General Hints on Running Mini-Scan Tests ..... 1

f To use the pipettes choose the correct volume for the sample, diluted sample or detector, and place the tip on pipette securely.

f Push the top plunger down and insert pipette below the top of the fluid and let plunger up slowly. Make sure the tip is filled to top. If not, release it and redo. When releasing fluid press down a couple of times.

f Always take a new tip for a new sample and discard the used tips.

f Plug the adaptor into the outlet (110-240) and into the mini-scan unit to charge until the small battery indicator is fully red (charge will be good for 4 hours).

f You will be given a dated sample ID number when you start running samples.

f Record the sample on paper and enter it into the Excel spreadsheet when finished with data retrieval.

f Place the flash drive into the USB port on the mini-scan. To collect data press Done when completing a test and the data will be delivered to the flash drive. To retrieve just insert the flash drive into your computer and download.

f It will appear in Excel format.

f There is space for the tech ID, date and sample identification.

f There is also room with each results for the dilution, sample type, etc., to be added into the Excel format.

f TG kits contain 10 tubes for dilution if necessary (grey caps). f FFA kits contain 4 tubes for dilution (grey caps). f AN kits contains 2 tubes for dilution (grey caps). f All kits contain calibrator tubes (For FFA and AN: 2 yellow-capped C1, 2 green-capped C2, and for TG, 6 clear-capped tubes).

f All kits contain 10 reagent tubes (red TG, Blue FFA, lavender AN). f Use the lower foam insert with tubes as your test tube rack. f Leave bottles and tubes in foam rack for ease of addition.

CALIBRATION

1. Press ‘FFA,’ then ‘CALIBRATE,’ then ‘CONTINUE’

NOTES:• Flash drive should be attached to Mini-Scan if results

need to be saved onto the flash drive.

• If already calibrated, the results will be displayed.

Turn on the mini-scan unit using the top button. Then select the button for the Free Fatty Acid Test.

FREE FATTY ACID TEST (FFA)Purpose: To test is to test feedstock for FFA from 0.1% to 5% with-out dilution, and 5% to 20% with one step dilution.

RANGE: 0.2 - 5% WITH FIRST DILUTION RANGE (EXTENDED): 5 - 25% WITH SECOND DILUTIONREPEATABILITY: +/- 0.2%CORRELATION TO TITRATION: 0.97

• Always listen for beep before removing tube.

• Draw up solution slowly, making sure there are no air bubbles in pipette tip.

2. Insert gray capped tube at prompt for ‘blank tube.’

3. Add 150 uL of reagent from brown bottle into yellow (C1) and green (C2) tube using yellow pipette.

4 . Cap and Shake each tube.

5. Insert green (C1) and yellow (C2) tube into Mini-scan when prompted.

NOTES:• The screen will display the slope, intercept and cor-

relation and if the calibration is good.

• This calibration is good for about 30 days.

• If calibration fails repeat the calibration.

6. ‘CALIBRATION SUCCEEDED’ will be displayed. Press SAVE.

TESTING SAMPLESThis test is to test samples for FFA from 0.1 to 5% without dilution, and 5-20% with one step dilution.

1. Press ‘FATTY ACID TEST,’ then ‘RUN TEST’

2. Insert grey capped tube when prompted for ‘BLANK.’

NOTES:• Flash drive should be attached to Mini-Scan if results

need to be saved onto the flash drive.

NOTES:• Take up sample slowly. If there are bubbles in pipette

tip, redo.

• If your sample is liquid, you can add 100ul directly to the blue capped tube.

• Use KimWipe to wipe outside pipette tip and tube.

• If your sample is not liquid or very cloudy then dilute as described below or warm briefly at 40 C in the ther-mal heater, microwave or hot plate.

• If the sample is difficult to warm, expected between 0 and 5%, is solid, or very cloudy, dilute your sample.

• To dilute:

Add two 200 uL of the sample into the grey capped tube.

Shake and warm if necessary, and then add 100ul of this dilution into blue capped tube.

3. Add 100 uL sample into blue capped tube using blue pipette

4. Put 100 uL reagent (brown bottle) into same tube. Cap and Shake.

5. Follow prompts and insert sample twice.

5. Press ‘DONE’ or ‘NEXT’ to read another sample.

• Result should be multiplied by 10 for final results.

• Low FFA readings are below 0.1%

• High FFA readings are above 5%
