The in vitro rooting phase takes place under sterile and fully conditioned circumstances making the VitroPlug a unique system. Introduction Vitro Plus The Vitro Plug 126 is the innovative method to cultivate and to process all conceivable seed- and tissue cultures. This technique has been developed in cooperation with Jiffy, Vitro Plus and Visser int. The unique grafting tech- nique for ferns and comparable crops has been patented, making use of the plug only possible under license. Products grown using the Vitro Plug system: Can be distributed worldwide directly from the growing chamber. Are suitable for automation. Are more reliable due to easier and faster hardening off process. The customer always should check if the radiation sticker has turned red and if the packaging is undamaged. If the sticker hasn’t turned red and/or the packaging is damaged the plugs shouldn’t be used. Unpack the bags with trays under Laminair flow cir- cumstances only, opened bags must be sealed airtight again before the remaining trays are taken out of the laminair flow. Destacking of the trays can be automated. If destacking is done manualy this needs to be done under sterile conditions (sterile gloves, pincets etc). As a standard, 400 ml liquid medium is dispersed evenly as possible between the plugs at a temperature of 50 degrees Celcius. The composition of the medium and PH are the same as the Stage III medium used for the particular crops in tissue culture, Agar is left out. Reopening of temporary stored trays filled with medi- um increases the risk of infection. Sealing the trays can be done with a hand sealer which should be placed within the sterile laminair flow. Incubation of plants in the growing chambers can take place under normal tissue culture circumstances.
