Resumo
• Concentrações oscilantes de LH e de FSH estão sob controle do GnRH hipotalâmico pulsátil
• Variações no padrão de pulso do GnRH associam-se a secreções divergentes de LH e de FSH
• Alterações na regulação gonadotrófica normal pelo GnRH associam-se a vários distúrbios clínicos
Os gonadotrofos hipofisários
• 7-15% das células da adenohipófise
• Grânulos mono e bi-hormonais
• As gonadotrofinas são compostas por 2 sub unidades
– α (homóloga)
– β (distinta para cada gonadotrofina)
• A sub-unidade β confere atividade hormonal específica para o heterodímero

Resumo:
• GnRH liga-se ao receptor GnRHR na superfície celular dos gonadotrofos e tem o potencial de estimular diversas cascatas de sinalização
• Mutações do GnRHR afetando “trafficking” são a base de muitas doenças
• Chaperones farmacológicos são um tratamento potencial para distúrbios do “trafficking”
• Síntese diferencial do genes da sub-unidade β das gonadotrofinas está associado a modificações de ativação / estabilidade de importantes proteínas regulatórias e fatores de transcrição

• Sítio de ação
• Papeis relativos de T & E2
• Determinar a contribição relativa dos esteroides sexuais vs inibina B para a regulação da secreção de FSH
