    PLANT STORY

Portulaca oleracea is an annual succulent with bright yellow

flowers surrounded by a cluster of leaves. The plant is known

as purslane, little hogweed or pusley and Machixian in China

and Machihyun in Korea meaning ‘horse teeth amaranth’ due

to its resemblance to horse’s teeth. It is an edible nutritious

vegetable, distributed all over the world and generally used in

    Korean and Chinese traditional medicines.

    PLANT STORY

In oriental herbal medicine, purslane is used to remove

toxic heat and substances, to arrest bleeding and to treat

infections and eczema. It is one of the richest green plant

sources of omega-3 fatty acids. The recent scientific

research showed that P. oleracea exhibited a wide range

of biological activity including anti-inflammatory, anti-

    oxidant and wound healing activities.

    ANTI-ALLERGIC RESPONSE

An allergic reaction takes place when the immune system of an

animal or human responds to the exposure of a variety of

    substances called allergens.

Histamine and ß-hexosaminidase are considered to be

markers of mast cell degranulation, mediators of inflammatory

responses. Thus, the ß -hexosaminidase activity in the medium

is used as a marker of mast cell degranulation. The inhibition

of ß -hexosaminidase release is required for an

    antiallergenic response.

    WHAT IS INFLAMMATION?

Inflammation is part of the complex biological responses to wide range

of harmful stimuli including injury, tissue necrosis, infection, and

irritants. The purpose of inflammation is to destroy (or contain) the

damaging agent, initiate repair processes and return the damaged

tissue to useful function. The symptoms of inflammation are redness,

swelling, heat, and pain, which are caused by increased blood flow

into tissue. The immune system is responsible of protecting our body

from the harmful stimuli and of maintaining homeostasis. Disorders of

the immune system can result in autoimmune diseases, inflammatory

diseases, and cancer. In an attempt to protect the body, the immune

system might overreact to the stimuli, and this might cause allergy or

    inflammatory reactions.

    INFLAMMATION MECHANISM

    PortulacaExtract-TLS

A variety of enzymes are secreted when cells on the skin tissue are stimulated by stimulus.

Nitric oxide is an important molecule in cell signaling and plays an important role in biological processes. Small amounts of nitric oxide help cell

activity, but as the amount and the reaction time become longer, intracellular COX-2 and LOX-5 levels are increased. Portulaca Extract-TLS has

been shown to inhibit NO and Lipoxygenase, which play an important role in inflammatory signals, and it is helps relieve and improve inflammation

    reaction.

    WOUND HEALING ASSAY

Scratch Wound Healing Assay has been widely adapted and

modified by researchers to study the effects of a variety of

experimental conditions, for instance, gene-knockdown or

chemical compound treatment, on cell migration and

    proliferation.

The basic principle of the assay is that, a “wound gap” in a cell

monolayer is created by scratch, followed by monitoring the

“healing” of this gap by cell migrating and growth towards the

center of the gap, hereby filling up the “gap”. Factors that alter

the motility and/or growth of the cell can lead to increased or

decreased rate of “healing” of the gap.

After 24 hours of culture, cells were scratched with a 200 μL pipette tip and cultured for additional 48 hours. Photos for the

stained monolayer were taken on a microscope.
