Antibody is a flexible molecule
Antigen-induced conformational changes in IgM
Planar or ‘Starfish’ conformation found in solution.
Does not fix complement
Staple or ‘crab’ conformation of IgM Conformation change induced by
binding to antigen. Efficient at fixing complement
σε «πρωτεϊνικό φορέα» “allergens” such as penicillin, possibly any
drug or chemical in food, cosmetics, paints, fabrics, etc.
Αντιγονικοί καθοριστές
• Antigenic determinants – Specific regions of a
given antigen recognized by a lymphocyte
Concept: Epitopes can bind in pockets or grooves or on extended surfaces in the binding site of antibodies.

pIgR & IgA are internalised
‘Stalk’ of the pIgR is degraded to release IgA containing part of the pIgR - the secretory component

IgA and pIgR are transported to the apical surface in vesicles
B cells located in the submucosa produce dimeric IgA
Polymeric Ig receptors are expressed on the basolateral surface of epithelial cells to capture IgA produced in the mucosa
Μονοκλωνικά αντισώματα
Laboratory produced antibodies Fusion of B-cells (synthesize antibody) &
tumor cell (immortal) Unlimited production of antibodies
Use in Research: specifically label proteins for cellular localization
(immunofluorescence microscopy), Medical diagnostic: Pregnancy test,
detection of tumor markers (surveillance after removal of tumor), diagnosis of infectious agents, STD, AIDS, etc.
Drug Treatment: Attachment of drugs to antibodies, target drugs to specific cells
Μονοκλωνικά αντισώματα
MHC
Μείζων σύμπλεγμα ιστοσυμβατότητας (MHC)
Η ενεργοποίηση των λεμφοκυττάρων γίνεται μέσω των μορίων MHC
Class I molecules display antigens on surface of nucleated cells, resulting in destruction of cells
Class II molecules display antigens on surface of antigen-presenting cells, resulting in activation
of immune cells
MHC molecular structure:
Peptides Binding to MHC Molecules
MHC I molecules bind short peptides, usually between 8 and 10 residues.
The typical length of a class I ligand comprises 9 amino acids.
MHC II ligands consist of 12 to 25 amino acids. A core of nine amino acids is essential for
peptide/MHC binding
