3. Hybridization procedures and parameters
Each hybridization description should include information about which labelled extract
(related to which biological material, which extract) and which array (e.g., array design, batch
and serial number) has been used in the experiment; and the hybridization protocol, normally
including:
· the solution (e.g., concentration of solutes)
· blocking agent
· wash procedure
· quantity of labeled target used
· time, concentration, volume, temperature
· description of the hybridization instruments

4. Measurement data and specifications of data processing
We distinguish between three levels of data processing raw data (images),–
image quantitations and gene expression data matrix. Each hybridization has at
least one image, each image has a corresponding image quantitation table, where
a row represents an array design element and a column to a different quantitation
types, such as mean or median pixel intensity. Several quantitation tables can be
combined using data processing metrics to obtain the ‘final’ gene expression
measurement table associated with the experiment.
1) Raw data description should include for each scan laboratory protocol for
scanning, including scanning hardware and software, scan parameters, including
laser power, spatial resolution, pixel space, PMT voltage: scanned images;
It should be noted that MGED does not have consensus whether the provision
of images is a part of MIAME.
2) Image analysis and quantitation: image analysis software specification and
version, availability, and the description or identification of the algorithm and all
the parameters used for each image the complete image analysis output (of the
particular image analysis software)
3) Normalized and summarized data gene expression data matrix data–
processing protocol, including normalization algorithm

검체는 실험자의 리 취 법에 라 체 의 양이나 품질에 많 향- , RNA
미친다 검체 내에 존재하는 체 의 품질이 지 있도록 취 리를 철. RNA ,
히 하도록 한다.
일 로 체 는 단일가닥 로 이루어 있어 실 등에 불 함 나타- RNA
내 에 상 에 실 에 의 보 체 의 분해를 야 하므로 체RNA ,
의 품질 지하 해 이하에 검체를 보 하거나 경우에 라RNA -70 ,℃
의 해 가 포함 용액에 검체를 고 이하에 보 한다RNase -70 .℃
실험에 충분한 양의 체 를 얻 하여 충분한 양의 검체를 사용하여 체- RNA
를 추출하며 매우 양의 체 가 함 어 있는 검체인 경우 사용 시RNA , RNA
료의 양 늘 체 를 추출한다RNA .
검체는 체 가 추출 지 포 내외의 의 향 지 도록 동결- RNA RNase
등의 법 통한 취 보 이 요하다, .
탁 쉬운 포주 같 시료인 경우 페놀류 등의 포 용매를 이용하여 추출-
하며 조직과 같 고체 시료의 경우 액체질소 막자사 등 이용하여 검체를 충,
분히 갈 추출하며 이 경우에 막자사 의 차 염 특히 주의하고 시료는 마쇄,
과 에 항상 동결상태를 지하도록 한다.

검체에 분리한 체 의 품질 독 체 분 용 로 재 과 복 지- RNA
야 하 에 양질의 체 를 얻 해 는 추출 시에RNA RNase genomic
의 염 차단해야 하고 용매처리를 하여 다당류를 거하고 분 과 차후DNA ,
실험에 해가 있는 추출 용해시의 충용액이나 침 단계에 의 염 충분히/
