IntroductionTask2.1description“Task2.1Establishadatamanagementandprocessingprotocolforharmonizationofdataforpooling, integration,qualitycontrolacrosssites.Lead:REGIONH;Participants:UmU,UOXF,MPIB, UB, MRC, VUMC (M1-M6) The task will develop and implement harmonizationprotocols,i.e.developtechnicalandsemanticdatastandards,andqualityprotocolstounifyandbridgethedifferentcodingstandardsoftargetvariablesbetweensites.Itwillalsodefineguidelinesforaddressingdifferencesindatamanagementsystemsandroutines,andMRimagequalitycontrolproceduresacrosssitestosecuretheefficientapplicationandutilizationofthedatatransfer,storageandprocessingsystemdevelopedinT3.1.Moreover,T2.1willfacilitatetheimplementationofthe innovativeposthocharmonizationalgorithms,developed inT3.2,tocorrect MR images for scanner-specific differences in tissue contrast and intensity, non-uniformity.Finally,T2.1willensuretheefficientimplementationandmanagementofthenewdried blood spot assays and online data collection system developed in T3.5 and 3.6,respectively.”BackgroundLifebrainisaEuropeanmulti-sitestudyaimedtocapitalizeonexistingmultidimensionalcrosssectional and longitudinal retrospectively acquired biological, behavioural and cognitive,medical,mentalhealth,andinvivoneuroimagingdata.TheobjectiveofLifebrainistoestablishasolidfoundationofknowledgeforunderstandinghowbrain,cognitiveandmentalhealthcanbeoptimizedthroughthelifespan.Identificationofdeterminantsofbrain,cognitiveandmentalhealthatdifferentstagesofliferequiresalargedatabaseofdetailedinformationaboutbrainimagingmeasures,cognitivefunction,mentalandmedicalhealth,aswellasbiologicalmeasures.Central to (large-scale) multi-site initiatives such as Lifebrain are data integration,standardizationandharmonization.TheinternationalEuropeanaspectaddsanextralayerofuniquepossibilitiesaswellaschallenges.ThecoordinatedresearcheffortswithinLifebrainallows formultiple analytic strategies (2), increase statistical power, allowmoreadvancedsubgroup analyses, enhance generalizability of findings and support cross validation orreplicationoffindingsacrossdatasets.

Statusoftask/state-of-theartinthetopicInJune2016,theinternationalJournalofEpidemiologypublishedthe“MaelstromResearchguidelinesforrigorousretrospectivedataharmonization”(2).Overanine-yearperiod,using“aphonesurveywith34majorinternationalresearchinitiatives,aseriesofworkshopswithexperts,andcasestudiesapplyingtheproposedguidelines”theauthorsdevelopedgenericguidelines for successful retrospective data harmonization that include severalinterdependentsteps:“
1. Definetheresearchquestion,objectivesandprotocol2. Assemblepre-existingknowledgeandselectstudies3. Definetargetedvariablesandevaluateharmonizationpotential4. Processdata5. Estimatequalityoftheharmonizeddataset(s)generated6. Disseminateandpreservefinalharmonizationproducts”
Thecurrentdeliverableprimarilyconcernssteps2and3.OverallLifebrainresearchquestionshavebeenoutlinedinthegrantapplicationandaredetailedinWP4:Demonstration.Steps4,5and6areoutofthescopeofthecurrentdeliverableandarethemainsubjectofotherworkpackages.ObjectivesThe overall objective of the deliverable is to provide a solid foundation for establishing aLifebraindatabaseconsistingofavailablebehavioural,cognitive,medicalandmentalhealth,neuroimaging and biological data, in-depth data harmonization and development andimplementationofguidelines forstandardizeddatamanagementandprocessingprotocolsacrosssites.Wewill:
• CategorizeavailabledataforallLifebrainstudiesandtimepoints• Identifyaninitialsetofvariablesofinterest• Acquire site-specific code books containing information on study design, standard

2. Descriptionofactivities The groundwork for the Lifebrain data harmonization effort was laid down in the grantapplicationandduringtheLifebrainKick-offmeetingheldinBrussels,January16-18,2017.Duringthefirst6monthsofLifebrain,theWP2.1 leader,RegionH,anddataharmonizationteamsofindividualLifebrainsitescommunicatedthroughemail,Slack,OneDrive,Skypeandtelephonetodiscussandorganizeallnecessaryinformation.1.1. Mappingthedata
ThefirsttaskwastomapalldataavailableandinprincipleshareablewithinLifebrain.Based on the discussion at the kick off meeting a spread sheet was created containingidentifieddimensionsandvariables.Thisspreadsheetwasthanpopulatedandifnecessaryextended by each site with information on data availability, and used tests, protocols,questionnairesetc.ThedimensionsthatwereincludedaredisplayedinFigure2.AdetailedoverviewofthevariableswithineachofdimensionsisprovidedinAppendix3.

1.2. Selectionofvariablesofinitialfocus
Based on the detailed overview of in principle shareable data it has been decided,whichvariablestoinitialfocuson(seeFigure3).Thedecisionwasinformedbyobservedavailabilityof data across sites, the focus of the research in WP4, specifically Task 4.2 “Analysis ofindividual pathways/mediator variables in relation to risk formental health problems andresilience” (M18-M48),andTask4.3“Analysis of individualpathways/mediator variables inrelation to risk for poor cognitivefunction and decline versusresilience”(M18-M48).Thedecisionwastofocusoureffortsonthedimensions:
• Studysetup/participants• Demographics• PhysicalMeasures• PhysicalHealth• MentalHealthand• CognitiveTests.
ThelevelofdetailobtainedfortheMRI,Biomarkers,andDNAdatadimensionswassufficienttoinformthespecifictasksonthesedimensionsi.e.

Next,spreadsheetsweredevelopedforgatheringdetailedinformationonrespectivelystudyprotocols, i.e. employed inclusionandexclusion criteria and theorder inwhichbiological,physical, behavioural and cognitive, and neuroimaging assessments were performed (seeFigure 4), and variablemeta-data i.e. site-specific information on variable name, variabledescription,allowablevalues,datatype,dataformat,allowablevalues,valuedescription,andcodingofmissingdata(seeFigure5).

2.1. Generalharmonizationprocedures Thecurrentdeliverablegatheredandorganized informationaboutall variablesof interestacrosscohortsinLifebrain.Whileharmonizationofsomeofthedimensions/variableswillbestraightforward,otherdimensions/variableswillrequiremoreefforttoharmonisegiventhelarge variation in assessment instruments used for some of the dimensions. In our dataharmonizationeffortswewillbuildonother initiatives,suchasEPOSA(16),andBIOSHARE(17)(seeFigure6).Intheautumnof2017,wewillestablishspecificworkgroupsfordataharmonizationofspecificdimensions/variables.Thetaskgroupswillgenerateguidelinesonhowtoharmonizespecificdimensions/variables,andonwhatanalyticapproachesarefeasibleand/orrecommendable.It is importanttodeterminewhethereachofthedimensions/variablesmeasuresthesameconceptinorderfortheharmonizationtoproceed.Ifthevariabledoesnotmeasurethesameconcept,harmonizationofthedatawillnotbepossible.Ifthevariabledoesmeasurethesameconcept,thentheharmonizationprocedurescanproceed.Anotherimportantstepistodeterminewhetherresponsecategoriesoverlap.Iftheydonotoverlap,variableswillbematchedoncategories,e.g.smokingyes/no.Iftheydooverlap,thetaskgroupwilldeterminepossibleharmonizationalgorithmsthatallowsforaslittle lossofinformationaspossible.
