In this interview, the Editor of the Aging Matters™ magazine, Phil Micans (PM) interviews Walter Pierpaoli, M.D., Ph.D. (WP)

Dr Pierpaoli is famous for his scientific and clinical published research into melatonin, the thymus, immunity and much more. In addition, he is also recognised for being one of the world’s foremost antiaging physicians. In this interview, Dr. Pierpaoli describes his latest work and how it impacts his ‘rotational theory of aging.’

PM: “Once again Dr Pierpaoli it is my pleasure to be talking with you on our favorite subject- aging! I know it is a subject that fascinates you, me and many of our readers.”

WP: “Quite right Phil, as always it is a delight to

be doing so. Who isn’t interested in how they are aging?”

PM: “Let us start then with the 64 billion Dollar question! Please tell us what you believe aging to be.”

WP: “In a nutshell, the pineal gland, via its links with the pituitary gland and the hypothalamus, is the biological clock network; it is a true scanner for growth, fertility, aging and also death.”

PM: “So in-effect, the pineal gland acts as a day-night counter limiting our lifespan?”

WP: “Yes, it seems blatantly obvious to me. Are we different from dogs, cats and mice? No, we are all mammals!”

PM: “I think that an age-clock counter explains why there have only been a handful of people who have made it past 120-years of age. If it were, for example, just down to free radicals as the cause of aging, there should have been some numbers of persons with excellent free-radical scavenging activity who would have had significantly longer lifespans by now. My assumption is based on the estimates that there have been 128 Billion homo-sapiens who have lived on Earth- that’s stated on Wikipedia so it must be right!!! (Ha ha)”

WP: “As you well know, I do not believe in the fairy tale of poisonous radicals. It is fashion in science, such as the other fairy tale, that you can prolong life by maintaining telomere length. It is a desperate attempt which makes me laugh. The body takes care of itself, if the central ‘clock’ is synchronized!”

PM: “Of course many people know that lifespan averages are increasing worldwide; but this is the mean lifespan for the average person, the maximum lifespan doesn’t seem to have expanded much- if at all. Roman records suggest there were individuals living up to 120-years, even thousands of years ago.This maximum lifespan of 120 years is even recorded in the Bible.”

WP: “What prevents man to live normally to 110, 120 or 130 years are diseases! Now today’s highly polluted environment facilitates all kind of diseases,

which when combined with bad genetics is the main cause for atherosclerosis and tumours, which kill 80-90% of the population. New generations are now doomed to age earlier. In fact, the long-lived people are those born before 1950, before the environmental disaster. They were raised and grew up in a more pristine nature- it is my own generation. Longer and healthier life now is a lie and I can see it in children! Believe it or not, healthy children do not exist anymore!”

PM: “So by understanding that the pineal can recognise day from night, by releasing melatonin during darkness, and that this action could be a counter, why is your theory of aging called the rotational theory?”

WP: “It is time to explain planetary and atom rotational movements, all being in cyclicity etc. Everything in the cosmos is rotational and totally synchronized, from the atoms to the galaxies! Every cell in our body follows a strict circadian, day-night cyclicity, lunar and seasonal periodicity. We are generated, grow, procreate and follow our species program in a completely rotational system, controlled by our ‘clock’ in the hypothalamic-pituitary-pineal network. We are a bunch of ‘tubes’ - arteries, gastrointestinal, urinary, etc., all strictly controlled by hormones, which in turn synchronically maintain cyclicity and periodicity, all linked to the rotational forces of the lunar-planetary system. After all, is the menstrual cycle different from the moon cycle? No it is not; but disruption of our adaptability to the rotational forces leads to diseases and early aging.”

PM: “So can melatonin, when taken as a night-time supplement, trick the age-clock? What have your animal experiments told you about their maximum lifespans and indeed the condition of their health in their later years?”

WP: “Melatonin a precise night signal from the pineal gland; my murine experiments clearly illustrate that by ‘putting to rest’ the pineal gland, with night administration of melatonin, that the mice do not become immortal, but they do prolong their vitality which enables them to reach their maximum life-span, which is 3 years and would correspond to 110-130 years in man. It is simple and clear, with

melatonin, which protects the pineal gland, we can prevent the ravage of time afflicting our later years.”

PM: “And how might this play out in humans in terms of lifespan and health?”

WP: “I do believe that ‘resetting the pineal clock’ with melatonin, formulated in a way which produces and mimics the physiological night peak, we can enter a new phase of human life which allows all of us to reach our maximum lifespan in a useful way.”

PM: “As a medical doctor I appreciate you have been treating many thousand of patients over decades, how have your patients’ health been improved by your protocols?”

WP: “From the publication of the USA bestseller, ‘The Melatonin Miracle’ in 1995, I was almost forced by people and circumstances to apply my discoveries to patients. I verified my re-synchronization method in several thousands of patients. All of them are cured from even the most hostile diseases such as cardiovascular; neurodegenerative a.o. Tumors can be easily prevented. I did not see a single case in my patients. A book is now being assembled collecting letters from the cured patients which is entitled; ‘The happy patients of Dr. Pierpaoli.’ Isn’t it the ultimate defiance?”

PM: “I happen to know that your protocols include more than melatonin supplementation; can you describe some of your other fundamental approaches to optimal health please?”

WP: “If the pineal gland controls our rotational adaptation to lunar and planetary cyclicity, obviously the night protection of the pineal gland with melatonin is fundamental. However, what will happen upon night protection of the pineal is a cascade of positive events which concern hormone inter-synchronization. In most cases I observe pituitary and thyroid dysfunctions, mostly hypothyroidism, which goes undetected owing to the laboratory assays which are totally misleading and wrong! I think this is a complete disaster. Clear or latent, insidious hypothyroidism is affecting 99% of the adult population. I do not know the reason for it and probably it is the main cause of pathologies in the new generations. But nobody talks of it! I

developed my own parameters. For example, I keep TSH at zero and ‘free’ T3 with ‘free’ T4 two-three times higher! TSH is a carcinogenic polypeptide similar to prolactin, whose value should not exceed 10 ng/ml. But it is prolactin that is the most dangerous carcinogenic, tumor-promoting agent in men and women. Also, in the course of my studies, in 1989, I discovered some totally unexpected activities for TRH, which has the totally misleading name of thyrotropin-releasing hormone. I wish to tell its story in a book. In fact, my observations on TRH activities over the past 25 years, allow me to say that a different level of basic regulation exists in the body- which goes back to the origin of life. This tiny molecule occupies a fundamental role, including the regulation of insulin production and release and consequently the precise control of blood sugar!”

PM: “Ah yes! TRH or thyrotropin releasing hormone; we did cover it in the lead story in Aging Matters™ issue 3, 2012 (copies of which are all online at: www.antiaging-systems.com/antiaging-magazine) Perhaps I could ask if you could please do a quick recap of the beneficial properties of TRH?”

WP: “My published experiments have shown that TRH may become a key molecule to invert the clock of aging. For example, it regenerates testicles in very old animals and also restores kidney function in sclerotic infiltrated kidneys. It seems to produce a regeneration of beta cells in the pancreas of diabetic animals. I am deeply interested to pursue clinical studies with TRH.”

PM: “I appreciate that you like to refer to TRH, much like melatonin, not as a hormone, but as a life generating molecule, why is that?”

WP: “The semantic attribution of ‘hormones’ to TRH and melatonin is an outrage to good sense and human intelligence. Both molecules are totally devoid of any side- or toxic effects even at huge concentration and even when given intravenously. In spite of this, European countries have waged a war against them, in particular Switzerland and Germany, obviously fearing their therapeutic activities when confronted by the profits generated by the industry of the chemical, man-made drugs.”

PM: “TRH is actually a tri-peptide and as many of our readers are aware, there is a wave of interest in the ways peptides work within the body now. So let us cut to the chase, after all, knowledge is worthless without power, or in this case - application. How do you recommend your patients use TRH, in terms of its dose and its timing?”

WP: “With 20-years of studies at my disposal, I developed an elegant method to allow TRH to enter the blood stream. In fact, the fragile molecule cannot be given orally because it is rapidly destroyed in the stomach. Also its production costs are prohibitive. Thus I recommend sublingual tablets of 5 mg and 10 mg. This method allows a direct entrance to the blood stream via the sublingual, superficial venous plexus.”

PM: “And does it need to be taken every day?”

WP: “We do not really know if TRH follows a circadian rhythm. However, in some of my experiments I observed that TRH shows more pronounced effects when taken in the morning. I suggest cycles of TRH, 5 mg sublingual tablets for periods of one month, on and off. I also suggest some blood tests of glycemia and lipids regularly in order to ascertain its benefit. It is particularly rewarding for patients suffering from Metabolic Syndrome X.”

PM: “And for melatonin, how should one take melatonin?”

WP: “The real value of melatonin is its formulation which allows its night release between 1am and 3am. Many years ago I formulated melatonin with zinc and selenium; it is sold in Italy as Melatonin Zn Se® and by IAS as MZS™. After many scientific studies showing how melatonin can dramatically enhance zinc levels, whose concentration in blood notoriously decreases with aging, it was the combination of melatonin with zinc and then particular excipients- that also determine a night peak- that have created this true biological bomb! One must know that zinc is a basic mineral for synthesis of more than 200 enzymes. One tablet at bedtime between 9pm and 11pm is proper. The dosage of 3 mg is suggested, some may require less,

some may require more.”

PM: “And are there any particular times to take melatonin?”

WP: “One should try to emulate the night peak; this means that nature has determined that melatonin levels are highest between the hours of 1am and 3am. The MZS™ when taken between 9pm and 11pm releases the melatonin into the blood during the period of 1am and 3am, therefore inducing this same as nature response.”

PM: “I have to say, that we here at IAS like original products, after all they are the ones that achieved the original results in the clinical studies- the results that the copycats will allude to! I know that your MZS™ works well because I’ve seen it in patients time and again. Even on those who were going to give up on melatonin because they had seen no benefit with the generic they had been using; as is often said in pharmacology, the devil is in the detail!”

WP: “Exactly Phil. Many people probably do not realise that many of the clinical melatonin studies have used our brand. For example, a study published in the New York Academy of Sciences in 2005 showed that our MZS™ can reverse both wet and dry versions of macular degeneration- can you imagine! Yet how many people are aware of this fact?”

PM: “Quite true, my joke is that if you want to keep something a secret - publish it! (Ha ha) After all, who can read everything? This fact and the pressures on doctors’ time, means that many rely on the pharmaceutical rep to keep them appraised of developments. Accordingly, they only hear what the drug companies are selling. Still, let us not dwell on this negativity; please tell us a bit about your latest projects.”

WP: “Right now Phil, I am working on two new books. One will describe TRH in detail– this is because the effects of TRH are both important and impressive, yet hardly anyone is aware of this news today. The second book will have examples of my patients, their before-and-after results and what was done in-between. I am writing that one, not because of my ego, but because I am so often asked what

I do to achieve such good outcomes. Right now, I have 1000 patients, none of them have cancer or heart disease or dementia- can you imagine?”

PM: “Excellent news! In the meantime, by the time folks read this interview, we should have your eBook ‘melatonin, the key of life’ available for download.”

WP: “I hope that will be helpful to people. I would like to make a statement about why it is vital to talk directly to the public.”

PM: “Please go ahead.”

WP: “A revolution can only be started by the people themselves, from the ground-up. Once the public wake up to the current medical mafia and understand what is truly possible with alternatives, by which I mean non-orthodox, but nonetheless scientifically proven approaches; then the world of health will become a very different place.”

My biochemist friend and patient, Bob, phoned me recently. In an urgent voice, he requested an immediate appointment. I was a bit surprised because several times previously he wanted to use only his traditional physician, who was covered by his medical insurance. I made the appointment for the following afternoon, wondering why Bob suddenly wanted my help.

The next day, Bob arrived in my office, flustered and haggard from lack of sleep. He explained that several of his senior friends had passed away in recent months; one of them had fallen in the bathroom at night and was sent to the hospital with head trauma. Apparently, this poor fellow had fallen because of adverse reactions to prescription medications—an all-too-common occurrence among America’s seniors. Unfortunately, the same thing

happened to beloved comedian George Burns, who slipped and fell fatally in his bathtub at age 97 years. Also unfortunate is the fact that most medical doctors do not know how to prevent bathroom falls. Contrary to what many people believe, an expensive medical insurance plan will not save them from many of the medical and lifestyle problems encountered during aging or from the diseases of aging!

Only Anti-Aging Medicine Solves Many Problems of AgingBob was also at risk from falling in the bathroom at night, and he asked me for a remedy. I explained to him that the answer is simple and straightforward: First, hire a carpenter for one day and have him install around the toilet and bathtub/shower areas stainless-steel 3-inch thick railings, which are available at most hardware stores. He would then be able to grab hold of these railings when he feels faint or off balance. He may also use them as additional towel or wet-clothing racks. people who are at risk of falling in the bathroom.

Bob thanked me for my help and promised to report to me the results from my suggested lifestyle changes.

As you can see from the above case study, preventative and anti-aging medicine offers more advanced and cutting-edge solutions for common problems encountered during aging.

Lower High Blood Pressure NaturallyIn the foregoing study, Bob has high blood pressure (bp), and he consumes bp-lowering medications that cause him to become dizzy and disoriented and lose his sense of balance. This is a common tragedy for modern American medicine and there should be better solutions. There are better solutions, but many remain unknown to medical practitioners.

Upon gaining Bob’s confidence in the quality of my advice, I persuaded him that the underlying cause of his high bp is a loss of hormones as he aged. This phenomenon is especially true for those over age 50 years, but it even plagues some people much younger.

In Bob’s case, I encouraged him to take comprehensive blood and urine tests. These tests confirmed my visual observations: Bob was deficient in the hormones DHEA, cortisol, thyroid, growth hormone, and testosterone.

The first two—DHEA and cortisol—were visually apparent, as depicted in the following younger-to-older photos of GOP Speaker of the House John Boehner.

Observe that because of aging, Mr. Boehner’s adrenal glands produce lower and lower amounts of vital hormones such as DHEA, cortisol, and

aldosterone. These adverse effects are evidenced by dark circles around his sunken eyes. Mr. Boehner, my patient Bob, and many others would benefit from daily supplements of hydrocortisone and DHEA. These dark circles and sunken eyes will all but disappear when one consumes these corrective hormones combined with a healthier diet—in other words, no junk food. These combined remedies cause the dark-circled pigmentation to disappear and will push forward the eyes in their sockets as gains are made in laying down new and healthy collagen and elastin. This simple remedy results in true regenerative medicine as practiced by knowledgeable anti-aging physicians during the past decade. Indeed, my own dark eye circles disappeared after about three months of daily DHEA treatment. My sunken eyes pushed forward in their orbitals (sockets) after about a year of treatment!

Also, notice the jowls Mr. Boehner acquired as he grew older. This aging sign is often referred to as ‘myxedema.’ Historically, men hid it by growing long beards. Today, we can prevent it with daily doses of growth hormone (or a growth hormone releasing peptide like Releasing-PRO™) and a natural thyroid like Armour® over a treatment period of approximately three years. or on our genes. Upon arriving at this genetic level, these hormones assert command and control of key biochemical processes. These key hormones and their helper proteins are human growth hormone (hGH or its stimulator, Releasing-PRO™), IGF-1, and TRH (pineal gland proteins). These three dominate at the top of the heap in directing a symphony of cellular processes throughout the body. These critical hormones and their helper proteins slowly mold and sculpt our bodies to a more youthful appearance and help us maintain a quick, firm, and more resolute-thinking brain.

My colleagues and I have conducted studies where patients consumed Releasing-PRO™ four times daily. We found that IGF-1 doubles from approximately 70 to 160 (plus or minus 15 ng/ml blood). This 160 level of IGF-1 will direct and sculpt the body to a more youthful appearance, as depicted below.

ConclusionsMany of the problems of aging can be corrected by making constructive changes to our living quarters and by supplementing with key hormones and peptides. Hyaluronic acid (HA) has been called “Nature’s Moisturizer.” It is found throughout the body because it forms part of the structural tissue that holds tissues together.

HA is a fundamental component of the extracellular matrix; it occupies the areas between cells and almost half (7.5 grams) of HA within the body is located in the collagen of skin, acting as a moisture retaining gel. It is found in many tissues in the body including the heart, eyes (vitreous humour), joints (synovial fluid), tendons and ligaments. Scientific studies have shown that HA improves skin and tissue hydration; works as a free radical scavenger

and stimulates the production of collagen in the skin. HA also keeps skin elastic and protects joints from repeated stresses. The glycation theory of aging is a modern outgrowth of the venerable cross-linking theory of aging, first proposed by Johan Bjorksten in 1942.1,2 Bjorksten was a chemist from Finland, who earned his Ph.D. degree at the University of Wisconsin in 1937. He went to work for the Ditto Corporation of America, which was the ‘Xerox’ of the day.

Protein cross-linking as cause of aging

While at Ditto Corporation, one of the main prob-lems Bjorksten faced was to develop a way to in-crease the ability of the Hectograph films to with-stand summer temperatures, exposure to water, mechanical stretching and friction, and to prevent their drying out and cracking. The ‘aging’ of hecto-

graph films was due to the cross-linking of protein molecules, which Bjorksten believed was also the cause of aging in humans. This was confirmed in a crude way by comparing the similarity between the shape of the mortality curve for humans (Fig. 1) with

a viscosity curve of gelatin during a cross-linking reaction (Fig. 2).3,4 The crosslinkage theory stated that the principal cause of aging was the linking together of two or more large molecules (macromol-ecules), which progressively linked with other mole-cules, impairing the functioning of cells, tissues, and organs, resulting in the aging of the organism.

Search for cross-linkage inhibitorsBjorksten focused on a potential means of prevent-ing and breaking up crosslinkages by using chelating agents that attach to metals within the body, enabling them to be excreted. Ethylene-diamine-tetra-acetic acid (EDTA) is a synthetic amino acid that removes metal-based crosslinkages by chelation, thereby “depolymerizing the gerogenic aggregates.” 5 Chelat-ing agents in current clinical practice besides EDTA (approved for use in the treatment of lead poisoning) are deferoxamine (Desferal® for acute iron intoxica-tion), and DMSA (used to treat lead and mercury intoxication). Physician members of the American College for Advancement in Medicine (ACAM) are proponents of intravenous chelation therapy with EDTA as a treatment for many chronic degenerative diseases, like atherosclerosis, hypertension, diabetes, and Alzheimer’s disease.6 Chelation with oral EDTA chelation is also becoming more widely used.[Ed. - EDTA is available in BCI®, Beyond Clean 2® and Beyond Fiber®]Other natural chelators include garlic, 7 chlorella, 8 lactic acid, citric acid, and malic acid. Bjorksten demonstrated that lithium was also an effective alu-minum chelator and crosslinkage inhibitor, stating that “lithium continues to be the most effective elec-trolyte for aluminum detachment.” 9[Ed. – Lithium orotate is available from IAS]Chelators as life-extending substancesA number of studies confirm that chelating agents, particularly, EDTA — may have life-extending prop-erties. Scientists demonstrated the life-extending effects of EDTA on lowly rotifers (small multi-celled

animals found in freshwater lakes and ponds).10-14 In the Soviet Union in the 1970s, Dr. T.L. Dubina performed a series of studies with EDTA on the life span of rats. In most of the studies, the mean life span of female rats treated with EDTA was increased by nearly 50%, and in one study the maximum lifespan increased 18-25% over the control animals. 15Aluminum—a powerful cross-linkerAluminum is a highly reactive metal that occurs freely in nature, and comprises over 8% of the earth’s crust. Although it also occurs very widely in human nutrition, (most people ingest 10-100 mg of alu-minum daily), it does not play a role in any known metabolic process. Aluminum is highly toxic, even in extremely small amounts, and although the neu-rotoxic effects of aluminum have been known for many years,16 scientists have recently shown that aluminum accumulation may contribute to Parkin-son’s disease17 Down’s syndrome18 and Alzheimer’s disease.19Aluminum uptake increases with ageIn 1955, during a talk on gelatin crosslinkages and aging at the Gerontological Society in Baltimore, Bjorksten discussed the relationship of aluminum to crosslinking. One of the attendees, Prof. H.H. Zinsser of Columbia University, was so interested in the concept that he and Bjorksten began a fruitful collaboration that was to last for seven years. Using spectrographic analysis, they examined the alumi-num content of 84 persons, ranging in age from 10 to 90 years (Fig. 3).

They found peak levels at age 40-50, followed by a drop and then leveling off, indicating that those whose aluminum accumulation peaked in middle age most likely did not survive the next ten years. 20 Zinsser’s data were confirmed independently by scientists who demonstrated a progressive increase of aluminum concentrations in the brain (Fig. 4).21

Bjorksten concluded that the progressive increases of aluminum with age were of great importance, and believed that the aluminum mechanism alone would limit human lifespan to 110-120 years of age even if no other cause of death intervened.22

Preventing aluminum accumulation and crosslinking

Bjorksten and his staff evaluated the ability of chelat-ing agents to remove aluminum-containing stains from the aortas of 5-6 month-old hogs (Fig. 5).23 it can be seen that EDTA was the most effective.

Lactic acid, similar to blood concentrations generat-ed by exercise were moderately effective. Of interest was the fact that 0.5 % procaine—the active ingredi-ent of Gerovital® (or GH3, the Romanian antiaging drug)—was also moderately effective in reducing the aluminum. This raises the question whether the metabolites of GH3—DMAE and PABA—might also have some effect in this regard.24 In fact, Drs. Imre Zs.-Nagy and Katalin Nagy demonstrated that both dimethylaminoethanol (DMAE) and centrophenox-ine (CPH) are indeed able to diminish the extent of crosslinking in old rats.25

Bjorksten’s team also evaluated the ability of various concentrations of lithium to remove tightly bound aluminum from tanned leather baseball covers.26, 27 Using a 0.05M concentration of lithium citrate, they completely demetalized the leather in approx-imately three months. Lithium is used clinically to treat manic depressive illness (MDI). Recent studies indicate that it may offer benefit in the prevention and treatment of Alzheimer’s disease, as well. 28 Perhaps long-term treatments with low doses of lith-ium may be an effective way to displace crosslinked/protein bound aluminum in animals and humans. Lithium orotate as a dietary supplement is the safest, most effective form of lithium available. Bjorksten spent his entire career searching for the cause and cure of crosslinks, which he believed would result in the reversal of aging and maintenance of prolonged youth. He believed that one cause of the crosslinks was heavy metal poisoning, (especially aluminum), which could be prevented and reversed with chela-tion therapy. He also identified the loss of fluidity of cell membranes as a contributing factor, and supported the use of membrane fluidizers such as centrophenoxine, phosphatidylserine, choline and lecithin.

Advanced Glycation End Products of Aging (AGEs)

In 1965, Dr. H.B. Bensusan proposed that a process known as the Maillard reaction, (the non-enzymatic

chemical reactions between proteins and carbohy-drates that cause cooked foods to turn brown) was what caused all long-lived proteins in the body to turn brown and become fluorescent, (under UV light), become more cross-linked, less soluble, less elastic, and less digestible by enzymes. In 1985, Monnier, Kohn and Cerami provided further details of the role of the Maillard reaction,1 and further developed the idea that the Maillard reaction causes premature aging and degenerative diseases such as diabetes and heart disease.29 In this regard, many scientists think the human body may be viewed as a ‘low temperature oven’ with a relatively long—ap-proximately 75 year —‘cooking cycle.’30 The Mail-lard reaction involves a chemical reaction between a sugar with a protein, into a complex known as a Schiff base. With continued exposure to the sugar, the Schiff base undergoes a ‘rearrangement’ known as non-enzymatic glycosylation that results in a more stable, less reversible substance, known as an Ama-dori product. In the human body, this process reach-es equilibrium over several weeks (Fig. 6).31

The Amadori product further degrades irreversibly into a number of highly reactive carbonyl (C=O) compounds called Advanced Glycation End prod-ucts, designated by the acronym AGE.32 AGE is a clever pun which reflects the proposed relationship of these reactive substances to aging and age-relat-ed diseases. AGEs can further react with other fats, proteins and nucleic acids to form largely indissol-uble crosslinks. The age-related accumulation of these AGE products has been demonstrated in many tissues of the body (Fig. 7).33

During long-term hyperglycemia (elevated blood sugar), as in diabetics, glycation and AGE formation may be quadrupled! This explains why diabetics suffer the premature onset of a wide range of age-re-lated complications including cataracts, retinopathy, neuropathy, nephropathy, atherosclerosis and osteo-porosis.34, 35

Crosslinkage Theory Gets New Life

Bjorksten was a talented petroleum chemist. Had he been a food chemist instead, he may have appre-ciated this link between the Maillard Reaction and crosslinking much earlier, and made even greater progress in developing preventive and therapeutic approaches to crosslinkage-induced aging. Through their insightful work in understanding this pro-cess, scientists like Brownlee, Cerami and Monnier provided renewed impetus and a ‘rebirth’ for the crosslinkage theory. Unfortunately, they did this with little attribution to Bjorksten, who had doggedly pur-sued this approach to aging for over 50 years.

AGE Breakers (and inhibitors)

There are a number of substances that have been used to delay and reverse glycation-induced crosslinkages (AGEs). Khalifah and his colleagues proposed a schematic of the formation of AGEs, and illustrates a number of specific therapeutic targets (Fig.8).31

Metformin (Glucophage®): Metformin is an anti-di-abetic biguanide that was derived from the herb, Goat’s rue (Galega officinalis). Biguanide drugs were recognized by Prof. Vladimir Dilman as early as the mid-1970s as the most effective antiaging drugs in existence. Metformin is an insulin receptor sensi-tizer, capable primarily of lowering blood sugar and insulin. Dilman also demonstrated that biguanides restored cortisol receptor sensitivity. Metformin has many other beneficial properties, including opti-mizing lipid profile, reducing body fat, maintaining levels of growth hormone, stimulating immunity, and extending the maximum lifespan of experimen-tal animals.36

Aminoguanidine (Pimegedine): Aminoguanidine is a substance that has been known for over 100 years. It is structurally very similar to guanidine, the active ingredient in the herb, Goat’s rue (the herbal proto-type for metformin). Aminoguanidine has aroused a great deal of interest in the last twenty years, due to its demonstrated ability to block the formation of AGEs and AGE-induced crosslinkages in both animal and human clinical studies. Aminoguanidine inhibits AGE formation, preventing AGE-induced crosslinks in collagen and other tissues. Fortunately,

aminoguanidine does not interfere with the forma-tion of normal collagen crosslinks, which are re-quired for structural integrity. Another mechanism by which aminoguanidine is believed to act is by enhancing the action of nitric oxide (the same mech-anism by which Viagra functions).37, 38 Aminogua-nidine also reduces the formation of lipofuscin (age pigment) and prevents or reduces cataracts, ather-osclerosis, diabetic retinopathy, nephropathy and neuropathy (Fig. 9). 30, 39-41

In a study of diabetic patients, after four weeks of therapy with aminoguanidine, LDL cholesterol decreased almost 30%, and total cholesterol and triglycerides both decreased almost 20%. Hemoglo-bin-AGE levels, a circulating marker of the degree of glycosylation, also decreased dramatically (13.8 U/mg Hb at the beginning of therapy, to 10.0 U/mg Hb after only four weeks).42 Aminoguanidine is very safe, as indicated by short-term human studies which used the astronomical dose of 1200 mg daily.43 (This is in comparison with a usual human dose of 100-300 mg daily). The dose required to cause death in half the animals (mice) to which it was administered (Lethal Dose 50 [LD50]) was 1800 mg/kg.37 That would be equivalent to a human dose of almost 300 grams!

Pyridoxal-5-Phosphate (P5P): P5P, the active form of vitamin B6, has been found to significantly re-duce the non-enzymatic glycosylation (formation of AGEs) of bovine serum albumin (BSA) with radio-active-labeled sugar. Of the substances tested, P5P

was exceeded only by aminoguanidine in its ability to inhibit AGE formation (Fig. 10). Combining P5P with metformin or aminoguanidine may enhance their AGE-inhibiting actions even more.44

Vitamin B1 (Thiamine) and Benfotiamine: In their book, Life Extension, Durk Pearson and Sandy Shaw reported that thiamine was an effective crosslink inhibitor.45 They were at that time consuming two grams of thiamine each day in their personal antiag-ing regimens. Thiamine may ultimately also prove to be an effective crosslinkage breaker as well as inhib-itor. Another thiamine derivative is the fat-soluble Benfotiamine. Benfotiamine is very helpful in the treatment of diabetic neuropathy at a dose of 300 mg twice daily.

Carnosine: Dr. Alan Hipkiss of the Division of Bio-molecular Sciences, King’s College London, reviewed the antiaging effects of carnosine and aminoguan-idine.46 Dr. Hipkiss believes that one of the major mechanisms of the antiaging effects of carnosine is its powerful effects as a crosslink inhibitor and breaker, and that the use of these substances might help to control age-related molecular dysfunction. Carnosine has been used in doses generally ranging from 500-2,000 mg/day.

Sorry for this long answer to your short question—but I believe that Dr. Bjorksten’s often overlooked contributions to the glycation/crosslinking theory need to be remembered and reemphasized.

(technically it is the closest legal molecule to GHB). Its approved use in Europe is to help with epilepsy, but like GHB, GABOB has been shown to improve growth hormone release significantly when taken before bedtime. GABOB does not have the ‘sleep inducing’ effects of GHB, but it can cause some sedation if taken in high doses.

Glutathione is a potent antioxidant that helps to prevent damage to important cellular components that can be caused by reactive oxygen species such as free radicals and peroxides. Glutathione is principally synthesized inside the body and is relatively poorly obtained via the diet. Hydergine is an extract of a fungus that grows on rye called ergoloid mesylate. Its approved uses have been to aid persons with memory disturbances, Alzheimer’s disease and even for persons who have suffered from electric shock, drowning or suffocation. This is because hydergine stabilises the levels of oxygen in the brain. It has been shown to improve oxygen levels when they are too low and even decrease levels when they are too high. The result of this optimised oxygen availability in the brain is an ability to do more mental work without becoming tired. Accordingly, hydergine can be a good tool to battle through boring paperwork or a long drive home in the dark. A fascinating animal experiment showed a special ability for hydergine to improve mitochondrial condition. The mitochondria are vital organelles found in every cell of our body responsible for the generation of ATP, the universal energy molecule. Unfortunately mitochondrial efficiency declines as we age. However, an Italian study highlighted that old animal mitochondria can be improved with hydergine supplementation.

Hydergine also stimulates the growth of dendrite nerve fibers. These connective strands are known to decline in aging. This is another antiaging feature of hydergine. Some have even linked the density and numbers of dendrite fibers to the level of I.Q. In summary, hydergine helps to protect against conditions of hypoxia and peroxidation, important factors to guard against a stroke and generalised brain injuries. Hydergine can extend the boundaries of mental workload and energy, fighting against boredom and may even provide some groundwork for raising the levels of intelligence. Melatonin (MZS™)

Melatonin is one of nature’s remarkable substances. It is made in response to darkness by the pineal gland. Melatonin’s benefits to help overcome jet lag and shift work have become quite well known and indeed it is useful for this purpose since adequate melatonin availability enables our bodies to be correctly in tune with our circadian rhythms. These circadian rhythms dictate our hormonal cyclicity and this cyclicity ultimately determine our aging and our level of immunity. Unfortunately melatonin secretions decline with aging. Professor Pierpaoli has published widely on melatonin, principally through the New York Academy of Science and at the meetings of the Stromboli conferences on aging and cancer- which IAS is delighted to support. What is important to know about melatonin is that Professor Pierpaoli advocates a 3mg dose to (his words) “put the pineal to sleep.” In addition, it is also vital to use a melatonin that produces a night peak of melatonin that is the same as the peak produced by the pineal itself. This he has achieved using specific excipients in his own formula ‘MZS™’ (which also contains the additional synergistic ingredients of zinc and selenium).

The results of low dose naltrexone, (widely known as LDN) therapy have been remarkable. Originally, the use of naltrexone was in the management of dependence on opioid and alcohol. But it was Dr. Bernard Bihari in the 1980’s who first used low dose naltrexone for the treatment of HIV and then for a number of other autoimmune diseases such as multiple sclerosis, Crohn’s disease and many immunological related diseases. Many MS patients have reported an improved quality of life. A leading study has established that by maintaining regular nightly LDN, less than 1% of the MS sufferers experience fresh MS attacks. IAS supplies the LDN dose of 4.5 mg.

population could be hypothyroid. Thyroid supplements come in two forms; natural supplements and synthetic supplements. It is almost always better to use a whole-natural thyroid extract because the synthetic versions usually only comprise one of the thyroid hormones, (such as, T3 or T4), whereas, whole-natural thyroids cover a fuller spectrum of thyroid hormones (including T1, T2, T3 and T4). Thyroid (Armour®; Synthroid®)

TRH is a hypothalamic hormone (technically a tri-peptide) whose real uses and benefits are just coming to the fore, primarily after research conducted by Dr. Walter Pierpaoli, the physician and scientist who exposed the benefits of melatonin to the world. Dr. Pierpaoli’s research suggests that TRH is a ‘master’ hormone responsible not just for helping to adjust thyroid hormones (its principle recognised role), but in helping to recorrect many imbalances throughout the body. For example in his animal experiments TRH supplements have encouraged old animals to:

• Re-establish spermogenesis, (reversing the age-related dysfunction of their testes).

• Correct kidney dysfunction, (a significant finding for renal issues).

• Correct pancreatic dysfunction, (a significant finding for diabetic issues).

• Improves many lipid profiles and aids weight loss when used over a few months.

TRH (Abaris™)

TA65 contains a highly purified specialized extract of the Chinese herb astragalus. What makes this supplement totally unique is that it is the first natural substance that has been proven to act as a telomerase activator and to extend telomeres in humans (in-vivo). Telomeres are the strands on the ends of chromosomes that shorter each time the cell divides. This is a significant part of the destructive process of aging. Animals with longer telomeres enjoy a more productive and healthier older age and appear to live longer. TA65 production is carefully controlled by TA Sciences, who purchased the technology from the Geron Corporation. Whilst other substances have claimed to delay the shortening of telomeres, only TA65 has been clinically proven (and published) to actually lengthen short telomeres in humans. If you want the cutting-edge of antiaging supplementation and the best possible chances for longer telomeres, then TA65 is currently your best (and indeed only) clinically proven choice.

it is the first 29 amino acids of GH, the part responsible for activation. This is what makes sermorelin stand head and shoulders over all previous agonists of GH. Sermorelin is GH site-specific and therefore very effective. What is more, sermorelin has a negative feedback loop (unlike GH itself), so the issues of overdosing and safety associated with injections of GH are minimized/ completely avoided. Sermorelin is stable at room temperature and efficacious when used sublingually. These are additional factors that favor sermorelin as the next generation of GH activators. Vasopressin is a porcine extracted source for this pituitary hormone which has a number of interesting roles.

Its approved medical use is to prevent frequent urination in conditions such as diabetes insipidus. For example, men with prostate conditions can benefit by less trips in the night to urinate.

But vasopressin is also responsible for the laying down of memories in the hippocampus and it has been used to treat amnesia. In particular, it is cited by many for memory imprinting, this is the learning of new material before it is experienced. Therefore taking 1 or 2 sprays into each nostril 15-minutes before an exam, meeting or lecture etc., can induce a better detailed recall of events later.
