NOWPak® Containers are the ideal packaging format for phosphoramidites used in large scale applications such as the synthesis of oligonucle-otide therapeutics. The disposable liners and pressurizable stainless steel overpak enable stringent control of water content and limit the potential for contamination during handling.

Dissolution of Phosphoramidites in NOWPak Containers

Containers are supplied with a pre-weighed amount of phosphoramidite. The amount of solvent (acetonitrile) added to the container is determined by the amount and molecular weight of phosphoramidite and the desired molarity of the solution.

1. Attach a dispensing head to the NOWPak container and place it on a digital scale.2. Connect the solvent line to the NOWPak container and tare the scale. 3. Open the valve and allow pressure to dispense the desired amount of solvent into the container. Dispense into the container the amount (by weight) of solvent shown in the table below.4. Disconnect the container, place on an orbital shaker, and mix.

Once dissolution is complete (4-5 hours), the container is connected directly to the oligonucleotide synthesizer.

Liner minimizes waste by ensuring maximum delivery of the phosphoramidite solution.

Two-piece metal closure assembly

ND-CD Connector

Optional aluminum breakseal closures are designed for re-use. Only the breakseal comes in contact with the fill material and is replaced prior to each use. These closures are used with NOWPak Dis-pense Connectors.

The NOWPak dispenser is designed to provide a clean, convenient and completely sealed method for dispensing chemicals from NOWPak containers and is used with the breakseal closure. The fluid path of these connectors consists of an inert fluoropolymer.
