Hair ELISA Drug Kits

Neogen has expanded its forensic drug detection product line by adding hair testing ELISA kits for the most commonly abused drugs on the illicit market—Cocaine, Methamphetamine, Opiates, Synthetic Opiates, PCP, and THC. Using hair samples to detect drugs of abuse has become common practice because of its advantages over traditional sample types. Advantages include:

• Hair is easy to handle and store• Collection is far less invasive than urine and blood• Brief periods of non-use do not affect test results • A larger window of drug detection• Simple retesting• Difficult to adulterate test resultsThese advantages have led hair samples to be extensively used in workplace testing, as well as in other areas, such as legal cases, occupational and traffic medicine, and for historical research.

Intended UseFor the determination of trace quantities of drug analytes in human hair samples.

Features of Neogen’s Hair ELISA Kits:• High sensitivity• Excellent reproducibility• Compatible with automated and semi-automated equipment• Available in 96 and 480 well assay

Assay PrincipleNeogen’s competitive direct ELISAs operate on the basis of competition between the drug or its metabolite in the sample and the drug-enzyme conjugate for a limited number of specific binding sites on the precoated microplate.

The simple assay procedure includes the addition of samples, calibrators or controls to the antibody-coated microplate followed by the addition of the enzyme conjugate (HRP). During incubation, the drug in the sample and the drug enzyme conjugate compete for binding sites on the antibody-

coated microplate. After the incubation, the microplate is washed and K-Blue® Substrate (TMB) is added. After the substrate incubation, the test can be read visually or with a microplate reader. The extent of color development is inversely proportional to the amount of drug in the sample or control/calibrator. The absence of the drug in the sample will result in a blue color (yellow if acid stop is used), whereas the presence of the drug will result in decreased or no color development.

Neogen recognizes that additional substrates and reagents may be required for your application. Therefore, Neogen makes available the following products that are optimized with our test kits.

Assay Procedure

Plates are precoated with the antibody. The plate is ready for use. DO NOT WASH.

Well

Antibodies

A sample or control is added to each well. Next, the drug-enzyme conjugate is added and the mixture is incubated at room temperature.

Conjugate

Sample drug

Wash the p la te to remove all unbound materials.

Wash

Unbound drug and drug conjugate

The bound materials now remain in the microplate.

Bound drug and drug conjugate

Add TMB substrate to each well and allow the color to develop.

Substrate

Qualitative results are obtained by measuring the absorbance reading at 450 nm or 650 nm if no acid stop is used.

Ease of Use• Quick and easy installation, user-friendly control system, and minimal

maintenance required

Excellent Performance• Precise liquid handling capability eliminates variations

Hamilton EquipmentThe Microlab STAR line of workstations was developed based on 50 years of expertise in liquid handling for fast and efficient assay automation and sample preparation. These innovative instruments were specifically designed for regulated laboratories to ensure full GMP-compliant operation.

The Microlab STAR automated liquid handling platform features:

• High throughput processing• Advanced pipetting technology• Easy-to-use VENUS software• Flexible design to allow easy instrument configuration• Broad range of testing applications

In addition to the Microlab STAR workstation Neogen offers the STARlet (medium - large volume sample lab) and the NIMBUS (small - medium volume sample lab) automated work stations.

Neogen offers a full line of Dynex and Hamilton automated equipment for use with our ELISA test kits. The equipment is available for direct purchase or through our Reagent Rental Program, which provides laboratories access to automation with a lower initial investment.

To take advantage of the Reagent Rental Program, customers agree to purchase a predetermined volume of test kits per year from Neogen, based on number of samples and required drug panel. Once an agreement is reached, Neogen will provide the automated equipment to the customer as a rental. The agreement is structured so the cost of the equipment rental, service contracts and test kits are merged, and spread over the duration of the agreement.

The Reagent Rental Program has proven to be a successful way for many of our customers to meet their automation requirements, while at the same time satisfying their ELISA test kit needs.

For assistance in determining your exact needs and selecting the best automated solution for your unique situation, or more details regarding the Reagent Rental Program, contact a Neogen sales representative.
