Speaker and Presentation Overview Dr. Bramasta Nugraha Project Leader University of Zürich Future Role of Cell Reagent Generation Towards More Predictive Model Systems and Readouts Dr. Bramasta Nugraha completed his undergraduate study at Nanyang Technological University Singapore in 2007 in School of Materials Science and Engineering where he had started to get involved in lab research since his second year study in biomaterials research. He further obtained his Ph.D. in Bioengineering (Tissue Engineering) at the National University of Singapore in 2013 under the guidance of Prof. Hanry Yu at the Dept. of Physiology School of Medicine and the Institute of Bioengineering and Nanotechnology A*STAR Singapore. During his Ph.D. dissertation he had involved in several research projects performed together with large pharmaceutical industries such as Johnson & Johnson, Roche, and Biopredic to utilize 3D liver/hepatocyte spheroids culture to do drug safety testing, hepatitis B & C infection studies, and hepatocyte progenitor cells maturation, respectively. The cleavable cellulosic sponge he developed in Singapore had been filed as patents in Singapore and in the US, as well as licensed to a biotechnology company in Taipei, Taiwan where he had involved in the technology transfer process. Upon Ph.D. dissertation completion, he was then awarded Roche Postdoctoral Fellowship in 2012 to continue his research in 3D cell culture in Basel, Switzerland in collaboration with Swiss Federal Institute of Technology Zurich/ETH Zürich. During his appointment at Roche, he broadened his expertise in a kidney 3D co-culture model to detect fibrosis onset with image-based analysis using 2-photon microscope and reporter cell lines. Currently, he is a project leader at the University of Zurich, Switzerland where he is developing cardiac organoids from various human-induced pluripotent stem cells to model human cardiac disease in vitro with robotic liquid handler automation system. Ilya Lukonin Postdoctoral Researcher and HCS Specialist Friedrich Miescher Institute Basel Regenerative Landscape of Intestinal Organoids Ilya has received his Bachelor’s and Master’s degrees in Biochemistry at the Freie Universität Berlin. During his Master’s studies he developed a profound interest for imaging and quantitative image analysis applied to cellular biology. To this end, for his Master thesis he joined the lab of Volker Haucke at the Leibnitz Institute for Molecular Pharmacology (FMP) where he studied endocytic processes using super-resolution microscopy and high throughput imaging. Next, he went on to build up his competencies in image analysis during his work as student assistant, developing computational tools for confocal and super-resolution microscopy. Driven by his interest for imaging and multivariate data analysis, he joined the lab of Prisca Liberali at the Friedrich Miescher Institute where he is investigating the molecular mechanisms that underlie the development of intestinal organoids by means of high content screening. PD Dr. Marianna Kruithof-de Julio Head of the Urology Research Laboratory University of Bern Organoids and Patient-derived Xenografts: Translational Research of Urological Cancers PD Dr. Marianna Kruithof-de Julio has expertise in patient-derived xenografts, organoids, mouse models of castration responsive prostate cancer, bioluminescence cell tracking and monitoring in the living mouse, and in animal models of both osteoblastic and osteolytic bone metastases and cancer stem cell of origin. Dr. Marianna Kruithof-de Julio (Dept. of Urology, University Hospital, Bern and Dept. for BioMedical Research, University of Bern) is the Head of the Urology Research Laboratory and is a leading expert in the field of PCa stem-cell biology. She has set-up the ex vivo model proposed in this study, and is an expert in in vivo experimentation. In addition, Dr. Kruithof-de Julio is the Director of the Organoid CORE facility, part of the Bern Cancer Research Cluster (BCRC) and Associate Member of the NCCR RNA and Disease.

Sylvia Suen, MSc.Research TechnicianUniversitair Medisch Centrum, Utrecht

Application of Human Colon Organoids in Cystic Fibrosis: A Multisite, International Validation StudyIn 2016 Sylvia Suen obtained her Master of Science in Medical Biology with a specialization in Clinical Cell Biology at the Radboud University Nijmegen, The Netherlands. During her Master’s she performed her first internship at the Radboud Institute for Molecular Life Sciences (RIMLS) in Nijmegen where she studied the Wnt signaling pathway in osteoarthritis. In her second (international) internship, at the University of Umeå, Sweden, she focused on the optimization of a 3D cell culturing system to enable the efficient culture and differentiation of mouse adipocyte stem cells. After becoming passionate about organoid technology she was determined to develop her skills in this specific subject. Since 2016 she works as lead research technician in the research team of Prof. Dr. Jeffrey Beekman, who was the founder of clinically applying human colon organoids in cystic fibrosis (CF) research and diagnostics through the development of the Forskolin induced swelling (FIS) assay. In the past years she has become an expert in the isolation and culture of human colon organoids and performing the FIS assay to perform research in cystic fibrosis. She also focused on the development and optimization of novel assays such as the Steady State Lumen assay (SLA) to enable further efficient organoid models to support CF research.

Human Neural Organoids for Treatment Testing and Neuroscience ResearchDr. Mosser studied Alzheimer’s disease at the Ecole Polytechnique Fédérale de Lausanne. Attracted by entrepreneurship, he has been working in the startup incubator Eclosion before he led his own startup project in the field of diagnosis. As the chief operating officer of Neurix, he brings his entrepreneurship know-how, manages product development, and ensures optimal functioning of the laboratory.

Two-dimensional Intestinal Stem Cell-derived Organoids as a Model to Test Nutritional Compounds for Intestinal HealthLaura M'Rabet obtained her Master’s degree in Molecular Sciences from Wageningen University and Ph.D. in Physiological Chemistry from Utrecht, where she gained experience in applied research and marketing in international industry.

In 2017, Utrecht Life Sciences seconded her to the Utrecht Science Park Foundation to open Utrecht Science Park's unique R&D infrastructure and forge connections between people and organisations from within and outside Utrecht Science Park.

Facilitating and conducting applied research is her passion. It is what inspires her on a daily basis, and has made her career an enjoyable experience.

Franziska Wienholz, Ph.D.Scientific Support Specialist EMEACorning Life Sciences

New Tools for 3D Research: Convenient Culture of Organoids and Efficient Scale-up of Spheroids Franziska received her Ph.D. from the Erasmus Medical Center (Rotterdam) in The Netherlands where she focused on the molecular regulation of DNA damage repair in order to prevent premature aging and cancer.

Since 2017 she is a Scientific Support Specialist at Corning and is involved in selecting the most appropriate product for the specific application of the researcher, and she subsequently supports the design, and set up of experiments by providing workflow solutions and protocols. During seminars and webinars, Franziska presents information and scientific data on products to enable scientists to use 3D cell culture for efficiently creating in vivo-like environments and predictive models.

Dr. Matthias Nees Principal InvestigatorUniversity of Turku

High Content Screening and Imaging in 2 to 4 DimensionsDr. Matthias Nees is the founder and coordinator of HCSLab, a service unit hosted and supported by the University of Turku, Institute of Biomedicine. Simultaneously, he is a team leader at the FICAN West Cancer Centre, a national research program under construction that has the goal to coordinate cancer treatment, research, and education in Finland. His group at FICAN West engages in personalized medicine and currently develops cell- and tissue-based model systems for chemosensitivity tests in vitro. Dr. Nees has previously worked at the VTT National Research Centre of Finland, where he was a group leader and principal investigator in the field of systems biology, high throughput and high content screening. Before moving to Finland in 2005, he held positions at the National Cancer Centre NCI at NIH in Bethesda, Maryland, USA, the Molecular Medicine Partnership Unit at EMBL Heidelberg, the German Cancer Research Centre DKFZ, and the University Hospital Heidelberg – always related to translational oncology, personalized medicine, and drug discovery. Since 2018, he is also a visiting professor at the Medical University in Lublin, Poland, with the goal to set up laboratory operations for personalized medicine specifically in the field of head and neck cancers.

Dr. Antti HassinenHead of HTS Imaging and Analysis UnitFIMM-HCA, Helsinki

3D High-content Imaging and Phenomics in Drug ProfilingAntti Hassinen is the head of the open-access High-Throughput Imaging and Analysis Unit (HCA-FIMM) in the Finnish Institute for Molecular Medicine (FIMM, HiLIFE) in the University of Helsinki, Finland. He has previously worked in the Faculty of Biochemistry and Molecular Medicine in the University of Oulu and gained his Ph.D. in the group of Docent Sakari Kellokumpu in 2012. The work was performed in the field of glycobiology and included studies on complex formation of glycosyltransferases in hypoxia and the homeostasis of the Golgi microenvironment. The work focused on the development of confocal and high throughput microscopy techniques with 2D and 3D cell cultures in close collaboration with the Oulu Bioimaging network OBI.
