The Lumat LB 9507 tube luminometer provides thesensitivity and versatility you need in a generalpurpose luminometer.

Advanced digital photon counting detection providesan extended dynamic range with linearity over morethan six decades.

Validated DLReadyTM, the BERTHOLD Lumat isideally suited for reporter gene assays as wellas all other important luminescence–basedapplications in research and diagnostics.

Low noise light detector The detection unit of the Lumat consists of astate–of–the–art ultra fast single photon countingphotomultiplier (PMT) with low noise. A selectionprocedure for the PMT tubes guarantees optimum performance and reliability of all instruments.

Unique design of measurement chamberThe special geometric design and the use of selectedcoatings in the measurement chamber providessuperior light collection efficiency.

The measurement chamber consists of 2 tube positionsin a rotating holder: while one sample is measured asecond tube can be placed in the loading position,resulting in a higher throughput comparable to auto-matic systems.

Variable volume injectorsThe Lumat is equipped with reagent injectors providingoptimum reliability and simplicity in liquid handling.

Based on the proprietary JET injection technologythe injectors combine high speed and reliablemixing of reagents with outstanding precision. Theinjected volume can be adjusted within the range25 to 300 µl.

The materials involved are able to withstand evencorrosive materials which are often used in chemiluminescence assays. The ”Unload” operationhelps the user to recover expensive reagents byreturning them back to the reagent reservoirs.

Injector cleaning solutionsFor proper maintenance of the injection system theCleanit solutions are recommended for regularcleaning. Daily cleaning ensures that accuracy andprecision as well as long life cycle of the injectors,will be maintained.

SoftwareThe software has been developed to provide easy,intuitive operation of the instrument’s powerfulfeatures. You can choose from a variety of basicmeasurement protocols.

Raw data protocolRecommended when the significance of the measurement is the integrated value of light emission over the measurement time – e.g. ATPmeasurements or reporter gene assays.

Dual label protocolAllows the automatic injection of reagents, thesequential measurement of two differentluminescent signals and the ratio calculation.

Kinetic measurementsCan be performed in three modes:1. Kinetics of a single sample within a preset time

window with a printout of individual values andreaction curve

2. Repeated operation to monitor long term reactions3. Endless counting of a single sample

Cut–off protocolCan be used for qualitative characterisation ofsample data by flagging result with attributes,

e.g. ”Positive”, ”Negative” or ”Intermediate”according to the preset limits.

Quantitative protocol (LIA, ILMA)A software option for full and convenient data evaluation is required for luminescent immuno-assays. It includes the use of measured standards,master curves and controls. Concentrations ofpatient samples are calculated according to anautomatic smoothed spline function including average calculations of replicates.

Data handlingThe measured and calculated sample values can beoutput to the built–in printer or to a computer viaserial interface with the WinTerm software.

QC luminescence test kitWith the QC luminescence test kit theperformance of your luminometer canbe checked. This is the ideal qualitycontrol (QC) method as performance ofboth injection and detection systems canbe monitored. The test kit follows a fast flash–type kinetic. The test kit contains a ”label” and two types ofstarter reagents.

Applications

Reporter Gene AssaysIn basic research of generegulation as well as in drugdiscovery the use of luciferases,ß–glucuronidases, ß–galacto-sidases and secreted alkalinephosphatases have become astandard tool offering the highest sensitivity.

Especially dual luminescence type assays, e.g. Dual–Luciferase® Reporter Gene Assay, havebecome a favourite means as they provide aninternal control for transfection efficiency orgeneral expression level.

ATP determinationA detection limit of less than 1 attomole of ATPper tube makes the Lumat one of the best suitedtube luminometers for the determination of cellviability, e.g. in tumour chemosensitivity assays,cell proliferation, antibiotic susceptibility testingor hygiene monitoring.

Luminescent immunoassays (LIA, ILMA)By exchanging colorimetric substrates ofhorse–radish peroxidase or phosphatases withluminescent ones a 100–fold increase in sensitivitycan be achieved. For immunoassay data evaluation the Lumat can be equipped withsophisticated and easy–to–use software.

DNA probe assaysSeveral diagnostic DNA probe assays based onluminescent read–out (e.g. Acridinium esters,Luminol) provide the most sensitive detectionand diagnosis of infectious diseases.
