QUALITY CONTROL
NEST boasts of a high power laminar flow clean room meeting the ISO 8 grade with a floor space of 2,000 square meters. These facilities are equipped with a central feeding system, high precision injection machinery from Japan, robot arms with automatic conveyers, hot runner molds, and vacuum plasma and corona treatment machines from Belgium, Germany and USA. Inner room moisture content and temperature are adjustable. In 2013, NEST installed a Rhodotron TT 200 from Belgium IBA, a world leader in the E-Beam sterilization industry. NEST completes a full process chain from the sourcing of the raw materials to the in-house sterilization of the final products ready for delivery to clients. NEST’s manufacturing site has been certified with an ISO 9001:2008 management system. Our sterilization procedures meet the ISO 11137:2006 and ISO 13485 standards.

FEATURES
NEST cell culture plates have round edges for better handling. Low profile and low-evaporation lid
design assures uniform growth. Both tissue culture treated and non-treated surfaces are available.
Non-treated growth surface is more hydrophobic than tissue culture surface and is similar to the
surface of a bacteriological Petri dish. Small feet on the bottom corners of each plate eliminate
scraping of the microscope lens.

Guidelines For Use:
1. Mix cell suspension with medium: Prepare cell suspension of required concentration in a container. Recommended
volume is about 30- 50mL per layer.
2. Add the mixed liquid into the Multi-layer Flask slowly with serological pipettes. To avoid foams and bubbles, allow
liquid stream to flow along the slope of the Multi-layer Flask. (Save a little liquid in pipette each time.)
3. Tips: A 10mL pipette allows media to be dispensed at the bottom of the vessel. A 25mL pipette allows media to be
dispensed just past the NEST Logo.
4. Hold the Multi-layer Flask upright with the Logo facing you and tilt clockwise to a 45° angle on a flat work surface
to partition the liquid into each layer.
5. While holding the Multi-layer Flask at a 45° angle, gently lay it flat onto the work surface with logo facing up.
6. After placing the Multi-layer Flask flat on a work surface, gently rock back and forth and side-to-side to distribute
cells evenly onto culture surfaces.
7. Tips: Take care to avoid foaming of medium, and not to spill liquid from each layer.
8. Repeat Step 3 to put the flask quickly and slightly into the incubator . Then, lay it flat as shown in Step 4. You may
choose to either aspirate or pour the media from Multi-layer Flask.
9. Aspirating method: To aspirate or remove media, tilt Multi-layer Flask, with the NEST Logo facing you, counter-
clock wise to a 45° angle while inverting the Multi-Flask toward you. Then, tilt Multi-layer Flask to the right,
continuing to aspirate all residual media.
10. Pouring method: With Logo facing you, pour spent media from Multi-layer Flask
11. Tips: Aspirate media using a NEST 2mL or 10mL aspirating pipette.
12. Wash with buffer for one time and add dissociating reagent (≥5mL per layer). Then, follow Steps 3-4 to distribute
to dissociating reagent to each layer.
13. Neutralize with inactivating solution and mix following Steps 3-4. Gently swirl to dislodge cells completely.
14. Follow Step 7 “Aspirating Method” protocol and collect cell suspension using a NEST 10mL serological pipette.
15. Follow Step 8 “Pouring Method”. Pour the cell suspension into a NEST conical tube.
16. Rinse with additional wash buffer if needed.

Narrow Mouth+ Narrow Mouth BiofactoryTM
Twin narrow mouth with vent and seal cap is designed for aseptic operation to reduce contamination risk with the help of tubing

Guidelines For Use
A. Inoculating
1. Preheat the BiofactoryTM and media to culturing temperature before use which can accelerate cell adhesion and increase
cloning efficiency. Warming up is very important especially for BiofactoryTM over 10 chambers because It takes longer
time to reach the setting temperature for bigger size vessel
2. Pour the media directly into the BiofactoryTM or with tubing system. Recommend media volume for each chamber is
150~200mL
3. Keep the BiofactoryTM edge-on after filling the media. The media will be evenly dispensed to each chamber.
4. Turn the BiofactoryTM 90 ° anti-clockwise slowly and steadily with the mouth on upside to equilibrate the media
5. Lay down the BiofactoryTM
to level position slowly to equilibrate the media without generating bubbles.
6. Put the BiofactoryTM into an incubator
B. Cell Harvest
1. Pour out the media when the cultivation is finished
2. Wash with CMF-PBS(40-50mL per chamber) at least once. Follow the operating steps
to ensure the culturing surface soak in the CMF-PBS completely. Gently shake back and
forth to wash away the residue media. Pour out the CMF-PBS and repeat the steps if
necessary.
3. Add appropriate preheated cell digestive solution (20-40 ml/layer), follow the
BiofactoryTM operating guideline to equilibrate the digestive solution. Tilt the BiofactoryTM
back and force gently to ensure the culturing surface soak in the dissociating solution.
Flap the BiofactoryTM slightly to help the cell detach from the surface. Dissociation
time could be referred with that of parallel condition in T Flasks or single chamber
BiofactoryTM
4. Wash the BiofactoryTM after dissociation, repeat the wash steps if many cell are found on
the wash solution
Preheat:
Preheat the CMF-PBS can reduce the wash time while preheat the media can accelerate the cell
attachment
Vent cap:
Keep the vent cap from alcohol which may wet the filter film, further affect the gas exchange and
liquid equilibrium
Culture temperature:
Slight variation of temperature will affect the cell yield. Pay attention to the culture temperature is
in accordance with the setting temperature.

Tips Coating
