Frequently Asked Questions about DIM-PRO PLUS What is Diindolylmethane? Diindolylmethane , or DIM, is a phytonutrient, found in broccoli and other cruciferous vegetables such as Brussel’s sprouts, cabbage and cauliflower. What does DIM do in the body? DIM influences the metabolism of estrogens in the body. This is an important health issue because estrogen has many effects on cell growth, health and function of estrogen responsive tissues (breast, uterus, ovaries, cervix, prostate, testes, and bone). There are three forms of naturally occurring estrogen compounds made in the body – estradiol, estrone, and estriol. There are also xenoestrogens; synthetic estrogen-like compounds that enter the body through the ingestion and/or inhalation of pesticides, drugs (hormone replacement therapy), fuels, and plastics. Combined, these estrogens represent the body’s systemic estrogen pool. Estrogen, whether synthetically derived or naturally formed, is broken down through detoxification pathways in the liver and eliminated from the body. This is a crucial step in estrogen metabolism as excessive exposure to circulating estrogens in the blood can trigger aberrant cell growth and function. There are two different detoxification pathways in the liver by which estrogen can be metabolized. One pathway yields estrogen derivatives known as 2-hydroxyestrone. The other pathway yields a 16-hydroxyestrone derivative. The 16-hydroxyestrones have been associated with breast and uterine cancers, cervical dysplasia, uterine fibroids, PMS, endometriosis, and obesity. These conditions are often the result of estrogen over-stimulation or dominance in the body. DIM works by positively influencing the production of 2-hydroxyestrone metabolites. As DIM increases these “good” estrogen metabolites, there is a simultaneous reduction in the levels of the 16-hydroxyestrone form. This shift in estrogen metabolism, evidenced by an increase in the 2:16 hydroxyestrone ratio, reduces the undue influence of the 16-hydroxyestrone metabolites on cell growth and function. What is BioResponse-DIM? BioResponse-DIM is a patented “Phytosorb” delivery system that is the only clinically proven method known to provide predictable absorption of DIM. In a recent human study as much as five times more diindolylmethane was absorbed from BioResponse-DIM, than from crystalline diindolylmethane. Could I obtain the same healthful benefits from DIM by simply eating more broccoli? Consuming a diet rich in cruciferous vegetables is beneficial form many reasons. These vegetables are rich in vitamins, minerals and fiber which promotes a healthy bowel. However, active therapeutic levels of DIM found in 2 capsules of BioResponse-DIM is equivalent to consuming over two pounds of raw broccoli. Consuming this amount of broccoli a day may be problematic for some. Why is glycine added to DIM-PRO PLUS? Glycine is an amino acid that is utilized in Phase II detoxification pathways in the liver. Through glycination, glycine forms a conjugate with a metabolic byproduct or drug rendering the newly formed compound water soluble and ready for elimination from the body through the urine and feces. Glycine is also a precursor amino acid that combines with glutamic acid and cysteine to form glutathione. Glutathione conjugation is a major Phase II detoxification pathway that aids in the removal of toxic intermediates. ® 11312 200th St. East Graham, WA 98338 Phone: 800-753-4145 Fax: 866-817-9931 www.medifood.info ®

What does DIM do in the body?DIM influences the metabolism of estrogens in the body. This is an important healthissue because estrogen has many effects on cell growth, health and function ofestrogen responsive tissues (breast, uterus, ovaries, cervix, prostate, testes, and bone).

There are three forms of naturally occurring estrogen compounds made in the body – estradiol, estrone, and estriol. There are also xenoestrogens; synthetic estrogen-like compounds that enter the body through the ingestion and/orinhalation of pesticides, drugs (hormone replacement therapy), fuels, and plastics. Combined, these estrogensrepresent the body’s systemic estrogen pool.

Estrogen, whether synthetically derived or naturally formed, is broken down through detoxification pathways in theliver and eliminated from the body. This is a crucial step in estrogen metabolism as excessive exposure to circulatingestrogens in the blood can trigger aberrant cell growth and function.

There are two different detoxification pathways in the liver by which estrogen can be metabolized. One pathwayyields estrogen derivatives known as 2-hydroxyestrone. The other pathway yields a 16-hydroxyestrone derivative. The 16-hydroxyestrones have been associated with breast and uterine cancers, cervical dysplasia, uterine fibroids,PMS, endometriosis, and obesity. These conditions are often the result of estrogen over-stimulation or dominance inthe body.

DIM works by positively influencing the production of 2-hydroxyestrone metabolites. As DIM increases these“good” estrogen metabolites, there is a simultaneous reduction in the levels of the 16-hydroxyestrone form. Thisshift in estrogen metabolism, evidenced by an increase in the 2:16 hydroxyestrone ratio, reduces the undue influenceof the 16-hydroxyestrone metabolites on cell growth and function.

DIM-PRO PLUSDim-Pro Plus nutritionally supports the activity of enzymes that metabolize estrogen and aspirin. Current research demonstrates that BioResponse diindolylmethane increases levels of good estrogens (2-hydroxyestrogens/2-OHE1) which tend to inhibit abnormal cell replication while reducing levels of “bad” estrogens (16-a-hydroxyestrone/16-a-OHE1 and 4-OH estrone).*

What is Dim?

Dim or 3,3’-diindolylmethane is a natural product produced during the autolytic breakdown of glucobrassicin, one of over 100 sulfur containing glucose conjugates, known as glucosinolates that occur exclusively in cruciferous vegetables such as broccoli, Brussels sprouts, cabbage, kale and mustard greens.

The immediate precursor of DIM in the plant is I3C or indole-3-carbinol, which is liberated from glucobrassicin by the action of myrosinase, an enzyme found in the cell wall of cruciferous vegetables, following disruption of the sub-cellular compartmentalization of enzyme and substrate via mastication.

Indole-3-carbinol undergoes extensive and rapid self-condensation reactions under the acidic conditions of the stomach to form several derivatives with distinctive biological activities.*

DIM is a major condensation product of I3C in vitro. DIM is also produced in vivo since it has been readily detected in the livers and feces of rodents administered I3C whereas I3C was not detected.* Biological Actions of DIM on Estrogen Metabolism

1. DIM alters estrogen metabolism in humans by up-regulating a specic cytochrome P450 enzyme (CYP1A1), which speeds up the conversion of estrone to 2-OH estrone (a "good" estrogen) at the expense of the conversion to 4-OH estrone (a "bad" estrogen) and 16-alpha OH estrone, also a cancer causing estrogen metabolite. Breast and prostate cancer risk increases if the ratio between 2-OH and 16-alpha OH estrone is skewed in favor of 16-alpha OH estrone production by the respective cytochrome P450 enzymes.*

2. Up-regulating CYP1A1 using DIM is especially desirable if the woman has a single nucleotide polymorphism on CYP1B1, which preferentially converts estrone into 4-OH estrone, another estrogen metabolite which increases a woman's risk for breast cancer by creating DNA adducts.*

In summary, DIM enhances the production of 2-OH estrone. As DIM increases these "good" estrogen metabolites, there is a simultaneous reduction in the levels of 4-OH estrone especially if a woman has a SNP on CYP1B1 (roughly 1 in 3 women do) and 16-alpha-OH estrone. This shift in estrogen metabolism, evidenced by an increase in the 2-OH :16 alpha OH estrone ratio and reduction in 4-OH estrone levels, reduces the risk of unregulated cell proliferation and growth and DNA adducts, respectively.*

Dierence between DIM and BioResponse-DIM (DIM-Pro Plus by Functional Integrative Therapies)

Crystalline diindolymethane is hydrophobic. It does not go into solution easily and therefore is poorly absorbed by the human body. BioResponse-DIM, on the other hand, is a patented delivery system wherein DIM is wrapped in phosphatidyl choline (lecithin) boosting absorption as much as ve times compared to crystalline DIM. DIM-Pro Plus by Functional Integrative Therapies contains not only BioResponse-DIM (75 mg per capsule) but also the amino acid glycine. Glycine is utilized in a Phase II detoxication process called glycination, where it forms a conjugate with various naturally occurring metabolic byproducts or pharmaceutical drugs to form water soluble compounds ready for elimination from the body through the urine and feces. In addition, glycine combines with glutamic acid and cysteine to form glutathione, an important tri-peptide in the removal of toxic heavy metals like mercury, lead and cadmium, pesticides, herbicides and insecticides via glutathione conjugation, another Phase II detoxication pathway.*

Who can benet from BioResponse DIM?

Women who are taking hormone-replacement therapy.*Women who are taking bio-identical hormones.*Women diagnosed with ER+ breast cancer and worried about recurrence.*Women considering or who have had IVF therapy.*Women who are taking birth control pills.*Premenopausal women who have severe menstrual signs and symptoms.*
