11. Zusammenfassung
Die vorliegende Dissertation ist ein Teil des Projektes „Oligosaccharid-analoge von Polysacchariden", in welchem unter anderem der Einfluß von
intra- und intermolekularenWasserstoffbrücken auf die Struktur und die
Eigenschaft von Cellulose anhand von Acetylenosacchariden untersucht
wird, welche sich aus D-Glucose, D-Cellobioseund den höheren ß-D-1,4-Glucanen ableiten. In diesen Analogen sind die exo-glycosidischenSauer¬stoffatome ganz oder teilweisedurch Butadiin-l,4-diylgruppen ersetzt.
Diese Arbeit beschreibt die binominale Synthese von oligomeren Acetyleno-cellobiosen (bis zur Stufe des Octameren) aus geschützten l,4'-trans diethi-
nylierten Cellobiosiden als Monomere. Die Herstellung der Monomere
erfolgte durch eine Glycosidsyntheseaus zwei ethinylierten Glucopyranosen.Der benzylierte Akzeptor 227 wurde aus Glucose in elf Stufen und einer
Ausbeute von 8% erhalten. Die Einführung der Ethinylgruppe von 227
erfolgte durch Addition von Lithium-Trimethylsilylacetylid an das Lacton
226, gefolgt von der vollständig diastereoselektivenReduktion des
entstandenenHalbacetals mit Triethylsilanin Gegenwart von Bortrifluorid-etherat. Ein effizienterer Zugangzu ethinylierten Glycosylakzeptorenwurdeaus Laevoglucosan (228) entwickelt.Durch retentive alkinylierende Acetal-spaltung der 2,4-disilylierten 1,6-Anhydroglucosen232 und 233 mit
Lithium-Trimethylsilylacetylid und frisch sublimiertemAluminiumtrichlo-rid in Gegenwart von einem Äquivalent sym.-Collidin und anschließendesaure Hydrolyseder Silylether wurde in Ausbeuten von 60 bis 70% das
kristalline 1,5-Anhydroglucit 234 erhalten. Die verschieden geschütztenGlycosylakzeptoren 235, 239, 241, 243, 245 und 247 wurden in drei bisvier Stufen und Ausbeuten zwischen 43 und 66% aus 234 erhalten. Die
Synthese der in Stellung vier ethinyliertenGlucosyldonoren 255-258, 260-
262, 264, 265 und 267 erfolgte ebenfalls aus Laevoglucosan (228) undumfaßte sieben bis acht Stufenbei Gesamtausbeuten zwischen 25 und 35%.Die besten Glycosidierungen des benzylierten Akzeptors 227 wurden mit
den acylierten Phenylthioglycosiden257, 258 und 267 erreicht (80-85% an
268 bzw. 269); die Glycosiderung des methoxybenzyl- und silylgeschütztenAkzeptors 247 mit dem kristallinen cc-D (260) oder ß-D-Trichloracetimidat261 lieferte in über 85% Ausbeutedas Monomere 273. Die Kristallstruktur
der anomeren Trichloracetimidate 260 und 261 wurde röntgenographischbestimmt(Dr. W. B. Schweizer, ETHZ) und repräsentiert die erste Festkör-

perstrukturdieser wichtigenKlasse von Glycosyldonoren.Im ungeschützten Monomeren 201 wurde eine starke intramolekulare
C(5)OH-0(5') Wasserstoffbrücke NMR-spektroskopisch ((D6)DMSO)nachgewiesen, welche bewirkt, daß die beiden Pyranoseringeum ca. 170°
zueinanderverdreht sind.Ein neues Paar orthogonaler Alkinschutzgruppen wurde entwickelt. Trime-
thylgermyl (TMG) und Trimethylsilyl (TMS) geschützte Dialkine wurden
unter geeigneten Bedingungenin hohen Ausbeutenprotodesilyliert oder pro-
todegermyliert. In ähnlicher Weise lassen sich TMG/TMS-Dialkinein fast
quantitativen AusbeutenBromo- und Iododegermylieren.Zusätzlich wurde
eine degermylierende oxidative Homokupplung von TMG/TMS-Dialkinenund eine Methode zur Umwandlungvon TMG-Alkinen in Alkinone ent¬
deckt.Durch binominale Synthese aus dem teilweise benzylierten TMG/TMS-Mo¬nomeren 311 wurden das Dimere 363 und das Tetramere 362 hergestellt.Während 363 sich noch acetolytisch debenzylieren ließ, versagte die
Methodebeim Tetrameren 362. Nach Abspaltung der Acylgruppenvon 365
wurde das Dimere 366 erhalten, dessen intramolekulareC(5)OH-O(50und C(7")OH-O(5"0Wasserstoffbrücken in (Ü6)DMSO NMR-spek¬troskopisch nachgewiesenwurden. Die binomiale Synthese des Tetrameren
379 und des Octameren 380 erfolgte aus dem orthogonal (TMG/TMS)geschützten Monomeren 313. Alle ungeschützten oligomeren Acetyleno-cellobiosen sind in Wasser, Methanol und DMSO löslich. Die iH-NMR
Spektren ((E>6)DMSO (201, 366, 379, 380), CD3OD (201, 366, 379) undD2O (380)) zeigten keine Hinweise auf intermolekulare Assoziationen. Die
Kraftfeldberechnung (MM3*) der Struktur des Octameren 380 in Wasser
ergab, daß eine 71-Helix gegenüber einer gewelltenlinearen Anordnung umca. 6 kcal-mol-l stabiler ist. In den 1H-NMR Spektren ((D6)DMSO, D20)von 380 waren keine Anzeichenfür diese (oder eine andere) Konformationzu erkennen.

Summary
The dissertation is part of the project "Oligosaccharide Analogues of
Polysaccharides", where the influence of hydrogen bonds on the structure
and properties of cellulose is assessed by comparing the structure ofcellulose with the one of a series of acetylenooligosaccharides obtained from
D-glucose, D-cellobiose, and higher ß-D-l,4-glycans. In these, all or some
exo-glycosidic oxygens are replaced by butadiynediyl units.
This work describes the binomial synthesis of higher oligomers (up to the
octamer) derived from \,A'-trans diethynylated cellobiosides as monomers.
The monomers were preparedby glycosidation of monoethynylated glucosylacceptors with monoethynylated glucosyl donors. The benzylated acceptor227 was preparedfrom D-glucose in 11 Steps and in an overall yield of 8%.The ethynylgroup was introduced by addition of lithium(trimethylsilyl)-acetylide to the lactone 226, followed by reductive dehydroxylationof thehemiacetal with triethylsilane in the presence of borontrifluoride-etherate.The ethynylated glucosyl acceptors were efficiently prepared from 1,6-anhydroglucose (228). Retentive ethynylating opening of the 1,3-dioxolanering of the 2,4-di-O-silylethers232 and 233 with lithium(trimethylsilyl)-acetylide and freshly sublimed aluminiumtrichloride in the presence of one
equivalent of syra-collidine, followed by acid catalyzed hydrolysis of the
silyl protecting groups resultedin the crystalline 1,5-anhydroglycitol 234 in
yields of 60 to 70%. The differently protected glucosyl acceptors 235, 239,241, 243, 245 and 247 were obtained from 234 in three to four Steps andin overall yields of 43 to 66%. The monoethynylatedglucosyldonors255-258, 260-262, 264, 265 and 267 were synthesized from 1,6-anhydro-glucose (228) in seven to eight steps and in overall yields between 25 and35%. The best results in the glycosidation of the benzylated acceptor 227were obtained with the acylated phenylthioglucosides257, 258 and 267
(80-85% yield of 268 or 269); glycosidation of the methoxybenzylated and
6-O-silylated acceptor 247 with the oc-D- (260) or /3-D-trichloroacetimidate261 gave the monomer273 in 85% yield. The structure of the ano-meric
pair of trichloroacetimidates260 and 261 was determined by single crystalX-ray analysis (Dr. W. B. Schweizer, ETHZ).The unprotectedmonomer201 possesses an intramolecular C(5)OH--O(50hydrogenbond as evidenced by NMR spectroscopy((D6)DMSO) which ledto a preferred conformation where the glycosidicallylinked pyranosyl units

are rotated by ca. 170°.A new pair of orthogonalalkyne protecting groups was developed.Several.trimethylgermyl- (TMG) and trimethylsilyl- (TMS) protected dialkyneswere synthesized and regioselectively deprotectedby protodesilylation orprotodegermylation. Similarly, TMG/TMS-dialkynes were bromo- and
iododegermylated in high yields (>90%). In addition, a new degermylatingoxidative homocoupling of TMG/TMS dialkynes and a method for thetransformation of TMG-alkynes into alkinones was discovered.The binomial synthesis of the dimer 363 and the tetramer 362 started from
the partially benzylatedTMG/TMS-monomer311. Acetolytic debenzylationof 363 gave 365, but the analoguous debenzylationof 362 provedunsatis-
factory. Deacetylationof 365 led to the unprotecteddimer 366. Intramole-cular C(5)OH-0(5') and C(7")OH-0(5'")hydrogen bonds within eachcellobiose unit of 366 were proven by NMR spectroscopyin (D6)DMSO.The tetramer 379 and the octamer 380 were obtained by a binomial
synthesis starting from the TMG/TMS-monomer313. All unprotectedoligomers (201, 366, 379, 380) are soluble in water, methanol and
dimethylsulfoxide.Their 1H-NMR spectra ((D6)DMSO (201, 366, 379,380), CD3OD (201,366,379) and D20 (380)) showed no signs ofintermolecularassociation. Calculation (MM3*) of the structure of theoctamer 380 in water indicated that a helical arrangement (71-Helix) is
preferred by ca. 6 kcal-mol-1 over a (corrugated) linear structure. Noevidence for this (or any other) conformation of 380 could be obtainedfrom the 1H-NMR spectra ((D6)DMSO,D20).
