The MPure-12 System employs an advanced magnetic bead separation technology that enables rapid and efficient purification of nucleic acids. This process includes four main steps: Lysis, binding, washing and elution. Purifying the nucleic acids with the MPure-12 System takes only 35 to 70 minutes depending on the selected protocol and kit. The MPure-12 System is a standalone benchtop system; no PC is needed. The MPure-12 extracts nucleic acids in three simple steps: Load, Run and Obtain.

The MPure-12 extracts samples of any type or quantity. For tough samples, lyse first with our FastPrep-24 5G, a standalone tissue homogenizer. The FastPrep-24 5G is capable of lysing any sam-ple type in 40 seconds or less, while providing a high yield and purity of the analyte. The FastPrep-24 5G is available with a vari-ety of sample holders that allow versatility in processing sample tubes of 2 to 50 mL, or ambient or cryogenic samples. Difficult to lyse samples are easily processed by lysing with the FastPrep-24 5G followed by the extraction of nucleic acids with the MPure-12 System.

MPure Kits contain everything needed for sample purification: Pre-filled reagent car-tridges, reaction chambers, sample tubes, elution tubes, disposable pipette tips, etc.

MPure-12 pre-filled reagent cartridges are designed to increase analyte yield and data quality while decreasing contamination risk and cost. Re-agent cartridges are pre-measured and film-sealed to avoid contamination; the MPure-12 System automatically pierces this film before moving the re-agent to the sample tube, eliminating operator handling errors. Each car-tridge contains reagents for one sample and cartridges may be run individ-ually, allowing flexibility in the number of samples processed (from 1 to 12) with no unnecessary waste of reagents.

Integrity -

TAE agarose gel electrophore sis of DNA isolated in replicates of 12 from 200 μLhuman whole blood taken from two different donors.

Argarose Gel Analysis of PCR products. Electrophoresis image shows high consistency and reproducibility on extraction results.

The DNA was isolated in repli-cates of 12 from 200 μL human whole blood taken from two dif-ferent donors. Elution volume was set to 100 μL. Integrity of DNA was shown by subjecting each eluate to TAE agarose gel electrophoresis together with a suitable molecular weight marker. All samples exhibit sin-gle band with molecular weight > 40 kb with no smear.

Reproducibility -

β-globin gene was detected by real-time qPCR following isolation with MPure-12 blood DNA kit.

The DNA was isolated by MPure-12 blood DNA ex-traction kit from twenty whole blood samples. The β-globin gene was detected by real-time qPCR. This data shows ultra high stability and reproducibil-ity of MPure-12’s nucleic acid purification capability.

Stability -

Real-time qPCR detec tion of the β-globin (A) and spiked par vovirus B-19 DNA (A,B). Whole blood DNA isolated with MPure-12 kit.

Sensitivity -

Using whole blood (in EDTA collection tube) spiked with serial-diluted human Parvo B19 Virus (in range of 5-500000 IU/l). 10ml eluate was used for real-time PCR reaction by RealStar® Par vovirus B19 PCR kit 1.0.

Scalability -

The DNA yield shows excellent scalability in different volume (100, 200, 300 and 400 μL) of extraction and WBCs count of the samples.

The DNA was extracted from whole blood by MPure-12 Blood DNA extraction kit 200. Real-time qPCR detection of the β-globin (A) and spiked parvovirus B-19 DNA (A,B). No significant influences on samples with different anticoagu-lants and storage conditions.

The DNA was isolated from different whole blood sam ples (WBCs count range 1.8 - 22 x 103 cells/μL). DNA yield (measured by Nanodrop 2000) shows excellent scalability in different volumes (100, 200, 300 and 400 μL) of extraction and WBCs count of the samples.

Whole blood (in EDTA collection tube) was spiked with serial-dilution of human Par-vo B19 Virus (in the range of 5-500,000 IU/L). 200 µL samples were extracted and eluted in 100 µL volume. 10 µL of eluate was used for real-time PCR reaction by RealStar® Parvovirus B19 PCR kit 1.0. As little as 5 IU spiked (about 1 IU in PCR reaction) sample can be detected, proving the excellent sensitivity and linearity of isola-tion.
