METODA PERUNUT 1-131 DAN PENGAKTIFAN NEUTRONDALAM EVALUASI HASIL IODINASI MINYAK CANDU

ABSTRAK
Minyak candu didapat dari biji tanaman varietas Pap
aver Somniferum dari famili Papaveraceae. Digolongkan
sebagai minyak pengering karena memiliki Bilangan Iod
lebih besar dari 130 atau kandungan asam linoleat diatas
60%. Beberapa publikasi melaporkan bahwa minyak candu
terdiri dari 11 sampai 30% asam oleat, 62 sampai 73% asam
linoleat dan 7 - 15% asam lemak jenuh yaitu 4 - 11 % asam
palmitat dan 2 - 4% asam stearat. Minyak candu yang
digunakan dalam penelitian ini memiliki sifat fisika dan
kimia sama dengan yang dimaksud dalam literatur karena
memiliki data-data karakteristik sama.
Dalam penelitian ini dilakukan hidrolisa minyak
candu dengan cara refluks dalam suasana basa kuat dan
pelarut alkohol. Hasilnya adalah 86 - 89 % berat asam-asam
lemak dengan rendemen 90-93% Asam-asam lemak
tersebut diesterifikasi dengan etanol menggunakan metoda azeotrop
Benoena - Alkohol dan katalis asam sulfat pekat, dihasilkan
etilester sekitar 87 - 91 %. Perkiraan B.M rata-rata minyak, asam-
asam lemak atau etilester dapat diketahui dari besarnya Bilangan
Penyabunan zat-oat tersebut masing-masing ialah 189, 197,172 dan B
M rata-rata didapat masing-masing 887,284 dan 326. Sementara itu
berdasarkan perhitungan adalah 875 -887, 284 dan 312. Dari Bilangan
Iod didapat informasi tentang banyaknya ikatan rangkap yang terdapat

dalam asam-asam lemak gliserida minyak candu. Dari
percobaan didapat bahwa Bilangan Iod minyak candu, asam-
asam lemak dan etilesternya adalah 135, 139 dan 122.
Matra dapat diperkirakan bahwa satu mol minyak dapat
menyerap rata-rata 5 mol Iod, masing-masing 3 mol asam
dan ester menyerap rata-rata 5 mol Iod.
Terhadap etilester dari asam-asam lemak minyak
candu dilakukan Iodinasi dengan menggunakan metoda Wijs.
Pereaksi aktif Iodomonoklorida dibuat dengan cares
mengalirkan gas klor ke dalam larutan Iodium dalam asam
asetat glasial, hasilnya dianalisa dengan metoda
Pengaktifan Neutron dan didapat perbandingan mol Iod dan
klor adalah 1 : 1. Dengan metoda tersebut diketahui Pula
bahwa Iodinasi berlangsung sekitar 70 sampai 100 %
dihasilkan Iodokloroetilester meskipun klor selalu
terikat lebih banyak dari Pada Iod. Untuk mendapatkan
Iodoetilester sebagai hasil akhir, makes klor ditukar
oleh Iod dengan menggunakan larutan Iodida dalam asam
asetat 98 % serta dibubuhi sejumlah natrium borohidrida
untuk mencegah oksidasi Iodida. Hasil pertukaran
dianalisa dengan metoda Pengaktifan Neutron dan
ditelusuri Pula dengan metoda Perunut I-131. Dari kedua
metoda didapat rendemen reaksi pertukaran yang kira-kira
sama yaitu sekitar 60 %. Reaksi Iodinasi keseluruhan
terjadi sekitar 70 - 80 % dihasilkan Iodoetilester.
Identifikasi hasil reaksi dengan metoda
Kromatografi kertas didapat perbedaan harga Rf antara
minyak candu, asam-asam lemak dan etilesternya yaitu

0,07;0,98 dan 0, 42 pada campuran eluen air: etanol : eter
sama dengan 4 : 2 : 3. Dalam eluen tersebut kelarutan
Iodokloroetilester dan hasil pertukarannya terlalu besar.
Kemudian dalam komposisi eluen air:etanol:titer sama
dengan 5:2:1,Iodokloroetileter dan hasil pertukarannya
kira-kira mempunyai Rf sama yaitu sekitar 0.9-0,95..
Dengan metoda spektrofotometri ultraviolet didapat
spektrum minyak candu, asam-asam lemak, etilester dan
Iodokloroetilester dengan A maksimum masing - masing
pada 289 ; 241 , 241 - 245 252 dan 263 241 nm. Hasil
pertukarannya memiliki spektrum dengan h maksimum 263 nm
dan sedikit serapan terlihat pada 241 nm. Digunakan asam
Iodoasetat dan asam kloroasetat sebagai pembanding dan
didapat spektrum dengan X maksimum
masing-masing 263 192 dan 192 nm. Berdasarkan literatur X
maksimum 192 nm berasal dari gugus asam asetat, maka 263
nm berasal dari gugus Iod.
Dari kedua cara identifikasi tersebut diatas dapat
disimpulkan bahwa Iodokloroetilester dengan hasil
pertukarannya masih memiliki sifat relatif sama, atau
belum seluruh klor dapat ditukar oleh Iod. Sesuai dengan
reaksi Iodinasi baru dicapai sekitar 70 - 80 % dihasilkan
Iodoetilester.

ABSTRACT
Poppy seed oil was obtained from the seed of
Papaver Somniverum of Papaveraceae family It was called
a drying oil because the Iodine Number was more than 130
or linoleic acids content was above 60 %. Many authors
reported that the poppy seed oil consists of 11 to 30 %
oleic acids, 62 to 73 % of linoleic acids and 7 to 15 %
saturated acids. The poppy seed oil used in this
experiment showed similar physical and chemical
properties with the reported poppy seed oil.
Prior to Iodination the oil was hydrolized then
esterified. The hydrolysis was carried out by refluxing
with alcoholic potassium hydroxide solution, which give
a yield of 86 - 89 % of free fatty acids or 90 -93 % of
the theoretical value. The ethylesters were obtained by
refluxing of the free fatty acids with a micture of
ethanol, benzene and concentrated sulphuric acid. The
yield was 87 - 91 % of the theoretical value. The
Saponification Number of the oil, fatty acids and
ethylesters were 189, 197, and 172 respectively,
corresponding to the average Molecular Weight of 887,
284 and 326, while the theoretical value were 875 - 887,
284 and 312. The Iodine Number of the oil, fatty acids
and ethylesters respectively were 135, 139 and 122. It
was found that one mol of the triglycerides absorbed
about five mols of halogen and three mols of fatty acids
or ethylesters absorbed five mols of halogen.

The ethylesters was Iodinated by the Wijs method.
Iodomonochloride was prepared by addition of chlorine
into a solution of Iodine in glacial acetic acid. Neutron
Activation Analysis confirmed that the reagent consist of
Iodomonochloride although the Iodination product
captivated more chlorine than Iodine atoms. The yield of
Iodination was 70 - 100 %. The chlorine atom was
eliminated from Iodoehloroethylesters by exchange with
Iodide solution in 98 % acetic acid. Sodium borohydrine
was added to the reaction micture, to stabilize the
Iodide ions. Neutron Activation Analysis and I-131
tracers were used to confirm the reaction. It was found
that 60 % of chlorine in Iodoehloroethylesters was
replaced by Iodine.
Paper chromatographic analysis showed Rf values as
follows : the oil 0,07 fatty acids 0,98 and the
ethylesters 0,42 . As developing solvent was used a
micture of H2O, ethanol and ether in the proportions of 4
: 2 : 3. The haloesters and the exchange product were
not separated. Using a solvent compositions of 5 : 2 : 1,
similar R f values were obtained, i.e. 0,9 and 0,9-0,95.
The ultraviolet spectra of the 'ethylester and fatty
acids showed the absorption peaks at 252 and 241 nm
respectively. While that of the oil and haloesters was at
241 and 289, 241 and 263 nm respectively. A similar
absorption spectrum was showed by the exchange product,
but the peak at 241 nm was appreciably lower. The peak

at 263 nm of Iodoacetic acid was due to absorption by
Iodine atoms. Both identification methods showed that
the haloesters still have similar characteristics with
the exchange product, because exchange of the chlorine
atoms was not complete.
