Practical PhD Course in Mouse Transgenesis Aarhus University October 26 to November 6 2015 Objectives: To introduce students to the principle techniques used to modify the murine genome such as transgenesis, knock out, knock in, TALEN- and CRISPR mutagenesis. The course includes hands on exercises and demonstrations of the following techniques: - Isolation of different stages of pre-implantation embryos - Culture of pre-implantation embryos - Pro-nuclear injection - Blastocyst injection - Preparation of embryonic fibroblasts - Derivation of ES cells from blastocysts - Culture, transfection and selection of embryonic stem cells - Isolation and analysis of single ES cell colonies - Cryopreservation of murine embryos and sperm The students will have the possibility to discuss their own projects with respect to the relevant techniques.
