BEYENE Dereje Degefie

Mr. Beyene’s research focused on “characterization and expression analysis of Isoamylase1 (Meisa1) starch gene in cassava and its wild relatives”. In this study a cDNA clone of a Manihot esculenta Isoamylase1 gene, a member of starch-debranching enzyme family in cassava storage root was characterized. The gene was observed to exist in two copies in the cassava genome. The gene expression level varies according to developmental stages of tissues (leave, petiole, stem, fibrous roots and storage roots) and storage roots. The gene is dominantly expressed in fibrous root other than other parts of the plant. Meisa1 expression reaches the peak during the early stages of storage root thickening after which it declines to undetectable levels as the storage root thickens. The overall expression pattern study showed that Meisa1 is involved in regulation of granule initiation and root differentiation into storage roots. The SNPs analysis of Meisa1 and G3pdh another gene involved in sugar metabolism show greater genetic diversity in cultivated than wild germplasm. The abundance of Meisa1 in fibrous root and early stage of fibrous roots differentiation to storage roots might be associated with storage root enlargement. Meisa1 was observed as more diverse in cultivated cassava than that in subsp. flabellifolia whereas G3pdh was more diverse in wild than cultivated. This may indicate the importance of Isoamylase1 in starch quality traits in cassava, a trait which is likely to have been the target for artificial selection by farmers and breeders, in addition to natural selection. Key findings of his studies are envisaged to be resourceful in exploitation of cassava’s full potential for various food and non-food starch applications.

Her pioneering research links for the first time sweetpotato root chemical composition and resistance. This research funded by the McKnight Foundation in collaboration with NARO, has provided basis on which breeding for weevil resistant sweetpotato varieties is to be done.

Conferment of the Degree of Doctor of Philosophy (Agricultural Extension Education)

Mr. Okiror’s research focused on the Primary School Agriculture (PSA) curriculum since primary schooling is the terminal level of education for majority of pupils. Vocational agricultural education in Uganda lacks a supervised practical component for transfer of learning. The study compared performance of pupils with school-based gardens and home gardens. Findings showed Supervised Agricultural Experience Projects (SAEPs) improve transfer of learning and that pupils effectively passed gardening information to parents despite English language constraints, timidity and parents to discounting school messages. The study showed that schools can contribute to farming at village level through SAEPs and outreach programs targeting parents. It contributes to the debate on vocational agriculture as a solution to youth unemployment than a dead end competing with priorities of basic Education for All.

Mr. Mbanzibwa’s research focused on assessment of the “genetic variability and population structure of Cassava brown streak disease (CBSD) associated viruses in East Africa”. CBSD continues to be a serious cause of cassava yield losses in East Africa. The causal agent of CBSD was previously thought to be Cassava brown streak virus (CBSV) alone. This study has clearly demonstrated that there are two distinct CBSD-associated viruses, namely CBSV and Uganda CDSV (UCBSV) following full genome sequencing of each of the viruses. The two viruses are spread all over in East Africa but there is predominance of CBSV in coastal and lowland areas while UCBSV is predominant around Lake Victoria. The UCBSV isolates were less virulent on Benthamina tabacum as compared to the isolates of CBSV. The virulence of these viruses observed in high altitude areas is most likely due to availability of susceptible cassava genotypes introduced through breeding programmes. Atypical of the family Potyviridae, CBSV and UCBSV genomes contained a HAM1-like protein located between the replicase and coat protein at the C-proximal region. The finding of the HAM1h in the genomes of CBSV and UCBSV was novel and unexpected that could play a role in colonization of plants in the family Euphorbiacea by viruses. The knowledge on distribution and nature of infection is expected to be of practical significance to cassava breeders in the region as it will make it possible to monitor varieties that are resistant to either or both viruses.

Mr. Tumuhimbise’s research focused on the potential of orange fleshed sweetpotatoes (OFSP) as a source of provitamin A carotenoids which are useful in alleviating vitamin deficiency. The study investigated changes in b-carotene bioaccessibility associated with processing and storage of OFSP and its relationship with the changes in tissue microstructure. In vitro models and in vivo models were used to investigate the amount of b-carotene released from the sweetpotato matrix and the amount finally absorbed in the body. The results showed that processing alters OFSP microstructure and improves b-carotene bioaccessibility. The findings further show that that consumption of OFSP is effective in combating vitamin A deficiency. The support of the supervisors; Prof J. Eng. Kaluuba investigated the fundamental limits of data transmission over a wireless channel and how to overcome this

problem through channel coding. The fundamental characteristic of wireless communications is that the communication path or channel is time-varying due to mobility of the user or of objects in the propagation path, and due to multipath signal fading, interference, and noise effects which degrade the received signal. The solution to the problem is to ensure that the intended message is obtainable from whatever is actually received, which is often a corrupted version. Channel coding, adds extra data bits to information bits to make the transmission of data more robust to disturbances present on the transmission channel. Eng. Kaluuba was able to establish the critical design parameters for robust codes and how to optimize these for best results, through simulation of their mathematical models using MATLAB. Prof. Livingstone Luboobi and Associate Prof. Tabani-Wani Gyavira supervised this work. Makerere University – CARNEGIE Project provided financial support.

Mr. Balyeidhusa’s research was in the field of African Trypanosomiasis. The research investigated the presence of human infective trypanosomes (Trypanosoma brucei gambiense) in domestic animals in northwest Uganda that cause Gambian type of sleeping sickness. No domestic animal examined in northwest Uganda was detected to have human infective trypanosomes. Probably domestic animals do not pose a potential risk to humans as reservoirs of T. b. gambiense parasites. However, control interventions should still consider treating domestic animals, since in West Africa, they have been implicated as reservoirs of T.b.gambiense. Also, the study revealed that tsetse flies feed on both domestic and wild animals. Tsetse control interventions should involve domestic animals. The research was supervised by Prof. John Enyaru and Assoc Prof. Fred Kironde. The study received funding from Makerere University Directorate of Research & Graduate Training, DAAD, and WHO/TDR.

Ms Nakalembe’s research focused on developing an inventory of the indigenous knowledge of indigenous mushrooms of Bunyoro Sub-region, Uganda. She further looked for molecules or compounds

in the mushrooms with potential for nutraceutical and drugs. A combination of standard field and laboratory methods were used in this study. Bunyoro sub-region has over 22 edible species of mushrooms used for food and medicine. Several useful molecules and compounds dominant in the most edible mushrooms were characterized, identified and tested, of which were of medicinal importance. The results of this study explain the indigenous use of these mushrooms in the treatment of various ailments including measles. Therefore, this research work opens further research for development of nutraceuticals, medicines and health products, especially for infants and the elderly. The supervisors of this project were Prof. John David Kabasa, Prof. Deogratias Olila and Prof. Taha Nabil and sponsors DAAD, Carnegie Corporation, SIG through the RISE-AFNNET Program and School of Graduate Studies, Makerere University, for funding this research.

Dr. Sengooba investigated the effect of different oxygen tensions on the growth of the respiratory system of Clarias carsonii and Clarias gariepinus species and its implications on fish farming in Uganda. Randomized laboratory based and field based studies were conducted. The gross anatomical and histological organization of the respiratory organs the gills, labyrinthine organs and suprabranchial membrane were quite similar in the two species. There was a positive correlation between growth of the physical respiratory structures and concentration of oxygen.Clariascarsonii was more adapted to low oxygen concentrations and a thus a promising candidate for aquaculture. The variation in oxygen concentration during the day which arises due to pond management

practices have implication on fish rearing especially for non air breathing fishes.
