Abstract

In order to identify molecular markers linked to rhizomania resistance gene, we used F2population

originated from the Holly source (containing Rz1resistance gene).In this study, 188 random ten mer

primers as singly and pair combinations for RAPD analysis were used in bulk segregant analysis (BSA)

of two bulks and F2population individuals. Nine primer pairs showed polymorphism in DNA bulks.

After testing these primer pairs on single plants of two bulks, some of them were selected for the next

analysis on F2 single plants. Then, the markers were evaluated using the frequency of recombinantplants method for estimating distance between markers and Rz1gene. The gene(s) were tagged using

four primer pairs of RAPD including R1 and R2 markers which are 2.32 and 8.3 cM apart from Rz1

gene in repulsion phase and C4 and C1 markers which are 21.4 and 27.5 cM apart from Rz1gene incoupling phase respectively.
