During the Sabbatical we accumulated promising data to support development of a new area of non-hormonal contraceptive drug discovery in females. Meetings Attended:

Michael W. Wolfe, Ph.D., Associate Professor Research in my laboratory has recently been focused in two areas. The first investigates the role of estrogen receptor alpha in growth and metabolism using receptor knockout rats. We are focusing on sex-specific differences in hepatic gene expression. In the second area, we are determining the role of SATB homeobox proteins in regulating placental development. These studies focus on SATB regulation of the epigenome and transcriptome in proliferating and differentiating trophoblasts. Meetings Attended:
