FloGel(FGIS-2)Fluorescent Gel Image System

步驟 : FloGel 接上電腦 (PC) ，由 PC 來控制。1. 打開總電源，暖機 10 分鐘 (CCD,themal printer,PC 開機，準備拍膠 )2. 一手戴手套拿 EtBr gel ，一手不戴打開 FloGel 的門，以保 持整個影像系統的乾淨不受 EtBr 的汙染。3. 將 Gel 擺在黑色玻璃 (UV filter) 的中央。

4. 打開 White light 開關至 (1) 的位置。

5. 在 PC 上打開 Scion Visicapture 。6. 按 Image – Start Live capturing 會出現左下面視窗。7. 按 Image – Show properties 會出現右下視窗。

8. 按 Preview – 影像縮小，可以看到 Gel 全部。9. 看著 PC moniter 調正 Gel 。10. 黑色 Filterwheel box ，旋轉到 EtBr filter ，或是其他的 filter e.g.

SyBr Green filter (option)

光圈

Filter wheel

Focus 細調

Zoom 放大縮小

11. 旋轉伸縮鏡頭 Zoom

1) 光圈調到適當 2)Zoom 放大縮小至適中 3)Focus 細調對焦至最清晰影像12. 關門，關 white light 開關至 (0) 的位置。13. 打開 UV light 開關至 (I) 的位置。14. 光圈調到最大，看影像是否清晰，若還要調整可重複 no,11

15. 按關掉 Preview ，讓影像回復正常大小。

如何設定影像內容 ?

選擇 Image—Show Properties

出現 Camera Properties ( 主要由 Setting 中的 Gain,Contrast,Black,Exposure, 來調節影像的清晰度 )

Readout Speed : 讀出速度，常用為 14MHz ，有效的為 7,14 MHz

Image Depth : 調色澤濃度，可設 8,10,12-bits s

Video signal 能夠被補償去產生 nature-tone images ；

當使 Video signal 失去能力則沒有補償且其輸出會成直線低。

Average : frame 數的平均Sum : frame 數的總合

Fame 數的累積 範圍從 2~65535

Preview : 680x512 image size preview ，此會增加 capture rate ，

有助於 camera 的定位和對焦。 Preview mode 不能被設定為 Bin mode 。

Bin : 680x512 的分辨率，較明亮的影像。除了 Preview 外，所有的 property setting

皆可在此模式中被調節。影像在 Bin mode 中不能被設定為 Preview mode 。Test Pattern : 測試 Camera 用。

Reset : 使所有的 Statistics 回到原始設定值

Camera Info : 現在選擇的 camera 的一般資訊

Camera : camera 的樣式

Revison : Camera 內部的校正數值

Serial Number : Camera 內部的連續數值

CCD : CCD 的樣式 Height : CCD chip 的高度

Width : CCD chip 的寬度 Depth : CCD chip 的深厚度
