Oneofthetoolsemployedinthedenguecontrolistheapplicationofinsecticidestocontrolitsvector.Inthiscontext,monitoringtheresistancestatusofAedes aegyptipopulations,aswellasdetectingbiochemicalmechanismsimplicatedwithresistanceareveryimportantparameterstodefinecontrolstrategiesofthisvector.Thepublicationofthishandbookthatgivesdetailedinstructionstothequantification,inAedes aegypti,oftheactivityofenzymesrelatedtoinsecticideresistance,throughmethodologiesadaptedtoourconditions,fulfillsanimportantgapandwillenabletheLaboratoriesoftheBrazilianAedes aegyptiResistanceMonitoringNetworktoac-complishthesebiochemicalassayscoordinatelyandinabetterway.Ontheotherhand,monitoringactivitieswillprovidetheHealthSurveillanceSecre-tary/BrazilianDengueControlProgramGeneralCoordinationrelevantinformationtohelptherationalcontrolofthedenguevectorinthecountry.Withthispublication,theHealthSurveillanceSecretary,togetherwiththeLabora-toryofPhysiologyandControlofArthropodVectors/OswaldoCruzFoundation/Riode Janeiro,presentlyNationalReferenceLaboratoryof theBrazilianAedes aegyptiInsecticideResistanceMonitoringNetwork,accomplishesitsroleinthescopeoftheNational Health Surveillance System, contributing to a higher effectiveness in thecontrolactionsofanimportantdiseaseinthecontextoftheepidemiologicalprofileofthecountry.

Theinitialdetectionandcharacterizationofaninstanceofinsecticideresistanceisgenerally accomplished using some form of bioassay. Bioassays are conducted toanswerthequestion:Willthisinsecticidecontrolthisvectoratthisplaceatthistime?Thisisthefundamentalpracticalquestionthatoperationalcontrolpersonnelmusthaveanswered,preferablybeforetheinsecticideispurchased.Whereresistanceisfound,thequestionthenbecomes:WhatdoIdonow?Itistoan-swerthisquestionthatbiochemicalandmolecularmethodsweredevelopedforde-tectingandassessinginindividualinsectsinsecticideresistancemechanismsandthepotentialforcrossresistanceanditsspecificityforparticulartypesofinsecticides.Microplate based biochemical resistance assays were initially developed at the Cen-tersforDiseaseControlinAtlantainthelate70s.Astheyweremorewidelyused,anumberofmodificationsinprotocoloccurred.Theseweremainlyduetothevaryingfiltersetsavailableincommercialmicropolate-readingspectrophotometersandtotheconvenienceinparticularlabsofusingdifferentproportionsofreagents,moreconve-nientbuffersanddilutionfactorsofinsecthomogenates.Ingeneral,twoprotocolshaveperhapsbeenmostwidelyused:theCDCandWorldHealthOrganizationprotocols.However,thestrengthofanyprotocolliesinitsadaptabilitytofulfilltheneedsofspe-cificcontrolprograms.Forthatreason,modifiedprotocolsandoriginalprotocolsaretobeencouragedwheretheyfillanoperationalneedinacountryorprogram.Theonlycaveatisthattheassayprotocolrepresentaccuratelyandpreciselythebiochem-istryofinsecticideresistanceinindividualinsects.TheLAFICAVEprotocoldescribedhereinfulfillstheserequirements,asIhaveper-sonallyverified,andshouldprovetobeavaluableadditiontoourtoolkitofmethodsforaddressingthecrucialissueofdetectinganddescribinginpracticaltermsresis-tanceinvectorsofdisease.Theauthorsareseekingtoprovideapracticallab-orientedmanual,includingwhatwillbeamostusefulCDrom,thatwillleavetheuserwiththesecurefeelingthattheyknowexactlywhattheyaredoingwhenperformingthesetests.Greatcarehasbeenexercisedinanempiricalassessmentofeveryaspectofthemethodology.AtCDC,weareplanningtolinkthisprotocoltoourresistancetrainingwebsite,furtherem-phasizingthatarobustmethodologycanbetailoredtofittheneedsofprograms.

1. INTRODUCTION
Chemicalinsecticideshaveupnowplayedanimportantroleintheinsectdiseasevec-torscontrol.InPublicHealth,themorewidelyusedinsecticidesactontheinsectcen-tralnervoussystem(CNS)andareclassifiedinfourmajorgroups,accordingtotheirchemical nature: organochlorines, organophosphates, carbamates and pyrethroids.Insectsbecomeresistanttothesecompoundsasaresultofeitheranalterationintheirtargetsitesoranincreaseddetoxificationefficacy.Thelattermechanismisknownasmetabolicresistance,andtheincreaseintheactivityofdetoxifyingenzymes(Glutha-tione-S-Transferases,Esterases,MixedFunctionOxidases)contributestotheelimi-nationordeactivationoftheinsecticidescirculatingthevector’sorganism.InBrazil,asaconsequenceoftheuseofinsecticidessince1967,denguevectorpopu-lationsareexposedtoanintenseselectionpressure.Inthissensetheanalysisofboththeresistancestatusofwildpopulationsandthemechanismsinvolvedisofrelevanceto the control of Aedes aegypti. Biochemical methods can be responsible for theearlydetectionofalteredspecimens,yieldinginformationconcerningtheresistancemechanismsofagivenpopulationaswellasapatternofcrossresistancetodifferentinsecticides.Thishandbookwaspreparedinordertogivedetailedinstructionstothequantifica-tionoftheenzymeactivityrelatedtoinsecticideresistanceinA. aegypti.TheworkispartofMoReNAa(BrazilianAedes aegyptiInsecticideResistanceMonitoringNet-work)activities,agroupoflaboratoriesthatacttogetherwiththePNCD(BrazilianDengue Control Program) and give support to the rational control of the denguevectorinthecountry.Theprotocolsdescribedinthishandbookaretheresultofmodificationsinthepro-cedures recommended by the World Health Organization (Hemingway 1998) andbytheCentersforDiseaseControl (Insecticide Resistance Workshop 1998)inordertoadaptthemethodologytoourconditionsandneeds.Theseprotocolshavebeenutilizedwithsuccessbyourgroupsince2002.Themainparametersevaluatedwere:mosquitohomogenizationvolume,natureandconcentrationofbuffersandreactives,sizeofthealiquotsofmosquitohomogenateusedineachassayandreactiontime.Weoptedtouseduplicatesamplesforallen-zymes.WealsoevaluatedtheeffectofcentrifugationonthequantificationofMixedFunctionOxidases,sincetheseenzymesareknowntobeassociatedwithmembranes(Montellano 2004).Furthermore,internalcontrols,correspondingtomosquitoesoftheinsecticidesusceptiblereferencestrain(weuseRockefellermosquitoes),werein-cludedineachassay.

AsEsterasesestãosendoquantificadascomtrêssubstratosdiferentes.Todososensaiosquantificam a atividade enzimática, com exceção do teste de Oxidases de FunçãoMista(MFO),queéumamedidaindiretadoconteúdodaenzima,umavezquequan-tificaogrupamentoheme,seugrupamentoprostético(verSeção 9.3).Otextofoiorganizadoemseçõesparafacilitaraconsulta,desdeocongelamentodosmosquitosatéainterpretaçãodosresultados.Detalhesdapreparaçãodosreagentese dos tampões também podem ser encontrados em seções separadas. We intend to evaluate other parameters, like the differential expression betweenmalesandfemales,throughoutdevelopmentorinadultmosquitoesofvaryingages.Currentlyweworkwithone-dayoldadultfemales.Asfarasbiochemicalassayswithadultsareconcerned,CDCmethodologyutililizesonlyfemaleswhileWHOproto-colalsorecommendstheuseofmales.Bycontrast,WHOprotocolworksonlywithyoungadults,one-dayold,whileCDChasnospecial recommendationon this is-sue.However,temporaldifferencesintheexpressionofenzymesrelatedtoresistancecan occur. Differential expression between larvae and adults is also an importantparametertoresistancemonitoringwhenit isnecessarytocomparebioassaysandbiochemicaltests.WedefinedprotocolstoquantifyAcethylcholinesterase,thetargetsiteintheCNSoforganophosphatesandcarbamates,andthemainenzymesrelatedtometabolicresis-tance.Inconjunction,theseenzymescandetermineresistancetothemajorinsecti-cideclassesemployedinPublicHealth.WedescribethequantificationofEsteraseswiththreedifferentsubstrates.Alltheprotocolsarebasedonthequantificationoftheenzymeactivity.TheexceptionistheMixedFunctionOxidases(MFO)assay,anindi-rectmeasureoftheenzymecontent,sinceitquantifiesthehemegroup,itsprostheticgroup(seeSection 9.3).Tofacilitatehandling,thetextisorganizedinseparatesections,fromfreezingofmos-quitoestointerpretationoftheresults.Detailsofpreparationofbuffersandreagentscanalsobefoundinseparatesections.Finally,fourExcelfilesaccompanythishand-book.Thesefilesenable,inasemi-automaticway,tocomparereplicasofeachsample,tocalculatetheenzymaticactivityofeachmosquito, tocorrectvaluesobtainedbythetotalproteinamountofeachmosquitoandtoelaborategraphswiththeactivityprofileofeachtestedpopulation.WehopethisworkcontributestotheAedes aegyptiinsecticideresistancemonitoringthat ispresentlybeingperformed in thecountry, stimulatingother laboratories toperformbiochemicalassays.Itcouldalsoserveasaninitialguidetotheevaluationofresistancemechanismsinotherinsectspecies.InitiallyplannedasahandbooktobeusedbytheMoReNAanetwork,wewerelaterencouragedtopresentthismaterialtoawiderpublic;thisisthereasonfortheEnglishversion.

4.1. Freezing
a)Useone-dayold(0-24hoursafteremergence)adultfemales,NONbloodfed2.b)FemalesfromthepopulationstobetestedareplacedstillaliveinsideEppendorf
tubes in pools of 40-50. Females of an insecticide susceptible reference strain,usedasinternalcontrolsoftheassays,arestoredingroupsofseven(7).
c)Tubes containing mosquitoes are immediately frozen (females still alive) at-70ºC3.
d)Whennecessary,removetubesfromthefreezerimmediately before use (Section
4.2, c).Maintainthetubesiced.Itisnotrecommendedtore-freezetheremain-ingmosquitoes.
4.2. Homogenization
a)Distribute45numberedEppendorftubesinabasincontainingice.b)Add 30 µl of water to each tube. Use preferably reagent grade water (ex:

10.1. File 1 – data transfer from Soft Max to Excel
ThespectrophotometerweuseinthelaboratoryisassociatedwiththeSoftMaxprogramthatshowstheabsorbancevalueforeachmicroplateposition.However,as mentioned above, the files for data processing were elaborated in the Excelprogram.WhendataaretransferreddirectlyfromSoftMaxtoExcel,theabsorbancevaluesareaccompaniedbyotherparameters,suchastemperatureandreadingtime,whicharenotnecessaryfortheprocessingsteps.These“excess”datashouldbeeliminated,andonlyabsorbancedatashouldberetained.TheprincipalpurposeofFile 1 istoprepareoriginaldatainsuchawayonlytheab-sorbancevaluesaremaintained.Thisprocedurefacilitatestransferenceofdatatotheotherfiles.Bydoingthisitisalsopossibletoworkinothercomputers,whereSoftMaxisnotinstalled.File 1containssixsheets.Thefirstonepresentsfillingoutinstructions;eachoftheremainingsheetswasdesignedtoreceivedatafromall theenzymesofacompletetest.Thesesheetsarereferredtoas“date1”to“date5”andshouldberenamedwiththedateoftheassaythatwillbeinserted.Eachsheethascellsassignedtoreceivetheoriginalabsorbancevaluesfromalltheen-zymes,fromthequantificationoftotalproteinsandfromtheproteinstandardcurve(BSA).Thesecells,thatareblue-labeledandsimulatethe96-wellmicroplates,followthenumberinganddistributionofthemosquitoespreviouslydefined:mosquitoes1to40correspondtothepopulationundertest,andmosquitoes41to45belongtothesusceptiblereferencestrain,asdescribedinSection 4.3.2.

11.2. Criteria for population classification
1)Graph representation: oneoption to theconventionalgraphic representa-tion,showninFigure 2,istheinclusion,inonlyonegraph,ofallpopulationsfromaseriesofevaluations.Thisprocedurefacilitatesthedirectcomparisonofenzymaticactivityamongpopulations.Inthisgraph,theinclusionoftheinsecticide-suscep-tible reference strain enables visualization of the activity alteration level of eachpopulation.Howeverweconsiderthatforthiskindofrepresentationparameterssuchasmeanandstandarddeviationarenotadequate:inseveralcasesindividualsofonepopula-tion do not follow the normal distribution for the characteristics evaluated in thescopeofthishandbook.Inparticular,populationswithanincipientenzymaticaltera-tion,presentingonlyafewaffectedspecimens,areespeciallyrelevanttoresistancestatusmonitoring.Identificationofthesepopulationsallowsinterventioninthefieldbeforeresistanceattainsathresholdthatrepresentsarisktovectorcontrol.Oneofthealternativestotheparameters“mean“and“standarddeviation“istheuseofthemedian.Inthiscase,differentpopulationsarerepresentedinbox-plotgraphs,wherethelineinsidetheboxindicatesthemedian,theboxlimitsrepresentthequar-tilesandtheverticallinesoutsidetheboxexhibitthelimitsofthedistribution(Figure2a).Anothergraphrepresentationthatmayalsobeadoptedappearsasacloudofpoints(scatterplot).Thisrepresentationdisplaysdirectlytheactivityofeachindi-vidual,givingaprecisedescriptionofthepopulationprofile(Figure2b).NeitherofthesetworepresentationsisavailableinFiles 1 to4,hereindescribed.OurlaboratoryprocedureistotransferdatafromFile 4toprogramGraphPadPrismver-sion4.00,inordertoconstructtheboxorscatterplot.

2)How to classify a profile as normal or altered
Enzymaticprofilecomparisonsamongpopulationsthroughparametricandnon-parametric statistical analysis revealed significant differences even when thesedifferences were not functionally relevant. This was, for instance, the case withACEactivityinBrazilianvectorpopulations:statisticalanalysisdemonstratedthatseveral field populations were significantly different from the susceptible refer-encestrain.However,according toWHOcriterion(a functionalcriterion), thisenzymeisconsideredsusceptibleiftheinhibitionofitsactivityinthepresenceofpropoxuris70%ormore(Hemingway, 1998).Whenthiscriterionwasemployedintheevaluatedpopulations,itwasverifiedthatACEactivityofallofthemshouldbeconsideredsusceptible.Wethendecidedtoworkinaccordancewiththedefinitionofaclassificationthatwasrepresentativeofthesefunctionaldifferences.Preciselyduetothis,wefirstevaluatedACEactivity,as thisenzyme isassociatedwitha functionalclassificationcriterionthatiswidelyaccepted(Hemingway, 1998).Aclassificationwasdefinedthatrepresentsadequatelythefunctionaldifferencesthatwere found, regardingnotonlyACEbutalsootherenzymes.Thisclassification isbasedontheuseofreferencestrain99percentileasthecutoffvalue.Ineachpopula-tion,thepercentageofspecimenswithactivityabovethe99percentileoftherefer-encestrainiscalculated.Someexamplesareexhibitedbelow:
