INTRODUCTION
CHAPERONES
Was discovered like heat
shock protein.
Its synthesis increases
with the high temperature.
Required for appropriate
unfolding, transport,
folding and degradation of
proteins.
Prevent protein
aggregation before folding
ending.
Increases the folding
velocity.
APOPTOSIS
Programmed cell
death.
Normal, clean and
tidy process
Removed cells with
genetic damage,
infected or old.
Can be activated for
internal or external
stimulation.
CORNEA
Transparent front part of
the eye.
Covers the iris, pupil,
and anterior chamber.
Functions: protection,
transmission and refraction.
Has 5 layers:
• Corneal epithelium
• Bowman's layer
• Corneal stroma: 90%
• Descemet's membrane
• Corneal endothelium
HSP27
Is a chaperone of the sHsp group
ATP-independent chaperone
Functions:
• Chaperone activity
• Thermotolerance
• Inhibition of apoptosis
• Regulation of cell development
• Cell differentiation
• Implicated in architecture of the cytoskeleton
• Cell migration
• Cell survival
• Tumor progression.
INTEGRATION
HSP27 is a chaperone who protects in a
stress situation, inhibiting the apoptosis
and activating the cell migration in human
corneal epithelial cells
GENERAL OBJECTIVE
GENERAL OBJECTIVE
Found the exactly role of HSP27
phosphorylated for the wound healing on
human corneal epithelial cells by certain
series of molecular methods that
demonstrate specificity for the apoptosis
and migration process

MATERIALES Y
MÉTODOS
LINEA CELULAR:• Es un cultivo celular
transferido o subcultivado
•Uniforme y homogéneo
•Podrá ser propagado,
transferido y almacenado
•De fácil manejo
En este estudio se usó
una línea celular de
células epiteliales de la
córnea humana
MATERIALES Y
MÉTODOS
WESTERN BLOT
•También llamado Inmunoblot
•Usada para
detectar proteínas específicas
en una muestra con la
interacción antígeno-anticuerpo
Pasos
1. Extracción y
solubilización de las
proteínas
2. Electroforesis en gel
(SDS-PAGE)
3. transferencia a
una membrana
absorbente
4. Detección de las
proteínas de la membrana
mediante el uso de anti-
cuerpos específicos
MATERIALES Y
MÉTODOS
WESTERN BLOT
Estudiar la presencia de
la proteína en el extracto
y analizar su cantidad
relativa respecto a otras
proteínas.
Para la visualización de
las bandas de interacción
antígeno-anticuerpo se
usó reactivos de
quimioluminiscencia
En el estudio se usó para
medir la expresión de
HSP27 después de le
lesión en las células
epiteliales de la córnea
humana
MATERIALES Y
MÉTODOS
TRANSFECCIÓN
Introducción de ADN
foráneo dentro del núcleo
de una célula eucariota
mediante plásmidos,
vectores víricos u otros
Se lleva a cabo al abrir
poros transitorios en la
membrana plasmática
para permitir paso de
material genético
En el estudio se usó para
introducir siRNAs-HSP27
específica y siRNAs de
control en las células
epiteliales de la córnea
humana, mediante
lipofección
MATERIALES Y
MÉTODOS
WESTERN BLOT DE TRASFECCIÓN
Este método se usó para
determinar el papel de
HSP27 en la
sobrevivencia celular.
MATERIALES Y
MÉTODOS
MIGRACIÓN CELULAR
Mecanismo por el cual la
célula se moviliza en
forma activa en
condiciones in vivo o in
vitro.
Utiliza como soporte
componentes de la
matriz extra celular, otras
células o sustratos no
biológicos.
Se da en la formación de
tejidos en el desarrollo
embrionario,
cicatrización y
la respuesta inmune.
En el estudio se midió
mediante fotografías en
puntos específicos para
cuantificar el número de
células en la migración de
las áreas lesionadas de las
células del epitelio cornea.
MATERIALES Y
MÉTODOS
MICROSCOPÍA CONFOCALTécnica de imagen
utilizada para incrementar
el contraste de las
fotografías, para
reconstruir imágenes en
tercera dimensión con una
resolución mayor.
En el estudio se usó
para analizar los
filamentos de actina y
su citoesqueleto.
MATERIALES Y
MÉTODOS
CITOMETRÍA DE FLUJO
Tecnología compleja, de
gran especificidad, que
permite analizar una
población celular.
Proporcionando gran
variedad de parámetros
como el recuento y
clasificación de células
según sus características
morfológicas
En el estudió se utilizó
este método para evaluar
la apoptosis celular.
MATERIALES Y
MÉTODOS
ENSAYO DE PROLIFERACIÓN CELULAR
La proliferación celular se
da en numerosos
procesos fisiológicos y
patológicos como el
crecimiento, la
cicatrización, la hipertrofia,
el desarrollo de tumores
La células en proceso de
proliferación pasan por
ciclo celular, durante el
cual las células se
replican y se dividen en
dos células hijas.
En el estudio de la
proliferación de las
células se midió mediante
densidad óptica, entre
mayor densidad, más
proliferación se había
dado.

CONCLUSIONS
CONCLUSIONS
• HSP27 phosphorylated was increased
after scratch wounding, so this protein is
related with the first steps of epithelial
wound healing.
• It’s very important to improve every day in
the way to induce apoptosis, because it's a
form to fight with cancer.
CONCLUSIONS
• With this discovering, scientists can think in new ways to treat corneal problems and it can revolutionize ophthalmology.
• We can elucidate the importance of molecular biology in medicine, with this research we have an idea of the behavior of proteins involved in specific cellular functions.
