PREFACE

Exposures to benzidine congener dyes in the workplace may increase the riskof cancer in the exposed workers. In particular, dyes based on the chemicalbenzidine may be carcinogenic; dyes based on o-tolidine or o-dianisidine mayalso pose a carcinogenic risk to workers. Cancer could be caused by the dyeitself, by leftover parent chemicals and impurities in the dye after itsmanufacture, or by chemicals resulting from breakdown of the dye in thebody. The risk of cancer extends beyond the manufacture of benzidinecongener dye where there is potential exposure to benzidine (a knowncarcinogen), o-tolidine, or o-dianisidine (suspected carcinogens). Workerswho use the dye or the dyed product may also be at risk and should be wellprotected.

The National Institute for Occupational Safety and Health (NIOSH) hasdeveloped this publication to alert supervisors, workers, safety committees,and safety directors to these hazards as well as to suggest the necessaryprotective measures. To "aid in determining whether or not benzidinecongener dyes are being used in a particular setting, Appendix A lists manyof their tradenames and synonyms, grouped by their parent chemical.

BACKGROUND

1. Those dyes tested showedcancer-causing potential.

In this publication. the termbenzidine congener dyes (BCDs)refers to commercial preparationsof those dyes produced from eitherbenzidine. ortho-to1idine. or ortho­dianisidine. The term "congener"(Latin for "same family") refers totheir similarities in. chemicalstructure.

Like many other technical grade dyeformulations. each commercial benz­idine congener dye may containseveral chemicals. At least fourof these chemicals--leftover parentcompound (the so-called "chemicalintermediate"). certain impurities.breakdown products. and the dyeitse1f--may pose serious healthhazards to workers.

Those BeDs tes ted were con­verted to the parent chemicalin the bodies of severalspecies of test animals.Available evidence supportsbiotransformation in humansof benzidine-based dyes tothe parent compound,benzidine. Although lessconclusive at this time,there is also evidence thathumans metabolize o-to1idine­based dyes to their parentchemicals.

The conclusionsscientists werelowing find ings:

Approximately 82 benzidine congenerdyes are now or have been widelyused in the u.s. They are marketedunder several hundred tradenames(many are listed in Appendix A ofthis publication). Although eachof these 82 dyes has not beenspecifically tested for healtheffects, they have severalimportant similarities. Thesesimilarities have led scientists atthe National Institute for Occupa­tional Safety and Health (NIOSH) toconclude. after extensive review ofthe data. that exposure to any ofthem in the workplace may .increasethe risk of cancer to exposedworkers.

Of the entire range of chemicalstested to date in the NationalCancer Institute's bioassay program,three benzidine-based dyes demon­strated the shortest "time-to­tumor" interval. These resultssuggested that the benzidine-baseddyes themselves might be very potentcarcinogens. Because the dyesthemselves--not simply leftover·intermediates, breakdown products,or contaminants--may be carcino­genic, it is not enough to protectagainst exposure to the dyes onlyduring dye manufacture. Workerswho apply the dye and workers whoprocess the dye are also exposed toa health hazard and must also bewell protected.

Dye house workers are probably themost significantly exposed workers.Much less is known about thepotential dye exposure of workersprocessing BCD-dyed fabrics,leather, and paper after thesematerials leave the dyeing plant.Generation of particulates from theBCD-dyed materials might occur, forexample, in cutting, sewing, knit­ting, weaving, and finishingoperations.

Although dye exposure in these. processing operations is not wellcharacterized, we do know that dyedparticulates are generated whenfabrics are prepared from dyedfibers, and that dyed particulatesare released during cutting andsewing operations.

Because the possibility of dyeexposure during processing activi­ties exists, but exposure data areat present not available, a deter­mination of whether such workersface any health hazard from workingwith BCD-dyed materials must awaitfurther investigation.

In summary, several benzidinecongener dyestuff constituent~-­

the dye itself, leftover parentcompound, breakdown products, andcertain impurities--may ,presentserious health hazards.

For the reasons discussed above,NIOSH recommends that workerexposure to benzidine-bas~d dyes bereduced to the lowest levelfeasibh, preferably by substitut­ing safer dyes for benz idine-baseddyes.

The available data on dyes based ono-tolidine and o-dianisidine, whileless extensive and definitivethan those for benz idine-baseddyes, suggest that these dyes, andcertain dye contaminants (includingbreakdown products occurring duringstorage or heating), may alsopresent a cancer risk to workers.Exposure to these two additionalclasses of dyes should therefore beminimized. NIOSH recommends, as aprudent public health measure, thatsubstitution of less toxic dyes beused as a control measure whereverpossible.

Q: Are only production workersexposed?

A: No. Production workers areoften the most routinely exposedworkers; but maintenance andcleanup crews can also be signif­icantly exposed. Such workersrisk exposure when local exhaustductwork requires cleaning, spillsmust be cleaned up, or processequipment must be disassembled.These individuals are of particu­lar concern because some of themost effective exposure controlsavaIlable to production staff, suchas process enclosure and localexhaust ventilation, are often theleast likely to be available tomarntenance workers.

The only sure way to find outwhether the ~ air in the worker I sbreathing zone is safe is to col­lect a sample of it, with apersonal sampler. The sampleshould then be analyzed by acertified industrial hygiene lab­oratory. Under the currentmedical and exposure records ac­cess standard, workers and theirrepresentatives have a legal rightto see the results of workplace airsampling tests performed by or fortheir employers.

Finally, some dye componentsmay enter -the body by absorptionthrough healthy or damaged skin, orthrough contact with the eyes.

A: Replacement of BCDs with knownsafe dyes is the most effectivemeans 0 f reduc ing exposure. Whensubstitution isn't a viable option,however, BCD exposure should bereduced to the lowest feasiblelevel.

A1though the amount. of exposurethat the body can safely tolerateis unknown, it is generally agreedtha t the smaller the exposure, theless the chance for developing anexposure-related disease. However,it is also possible that the chanceof the disease developing willremain the same, but that at lowerexposure levels it will take moretime for the disease to develop.

One very effective method forreducing inhalation exposureinvolves changing the form in whichthe dye is hand led. For example,dyes can be supplied in. paste,1 iquid, or pelletized forms, or insoluble bags. Note that handlingdyes in these forms does notnecessarily reduce the need foreffective protection against skinexposure (discussed below).

Inhalation (as well as skin expo­sure) can be further reduced, oftensubstantially, by preventing dyesfrom contaminating workplace airand work surfaces (floors, benches,machinery, etc.). A particularlyeffective control method involvescontaining those operations wheredye is transferred, weighed, mixed,metered, sampled, packaged, orotherwise handled. The technicalterm for this technique is processenclosure.

Localexhaust ventilation is frequentlyapplied to such enclosures to makesure that any openings permittingleakage will cause air to flow intothe enclosed area rather than Ql;tof it. Another example of processenclosure involves the use ofliquid metering devices or gravityfeeds for charging operations.By eliminating pouring and measur­ing steps, these· devices cansignificantly reduce exposure due.to spills.

Where process enclosure is notpractical, concentrations of air­borne dyes can still besubstantially reduced . by localexhaust devices, such as hoods anddowndraft or crossdraft tables.When properly designed, fabricated,and maintained, these devicescapture the dye particles close tothe point of release and preventthem from entering the worker'sbreathing zone. However, whenlocal exhaust is used, adequatefilters or traps should be providedin the exhaust air line before theair is released inside or outsidethe plant. It is also importantthat such filtration apparatus bedesigned to allow safe mainte­nance, and that the exhaust systemcomponents are kept well main­tained.

Engineering controls, such as pro­cess enclosure arid local exhaustventilation, can also substantiallyreduce dye exposure in dye applyingoperations, as well as in process­ing industry (post-application)operations where exposure is docu­mented.

restricted areas of the workplace.Such areas need to be clearlyposted with appropriate warningsigns, and access to them should belimited to workers informed of boththe potential hazards and propercontrol measures.

PERSONAL PROTECTIVE EQUIPMENT

Q: How can workers be protectedduring emergencies, maintenancework, equipment failures, andcleaning operations against expo­sure to hazardous dyes .that escapeand become airborne despite ef­forts to contain, capture, orremove them?

A: If hazardous dyes escape intoworkplace air, then (1) workersmust be individually protectedagainst breathing them or cominginto skin contact with them, and(2) settled dye particles must beremoved and immobilized to pre­vent them from becoming airborneagain.

Inhalation of hazardous dyes thatescape capture or containment ef­forts can be minimized by use of apositive-pressure respirator (airline or battery powered). Theseprovide air to the facepieceunder slight· pressure. The pres­sure (1) helps prevent airborne dyefrom leaking into the mask duringinhalation, and (2) reduces theburden of wearing a respirator bydiminishing breathing effort.

Persons wearing respirators otherthan supplied-air types typic~lly

breathe with greater effort andsweat more, and may be inadequatelyprotected due to a less than per­fect seal. A snug fit is essentialwhere the edges contact the face.

For situations where an air linerespirator would not allow suffi­cient mobility, a half-mask batterypowered air purifying respiratorwith high efficiency fil ters isrecommended. Powered air purifyingrespirators are inferior, however,to air line respirators for use inareas with substantial dust levels,or in confined spaces. Under emer­gency conditions, only respiratorswith a self-contained air supplyare suitable.

Note: Since the particles that canreach the lungs are light enough tostay suspended in workplace airlong after heavier particles havesettled, airflow patterns may carrythem to work positions far fromtheir source, where workers may notbe wearing respirators. Becausethese respirable particles are

small enough to be invisible.persons at nearby work stations maybe unaware of what they may bebreathing. Airflow patterns caneven move these microscopicparticles to more remote areas.such as lunch rooms. managementoffices. and other areas nottypically thought of as hazardous.

This illustrates one more reasonwhy engineering controls arestrongly preferred over respiratoruse for protection against BCDhazards; all employees can be pro­tected, n~ just the workers knownto be exposed.

Settled dye particles must be re­moved without scattering them backinto the air. This means no b10w­downs with compressed air. and nodry sweeping. The only safemethods for cleaning settled dyefrom equipment, ductwork, ledges,floor s. etc. are: (l) vacuuming(using high-efficiency filtervacuums. such as those approved forhandling asbestos), and (2) wetmethods (such as first sprayingcontaminated surfaces with a finewater mist and then hosing themdown with additional water).

Vacuuming is also recommended ·forthe initial removal of dust fromheavily contaminated clothing. bothimmediately after substantial con­tamination and routinely beforechanging clothes.

Q: What type of personal protec­tive equipment should be worn byworkers who cannot otherwise avoidskin contact with a benzidine con­gener dye?

A: Skin contact is typically mini­mized by wearing gloves andprotective coveralls. .

Gloves are especially important.since they prevent skin contactwith parent compounds and othercontaminants. The gloves must beimpermeable to the extent practi­cal. They must be washed beforebeing removed. and checked forpinholes. tears. and similar de­fects before reuse. Becausewearing gloves contaminated on theinside exposes the skin to thecontaminant, gloves with dye res­idue inside must be replaced orthoroughly cleaned. Longs1eevedstyle gloves are recommended toensure that wrists and forearmsaren't contaminated by materialentering the cuffs or getting onexposed arms.

Full-body coveralls are alsorecommended for protecting skinfrom contact with hazardous dyes.Coveralls should be changed atleast daily. and sooner if theybecome contaminated because ofleaks, spills, discharges, etc.Use of disposable outer garments isstrongly recommended for tworeasons. First, complete removalof BCDs from contaminated garmentscan be difficult. Reducingagents, such as those commonly usedin dye stripping processes, releasethe toxic parent compound from thedye. which may become entrapped onthe garments being laundered.

Second, if contaminated workclothes are worn home, or are takenhome for laundering, family membersmay be inadvertently exposed.

to clean dye-stained skin impairsthe skin's natural protectionagainst the environment. Onceimpaired. the skin more readilyabsorbs many hazardous chemicalsand becomes more susceptible toinfection and dermatitis. The useof any cleaner that contains astrong reducing agent. such assodium hydrosulfite. is especiallydangerous since this chemicaltransforms benzidine congener dyesback to their toxic parentcompounds. Showers should beavailable where dyestuff exposurecan occur. and workers should berequired to shower whenever sig­nificantly contaminated.

Q: How do workers get the re­quired gloves. coveralls. and(where appropriate) respirator?

A: Under Federal law employers areresponsible for protecting workersaga ins t workplace hazards, and arerequired to provide all necessaryprotective equipment. This equip­ment must be effective, properlymaintained, and readily availableto workers.

If reusable protective garments areused. the washing of coveralls.gloves. etc. should be performed orarranged for by the employer. sincelaundering of BCD-contaminated gar­ments necessitates special handling.Reducing agents should not be usedto remove BCD stains. due to con­vers ion of these dyes back to theparent compounds, and the possi­bility that these would be entrappedon the seemingly clean garment (oron other garments in the batchbeing washed).

The recommended alternative tolaundering is to provide a freshdisposable outer garment daily. and

to require it to be changed duringthe workday if it· becomes contami­nated. This avoids worker exposureduring laundering. and the possi­bility of residual contaminantsbeing present after washing.

Q: Is all footwear about equallyprotective against dye exposure?

A: No. Canvas sneakers and otherfabric footwear as well as leathershoes or boots will absorb spilleddye. especially when the dye is inliquid form or the footwear iswet. Dye "retained on absorbentfootwear may later contact theskin; the moist environment infootwear is conducive to skinabsorption of various dye compo­nents.

For these reasons, washable, non­absorbent footwear is recommended.Such footwear should be cleanedafter each shift and stored in aplace free from contamination. Analternative to using nonabsorbentfootwear is to use disposable shoecovers on existing footwear.

Q: Wearing full-body coveralls,nonabsorbent footwear, nonabsor­bent long-sleeved gloves, and (ifappropriate) a respirator couldmake working in hot work areasextremely ~uncomfortable. Arethere alternatives?

A: Yes. It is widely recognizedthat protecting workers individu­ally with personal protectiveequipment can impose considerablestress on them. In certainenvironments. such as high heatplus high humidity. this burden maybe serious enough to l.ncrease therisk of accidental l.nJuries, dueto lowered alertness or impaired

Becauseexposure is completely eliminated.substitut ion of a nonhazardous dyecan be the simplest and most effec­tive alternative. Where dyesubstitutes that are known to besafe are not available. processenclosure with local exhaust venti­lation to keep the enclosure undernegative pressure may provide ade­quate control.

Where work is performed under hotand/or humid conditions. and use ofpersonal protective equipment isunavoidable. the environment ~f thework area involved should be madeas favorable as possible. Forexample. ventilation can be in­creased to remove excess heat orhumidity. or air conditioning canbe installed. Frequent rest breaksmay provide additional protectionagainst severe discomfort.

Obviously many work areas could'onlybe air conditioned with great dif­ficulty; this is yet another reasonwhy engineering controls and othermeasures that control contaminantsat the source have been favored overuse of respirators. protective gar­ments. and other personal protectiveexposure controls.

A: Cases showed a range of 1-40years between first exposure andthe diagnosis of cancer in exposedworkers. I t is importan t to notethat the minimum amount or lengthof exposure required to initiatetumor develOpment is not known.

< BCD TRADENAMES AND GENERIC NAMES I BY PARENT CHEMICAL

Many different tradenames and generic names are used for benzidine congenerdyes. As an aid for identification, Appendix A lists tradenames and genericnames used for BeDs. Dye names are listed alphabetically and grouped bytheir parent chemical (benzidine, o-tolidine, or o-dianisidine). All threelists should be checked for the dye name in question. Both imported anddomestically produced dyes are listed.

Appendix A also lists many tradename dyestuffs that are no longer produced.These are included to (1) help workers find out whether they had beenexposed to BCDs in the past, and (2) permit recognition of older inventoriesof discontinued dyes that could still be used.

Colour Index (C. I.) numbers and generic names are included for those dyesidentified by the Toxicology Information Response Center, Oak Ridge NationalLaboratory as being based on benzidine, o-tolidine, or o-dianisidine. Theselists are not, however, exhaustive inventories of every BCD tradenameproduct. To ascertain whether a dye not listed is a benzidine congener dye,consult the Colour Index or contact NIOSH.

Metallized o-dianisidine-based dyes are noted by a "pound signll (#). Thesedyes, according to industry sources, may be less hazardous to handle thanother benzidine congener dyes. Since NIOSH does not now have enoughinformation to determine whether the commercial grade metallized dyespresent less of a health hazard, the "pound signll UF) should not beconstrued as NIOSH support for any safety claims.
