Challenges in Proteomics
2006-2007
Mise en place d’une nouvelle génération d’instruments :
- LTQ-FT-Orbitrap
Développement de nouvelles stratégies :
- Identification de protéines de faible abondance
Un spectromètre de masse hybride de nouvelle génération : LTQ-FT-Orbitrap
• Mass resolution > 60,000 (FWHM) at m/z 400 at 1 sec cycle
• Max. resolution > 100,000 at m/z 400 in a full scan
• Mass accuracy < 5 ppm external calibration (2 days old)• < 2 ppm internal calibration (lock mass)
• Mass range : 50 – 2,000; 200 – 4,000
• Sensitivity : sub-femtomol on column
• Dynamic range : 3000 in a single scan
• Throughput : 3 accurate mass scans per second or 4 scans per second parallel detection (1 HR Orbitrap scan + 3 LR LTQ MS/MS scans)
