One hundred micrograms of proteins, present in rat liver homogenates, subcellular fractions, and purified peroxisomes, was subjected to SDS – PAGE, transferred to nitrocellulose, and analyzed for the presence of PTS1- proteins by using a bacterial lysate containing a vector encoding the biotinylated PTS1-receptor (A, C, and D) or a vector encoding only the biotinylated fusion part (B) as described under Materials and Methods. In C and D, the bacterial lysate was combined with 60 mM SKL- peptide and 60 mM UNSKL-peptide as competitor, respectively. The blue bands are endogenous biotinylated proteins, and the red bands are PTS1-proteins. (H) homogenate, (E) postnuclear supernatant, (N) nuclear fraction, (M) heavy mitochondrial fraction, (L) light mitochon- drial fraction, (P) microsomal fraction, (S) cytosolic fraction, (PO) purified peroxisomes, and (PO*) purified peroxisomes of a clofibrate-treated rat. PTS1-containing proteins are indicated by arrows at the right side of D (ACOX-A, the 70-kDa subunit of palmitoyl-CoA oxidase; ACOX- C, the 23-kDa subunit of palmitoyl-CoA oxidase; AGT, L-alanine:glyoxylate aminotransferase 1; MFP, the inducible multifunctional protein; SCPx, the 58-kDa sterol carrier protein; UOX, urate oxidase). Proteins recognized by the anti-SKL antibodies are marked by an asterisk.
