Abstract
The present dissertation thesis deals with several research topics involving development and
use of cyclic hypervalent fluoroalkyl iodine(III)-reagents in electrophilic fluoroalkylation
reactions of a variety of nucleophiles.
The second chapter describes efforts in improving the current syntheses of the incresingly
popular electrophilic trifluoromethylation reagents on the basis of hypervalent iodine(III)
deveIoped in 2006 by Patrick Eisenberger. Simplified syntheses suited for safer large scale
preparation of these two hypervalent CF3-iodine reagents R4 and R5 are reported.

In both cases, the stoichiometric oxidants sodium metaperiodate and t-butyl hypochlorite
have been replaced by cheaper and easier to handle trichloroisocyanuric acid. Reagent R4
was accessed in a one-pot procedure from 2-iodobenzoic acid in 72% yield. Reagent R5 was
prepared via its fluoroiodane in a considerably shorter time and with no need of a precisely
defined temperature regime. Both syntheses were optimized for use on larger scale.
In the third chapter, α-trifluoromethylation of enolizable carbonyl compounds is described. In
its first part, diastereoselective trifluoromethylation of Evans-type acyloxazolidinones is
reported, a project conducted in collaboration with Dr. Dominique Cahard from INSA, CNRS,
Rouen, France. Acyl-oxazolidinones were deprotonated with LiHMDS and reacted with R4 to
give α-trifluoromethylated products in up to 91% combined isolated yields and 97:3 dr.

Use of sodium and potassium hexamethyldisilazide instead of LiHMDS led to inferior
reactivity and diastereoselectivities, giving an indication that the Lewis acidity and higher
oxophilicity of the Li cation might have been responsible for both reagent activation and
tighter chelation of reactive intermediates.The resulting isolated diastereopure products could
be further transformed into valuable products without racemization.
In the second part, a continuation of the project initiated by Dr. Raffael Koller was carried out.
A range of five- and six- membered lactones were transformed via their lithium enolates to
their trimethylsilyl ketene acetals and these were reacted in the presence of 1 mol %
TMSNTf2, a powerful silylating reagent, to the corresponding tertiary or quaternary α-
trifluoromethylated lactones in up to 96% isolated yield. The reaction was shown to be
particularly insensitive to steric bulk in the α-position. Aryl-substituted lactones generally
gave worse results, most likely as a result of diminished electron density of the
corresponding silyl ketene acetals.

In the fourth part of the third chapter, 3-substituted N-Boc-oxindoles were trifluoromethylated
with R4 or R5.

Upon lithiation with LDA, these substrates underwent trifluoromethylation with R4 in up to
70% NMR yield. Alternatively, these substrates could be also trifluoromethylated with R5 in
the presence of 10 mol % MgBr2-diethyl etherate in up to 76% NMR yield, giving access to
quaternary trifluoromethylated oxindoles. We speculated that Mg2+ salts play a dual catalytic
role - firstly, bidentate oxygen chelation of Boc-3-alkyl-oxindoles with Mg2+ cation leading to
enolization of the substrate and secondly, activation of R5 through coordination of its
alcoxide moiety. The reactions was shown to be relatively sensitive to the steric bulk of the R
group.
In the fourth chapter, a protocol for mild trifluoromethylation of N,N-disubstituted
hydroxylamines with R4 and R5 is described. The trifluoromethylation method was shown to
be tolerant towards a variety of functional groups, including nitriles, alcohols, ketones, esters,
amides, imides and nitrogen heterocycles.

The key step in achieving a clean reaction outcome is the activation of the CF3-reagent with
either TMSOTf or LiClO4 and partial or full deprotonation of the substrate with
tetramethylguanidine or LDA. Preliminary mechanistic studies suggest that the reaction
follows a radical pathway in which the deprotonated hydroxylamine and a Lewis or Brønsted
acid-activated CF3 reagent engage in a single electron transfer step generating a pair of
radicals which recombine to afford the desired product. The trifluoromethylation procedure
was successfully applied in the modification of secondary nitrogen groups of
pharmaceutically relevant targets (Fluoxetine and Mefloquine), affording new derivatives
containing a novel N-trifluoromethoxy moiety.

The fifth chapter describes the development and use of novel cyclic fluoroalkyl hypervalent
iodine reagents featuring different -CF2CF2R moieties, a project conducted in collaboration
with Dr. Petr Beier, Institute of Organic Chemistry and Biochemistry, Academy of Science of
Czech Republic, Prague. Using BrCF2CF2Br as the -CF2CF2-synthon source, the reported
compounds PhSCF2CF2SiMe3, PhSO2CF2CF2SiMe3, PhOCF2CF2SiMe3 and ImCF2CF2SiMe3
were synthetized. Using these fluoroalkylated silanes in the rapid fluoroiodane-based
Umpolung protocol whose basis was developed in the second chapter, novel hitherto
unknown reagents were synthetized in isolated yields up to 85%. The reactivity of these
reagents was shown to roughly parallel the known reactivity of R4 and R5. Using slightly
modified procedures developed for trifluoromethylation with R4 and R5, thiols,
hydroxylamines and phosphines were fluoroalkylated in good to excellent yields.

In the last chapter, copper-catalyzed trifluoromethylation of terminal acetylenes is described.
Screening of optimal reaction conditions identified the combination 1 mol% CuCl, 1.2 mol%
1,10-phenantroline, 10 mol% benzyltriethylammonium chloride and 1 equiv R5 in MeCN to
give best results in trifluoromethylation of phenylacetylene in 70% NMR yield.

Zusammenfassung der Dissertationsarbeit
Die vorliegende Dissertationsarbeit behandelt mehrere Forschungsthemen in Bezug auf die
Entwicklung und Anwendung von hypervalenten Fluoroalkyl Iod(III)-Reagenzien in
elektrophilen Fluoroalkylierungsreaktionen mit einer Reihe von verschiedenen Nukleophilen.
Das zweite Kapitel beschreibt die Verbesserung der bisherigen Synthesen der immer
populärer werdenden elektrophilen Trifluoromethylierungsreagenzien auf Basis vom
hypervalenten Iod(III), ursprünglich entwickelt in 2006 von Dr. Patrick Eisenberger.

In beiden Fällen wurden die stöchiometrischen Oxidationsmitteln Natrium Metaperiodat und
t-Butyl Hypochlorit durch die wesentlich billigere und einfacher zu handhabende
Trichlorisocyanursäure ersetzt. Reagenz R4 wurde im Eintopfverfahren ausgehend von 2-
Iodobenzoesäure in 72% Ausbeute synthetisiert. Reagenz R5 wurde über sein Fluoroiodan
in erheblich kürzerer Zeit und ohne Notwendigkeit eines rigoros definierten
Temperaturgradienten dargestellt. Beide Synthesen wurden für den Einsatz im grösseren
Massstab optimiert.
Im zweiten Kapitel wird die α-Trifluoromethylierung von enolisierbaren Carbonylverbindungen
behandelt. In seinem ersten Unterteil, wird die diastereoselektive Trifluoromethylierung von
Evansschen Acyloxazolidinonen wird beschrieben, ein Projekt, das in Zusammenarbeit mit
Dr. Dominique Cahard aus INSA, CNRS, Rouen, Frankreich, durchgeführt wurde.
Acyloxazolidinone wurden mit LiHMDS deprotoniert und mit R4 unter Bildung von α-
trifluoromethylierten Produkten in kombinierten isolierten Ausbeuten bis zu 91% und
Diastereomerenverhältnissen bis 97:3 umgesetzt.

Die Verwendung von Natrium und Kalium Hexamethyldisilazid statt LiHMDS führte zu
schlechterer Reaktivität und Diastereoselektivität, ein möglicher Hinweis dafür, dass die
Lewis Acidität und Oxophilie des Lithium Kations einen Beitrag zu Reagenzaktivierung und
stabilerer Chelatisierung reaktiver Intermediate geleistet hat. Die resultierenden isolierten
diastereomerenreinen Produkte konnten weiter zu wertvollen Produkten ohne Racemisierung
ungesetzt werden.
Im zweiten Unterteil wird eine Fortsetzung des von Dr. R. Koller initiierten Projekten
beschrieben. Eine Reihe von fünf- und sechgliedrigen Laktonen wurde über ihre Li-Enolate
zu den Silylketenacetalen umgewandelt, und diese wurden anschliessend mit R5 in
Gegenwart von 1 mol% TMSNTf2, eines starken Silylierungsmittels, zu den entsprechenden
tertiären und quaternären trifluoromethylierten Laktonen in bis zu 96% isolierter Ausbeute
umgesetzt. Die Reaktion erwies sich als unempfindlich gegenüber der sterischen Hinderung
in α-Position. Arylsubstituierte Laktone gaben generell schlechtere Resulate,
meistwahrscheinlich als Konsequenz der verminderten Elektronendichte des
entsprechenden Silylketeneacetals.

Im dritten Unterteil, 3-substituierten N-Boc-Oxindole wurden mit R4 und R5
trifluoromethyliert.

Nach Lithiierung mit LDA konnten diese Substrate mit R4 mit bis zu 70% NMR-Ausbeute
trifluoromethyliert werden. Alternativ konnten sie mit R5 in Gegenwart von 10 mol% MgBr2-
Diethyletherat in bis zu 76% NMR-Ausbeuten trifluoromethyliert werden, wobei quaternäre
trifluoromethylierte Oxindole isoliert werden konnten. Wir vermuten, dass Mg2+ Salze eine
doppelte Rolle spielen. Erstens führt bidentate Chelatisierung mit dem Mg2+ Kation zum
höheren Grad der Enolisierung des Substrats und zweitens zur Aktivierung von R5 durch
Koordinierung seiner Alkoxid-Gruppe.
Im vierten Kapitel wird ein Protokoll für die milde Trifluoromethylierung von N,N-
disubstituierten Hydroxylaminen mit R4 und R5 beschrieben. Die
Trifluoromethylierungsmethode hat sich durch eine gute Toleranz gegenüber einer Palette
verschiedener funktionellen Gruppen ausgezeichnet, inklusive Nitrile, Alkohole, Ketone,
Ester, Amide, Imide und Stickstoffheterozyklen.

Der Schlüsselschritt in der Optimierung der Reaktion, sodass sie einen sauberen Ablauf
aufweist, war die simultane Aktivierung vom CF3-Reagenz mit entweder TMSOTf oder
LiClO4, und parzielle oder vollständige Deprotonierung des Substrats mit
Tetramethylguanidin oder LDA. Vorläufige mechanistische Experimente deuten darauf hin,
das die Reaktion auf einem radikalischem Wege abläuft, in dem das deprotonierte
Hydroxylaminsubstrat und Lewis oder Brønsted-Säure aktiviertes CF3-Reagenz in einem Ein-
Elektron-Übertragungsschritt ein Radikalpaar bilden, das unter Radikalrekombinierung das
gewünschte Produkt liefert. Die Trifluoromethylierungsprozedur wurde erfolgreich für die
Modifikation der sekundären Amingruppe der pharmazeutisch relevanten Moleküle Fluoxetin
und Mefloquin eingesetzt, und somit konnte man Derivate mit neuartiger N-Trifluoromethoxy
Gruppe erhalten.

Das fünfte Kapitel beschreibt die Entwicklung und Verwendung neuartiger zyklischen
fluoroalkylsubstituierten hypervalenten Iod (III)-Reagenzien mit unterschiedlichen -CF2CF2R
Gruppierungen, ein Projekt dass in Zusammenarbeit mit Dr. Petr Beier vom Institut der
Organischen Chemie und Biochemie der Akademie der Wissenschaften der Tschechischen
Republik in Prag, enstand. Unter Verwendung von BrCF2CF2Br als -CF2CF2- Synthonquelle,
wurden die bekannten Verbindungen PhSCF2CF2SiMe3, PhSO2CF2CF2SiMe3 und
ImCF2CF2SiMe3 wurden hergestellt. Diese fluoroalkylierten Silane konnten in der im zweiten
Kapitel ausgearbeiteten Fluoroiodan basierten Umpolung zu den entsprechenden bis jetzt
unbekannten hypervalenten Iod(III)-Reagenzien mit isolierten Ausbeuten bis zu 85%
umgesetzt werden. Im Allgemeinen liess sich beobachten, dass die Reaktivität dieser
Reagenzien grob der von R4 und R5 ähnelt. Unter Verwendung leicht modifizierter
Prozeduren, die ursprünglich für R4 und R5 entwickelt wurden, konnten Thiole,
Hydroxylamine und Phosphine in guten bis exzellenten Ausbeuten fluoroalkyliert werden.

Im letzten Kapitel wird die Kupfer-katalysierte Trifluoromethylierung von terminalen
Acetylenen beschrieben. Anhand vom Screening der optimalen Reaktionsbedingungen
wurde festgestelt, dass die Kombination von 1 mol % CuCl, 1.2 mol% 1,10-Phenantrolin, 10
mol% Benzyltriethylammonium Chlorid und 1 Äq R5 in MeCN die Trifluoromethylierung von
Phenylacetylen in bis zu 70% NMR Ausbeute ermöglichte.
