?Membrane Protein Purification Kit Membrane Protein Purification Kit is designed for efficient detergent screening of histidine-tagged membrane proteins. Small aliquots of cell membranes are solubilized in different detergents, followed by rapid purification using His Mag Sepharose Ni magnetic beads. Reliable recovery of captured protein is achieved using a magnet.

The purification step is performed directly after solubilization using the same detergent. Analysis and evaluation can be performed by a number of methods such as Western blotting, gel filtration, or light scattering.

Membrane Protein Purification Kit offers the following benefits:

Mag Sepharose beads are magnetic beads designed for simplified enrichment of target proteins by immunoprecipitation, pull-down applications, or enrichment of phosphopeptides using titanium dioxide chromatography. Captured protein and peptide fragment targets are immobilized to ligands attached to Mag Sepharose beads and collection of beads and captured protein is achieved using a magnet.

Two new products have been added to the range of Mag Sepharose beads:

Protein A Mag sepharose Xtra and Protein G Mag sepharose Xtra are designed for efficient, high capacity small-scale purification/screening of monoclonal and polyclonal antibodies from various species. Antibodies are captured with high specificity on protein A and protein G ligands attached to Mag Sepharose. Protein A and Protein G Mag Sepharose beads allow simple capture of antibodies from low microliter to high milliliter scales. Purification of more than 350 µg of human IgG per purification run is possible.

His Mag sepharose ni is designed for efficient, small-scale purification/screening of histidine-tagged proteins. Histidine-tagged proteins are captured on the Ni2+ ligand coupled to Mag Sepharose magnetic beads. Purification of up to 500 µg of histidine-tagged protein per purification is possible.

The high density of the beads allows rapid capture with magnetic devices while the visibility of the beads ensures reliable collection of the target protein. In addition, the beads are hydrophilic and nonadherent and this reduces smearing effects and aggregate formation. Together with MagRack 6, a separation tool for handling the beads in microcentrifuge tubes, up to six samples can be processed in parallel. Large numbers of samples can be screened in parallel and throughput increased even further with the use of a robotic instrument.

This handbook describes the methodology for using gradients of Ficoll-Paque™ media for density centrifugation of human mononuclear cells. Methodology and applications of the sterile, density gradient media Ficoll-Paque PLUS and Ficoll-Paque PREMIUM are described in detail. These media of 1.077 g/ml density are supplied ready-to-use for the purification of mononuclear cells from human peripheral blood.

This update further introduces the use of Ficoll-Paque PREMIUM 1.073 and 1.084, which are centrifugation media for the isolation of lower- density cells such as monocytes and higher-density mononuclear cells, respectively. Ficoll-Paque PREMIUM products are based on Ficoll-Paque PLUS but are manufactured in a GMP-compliant environment and under a Quality Management System certified to ISO 13485:2003. Ficoll-Paque PREMIUM can be incorporated into existing procedures where Ficoll-Paque PLUS is used, enabling the isolation of human mononuclear cells from bone marrow, peripheral blood, and umbilical cord blood.

The HiScreen column line has been extended with four new columns—HiScreen Blue FF, HiScreen Ni FF, HiScreen DEAE FF, and HiScreen Butyl HP. The different functional ligands of the new columns allow purification by affinity (AC), ion exchange (IEX), or hydrophobic interaction chromatography (HIC), respectively.

The four new columns are:

> Hiscreen Blue FF: Prepacked with Blue Sepharose™ Fast Flow containing the Cibacron™ Blue 3G ligand to enable purification or removal of proteins such as albumin, interferon, lipoproteins, and blood coagulation factors.

> Hiscreen ni FF: Prepacked with Ni Sepharose 6 Fast Flow immobilized with a Ni2+ charged ligand for capture and purification of histidine-tagged proteins

> Hiscreen DeAe FF: Prepacked with DEAE Sepharose Fast Flow, which contains the weak anion exchanger group DEAE for protein capture applications by IEX.

> Hiscreen Butyl HP: Prepacked with Butyl Sepharose High Performance, which allows high-resolution polishing applications by HIC.

HiScreen columns have small bed volumes (4.7 ml) for fast results and minimal sample/buffer consumption. The columns have a 10 cm bed height and capacity can be increased by connecting columns in series to achieve a 20 cm bed height.
