The MW ladder marker was loaded on lane 1. The characteristic smearing patterns of Ph-L12, as shown in lanes 3 and 9, appeared in all the lanes of complexes (lane 4 to 8). The Ph-L12 area of smearing was indicated in the right side of gel panel. The band of WT Ph-P0 (closed circle) or its mutants (closed triangle) was also detected in individual lanes. Fig. S5. Alignment of the amino acid sequences of P0 and L10. The amino acid sequences of

P0 from 11 species of Eukarya and 10 species of Archaea, and of L10 from 28 species of

Bacteria were aligned by clustalW algorithm followed by manual inspection. The alignment is

separated by spaces into three domains, upper, middle and lower, corresponding to the

urkingdoms, Eukarya, Archaea, and Bacteria. The positions of the N-terminal and C-terminal

regions with the symbol ‘x’ attached on the space between the archaeal and bacterial domains

could not be aligned between the eukaryal/archaeal and the bacterial sequences, while those of

the C-terminal region with the symbol ‘x’ between the eukaryal and archaeal domains could not

be aligned between the eukaryal and archaeal sequences. Selected positions used for

phylogenetic analysis were marked with ‘*’ above the alignment. The putative binding sites of

stalk dimers were masked based on the results of biochemical experiments on the silkworm

(Bmo) P0 for eukaryal and archaeal sequences (31), and on the reported structural information

for Thermotoga maritima (Tma) L10 for bacterial sequences (23). The position of the inserted

sequences is enclosed by a rectangle. In this portion all archaeal sequences have insertions

compared to eukaryotic sequences, while several bacterial sequences have insertions compared

to other bacterial sequences. The archaeal and bacterial insertions could not be aligned either.
