Resume of Ph.D. Work
In the Ph.D programme I had studied various aspects of Salmonella
contamination in fish and crustaceans such as their prevalence, antibiotic resistance
pattern and survival strategies in natural waters such as fresh water and sea water.
More than 1000 samples were analysed for the prevalence of Salmonella. Confirmed
strains were serotyped at National Salmonella and Escherichia Centre, Central
Research Institute, Kasauli, Himachal Pradesh, India. All the serotypes were subjected
to antibiotic sensitivity sensing in order to find out the level of resistance acquired by
them and multiple antibiotic resistance (MAR) indexing in order to assess the risk
associated with these strains. Important strains were tested for their survival strategies
in natural waters such as fresh water and sea water. The ability of the predominant
serotypes to adjust to the changes in salinity and temperature were also studied using
microcosms. The major findings were as follows:
Fourteen percentage of fishes and 17 percentage of crustaceans were
contaminated with Salmonella.
Serotyping revealed the presence of Salmonella weltevreden, S. mgulani, S.
typhi, S. typhimurium, and S. senftenberg.
S. senftenberg was encountered in only crustaceans while the other serotypes
were encountered in both fishes and crustaceans.
Antibiotic sensitivity testing revealed high degree of multiple antibiotic
resistance among the strains.
Multiple antibiotic reisistance (MAR) indexing of the strains showed that all
the strains were originated from high risk sources of contamination such as
human, poultry and dairy cattle, where antibiotics are often used.
Survival studies using microcosms of freshwater and seawter was carried out
to find out the role of various physico-chemical and biological parameters in
the self-purificationof natural waters when contaminated with Salmonella. The
results revealed biological factors such as predatioin, antibiosis, competition
and protozoan bacteriovory were the most important factors in neutralising the
Salmonella contaminants in natural waters. Physical factors such as uv and
visible light exerted only a secondary effect, while the chemical composition
of natural waters did not exert much negative impact.
Experiments to reveal the ability of selected strains of Salmonella to tolerate
changes in salinity and temperature showed that they are well suited to
estuarine conditions.
The findings of Ph.D were published in leading international journals
such as Food Microbiology (Academic Press Ltd., UK) and Letters in Applied
Microbiology (Society for Applied Bacteriology, UK).
During Ph.D I had also done considerable works on the bacterium
Escherichia coli and the finding were published in international journals of repute
such as World Journal of Microbiology and Biotechnology (Rapid Communication of
Oxford Ltd. UK) and Zentralblatt fur Hygiene und Umweltzmedizin (Gustav Fisher
Verlag, Stuttgart and New York).
