formation that has been found responsible for various outbreaks of foodborne pathogens.

Biofilms are the hydrated matrix of extracellular polymeric substances produced by bacteria

that adhere them to the surface helping the bacterial communities to live in a sessile and

protected environment. Foodborne pathogens exist in natural environment and they form

biofilms on food and food contact surfaces. These biofilms provide bacteria the resistance

against antibiotics, chemical agents and environmental changes as well as helps bacteria to

overcome the host immune responses. Consequently, removal of these foodborne bacteria

and their resistant biofilm is a major challenge for the food industry. Quorum sensing (QS) is

used both by Gram-positive and Gram-negative bacteria including foodborne pathogens for

communication through the production of autoinducers, which are small signal molecules.

QS regulates the expression of several genes in response to population density and controls

the productions various enzymes that are directly or indirectly involved in food spoilage

such as proteolytic, lipolytic, chitinolytic, and pectinolytic enzymes. Furthermore, QS is also

involved in the activation of certain genes in bacteria to secrete extracellular matrix like EPS

and proteins and hence, helps in the formation of biofilm that is resistant to drugs and other

adverse environmental factors. In recent years, detection of quorum sensing signal

molecules in spoiled food items has added a new dimension in the study of food spoilage and

its control. Thus, there is an urgent need to develop quorum-sensing inhibitors that target

the synthesis of autoinducer molecules or block the signaling systems resulting in reduced

biofilm formation to prevent the food spoilage. A number of strategies are currently being

investigated to develop QS inhibitors that can interfere with biofilm formation by foodborne

bacteria so that spoilage of food can be prevented. Considering the need of new anti-QS and

biofilm agents, we screened the rich diversity of Indian medicinal plants, essential oils and

antibiotics. Following outcomes were obtained during the PhD work:

Pseudomomas a foodborne pathogen, resistant to antibacterials, having biofilm

forming capacity and could produce one or more quorum sensing signals (AHL)

molecules.

QS regulated virulence factors and biofilm in Pseudomonasaeruginosa and Aeromonas

hydrophila were inhibited in a concentration dependent manner by antibiotics

Methanol extract of three plants (Mangiferaindica,Psoraleacorylifolia and Trigonella

foenumgraceum) significantly inhibited virulence factors like elastase, total protease,

chtinase, pyocyanin production, EPS, swarming motility and biofilm formation to

varying levels in a concentration dependent manner.

Clove and peppermint oil inhibited biofilm and QS regulated virulence traits of PAO1,

PAF79 and A.hyrophila WAF38. The efficacy of the oils was found mainly attributed

to their essential active constituents, eugenol and menthol.

β-galactosidase assay revealed that all the test agents significantly reduced AHL levels

at respective sub-MICs. Reduced AHL levels are indicative of impaired las-controlled

transcription, therefore reduced expression of lasB gene.

Our results on QS and biofilm inhibition against foodborne pathogens may form the

basis for future research work to address biofilm control and removal in the food

industry.

My PhD work has yielded 11research article published in journals of International

and National repute (mostly ISI Journals) with highest impact factor of 4.07.
