How to choose an SPE product1.Characterize the sample

Factors such as the analyte’s polarity relative to thematrix, the presence of charged functional groups, solubility, molecular weight, etc., determine how strongthe analyte is retained by the packed bed.

2.Select a retention strategyThere are two basic methods for sample treatment:a. Select the packing bed to retain the desired analyte.

The contaminants are washed off and the desired analyte is then eluted for analysis.

b. Select the packing bed to retain the contaminantsand the desired analyte passes directly through.

3.Select the proper packing typeSelect the proper packing type for the cleanest extract with the highest recovery. a. Reversed-phase packings are hydrophobic, silica-based

materials that retain moderately polar to non-polarcompounds from a polar matrix while washing off polarinterferences. Or you can retain non-polar contaminantswhile the polar compounds pass through unretained.

b. Normal-phase packings are hydrophilic, silica-basedmaterials that retain polar compounds from a non-polar matrix while washing off non-polar interferences.Or you can retain polar contaminants while non-polarcompounds pass through unretained.

c. Ion-exchange resins retain charged compounds orremove ionic interferences.

4.Optimize conditions for best resultsSelect proper bed size and suitable conditioning, washand elution solvents.a. Poor sample recovery often occurs when the packed

bed dimensions are not optimized. An excessive bedweight results in incomplete elution while an insuf�cientbed weight results in incomplete retention.

b. Consider the solvent strength relative to the packing material. The �nal conditioning solvent should be weak so it doesn’t act as an eluting solvent. Buffers should be used to control ionization of potentiallycharged compounds.

c. Wash solvents should remove weakly retained interferences without being strong enough to elutethe analyte.

d. Elution solvents should be strong enough to completely elute an analyte in a small volume (1–2mL).

Choosing the Right Cartridges

Cartridge Size —Sample Capacity

Sizes: 1 mL, 3 mL, 6 mL, 60 mL(Specialty sizes available upon request)The SPE Cartidge determines the volume of the sample solution, which can be loaded onto the sorbent bed in a single step.

Resin Bed Mass—Analyte Capacity

Sizes: 10 mg, 30 mg, 60 mg, 200 mg, 1 g (Specialty bed weights available upon request)The resin bed mass determines the maximum amount of analyte retained by the sorbent.

Tips: Choosing the proper bed mass reduces leaching of sample components

Tips for Choosing Proper Resin Mass

Polymeric Sorbents—polymeric resins provide largersurface area than silica-based resins, offering higher capacity on the order of 10-15% of the resin mass. Thisallows for the use of smaller bed volumes and improved detection limts.

Ion Exchange Sorbents—the number of charged sites on the resin available to interact with the analyte determines capacity.Tip: Choosing a slightly larger resin mass is advisable, since mostSPE samples are loaded with contaminants and interferences

Pore Size—the mean diameter of the pores on the sorbent particle surface

The analyte molecule in an SPE method should t easily through the pores of the sorbent (i.e. for largemolecules, a sorbent particle with extra large pores should be selected).

Resin Bed Volume—Interstitial and Pore Volume

The resin bed volume controls the minimum solventvolume in an SPE method. To successfully condition,wash and elute from the cartridge typically requiressolvent volumes 4 to 8 times that of the sorbent bed.Using less than 4 to 8 sorbent bed volumes contributes toincomplete solvation and low, inconsistent recoveries.

Tips: Polymeric SPE products usually require 200-250 uL per 100mg of sorbent.

MicroLute™ ManifoldThe MicroLute™ vacuum manifold from Porvair Sciences isprecision machined from crystal clear acrylic (top plate) andacetal polymer (plenum chamber). The acrylic top plate allowsvisual access to the plenum chamber for checking progress ofthe separation process.

• Designed to take most filter plates manufactured to the SBS/ANSI specification with long drip directors

Universal Robotic ManifoldBased on the successful MicroLute™ Acrylic Manifold, thenew Universal Robotic Manifold is designed to be easilyassembled and disassembled by robotic manipulators,enabling productive automation of SPE or DNA clean-up pro-cedures. Vacuum Manifolds

Vacuum manifolds for SPE sample preparation, filtration, andelusion are available in 12, 16, and 24 port configurations fromCiro Manufacturing Corp. These manifolds permit consistentextraction and filtration results. Multiple sample processingwith these manifolds simplifies procedures and savestime.The manifolds consist of a clear glass chamber and lid towhich a vacuum is applied to draw a sample through an SPEcolumn, cartridge, or disk.Adjustable racks placed in the glass vacuum chamber willaccommodate a variety of sample collection vessels,including test tubes, autosampler vials, volumetric flasks, andErlenmeyer flasks. Eluants are deposited directly into thecollection vessel of choice via polypropylene, optionalstainless steel, or Teflon needles.Drying attachments for the 12 and 24 port manifolds will directa flow of air or nitrogen into the collection vessels to dryeluants prior to further analysis. Drying attachments can alsobe connected, via adapters, to SPE columns or cartridges inorder to dry the column or cartridge prior to final elusion.Optional disposable solvent resistant polypropylene liners areavailable for the twelve port manifolds.These waste linersgreatly simplify sample preparation, solvent disposal, andclean-up.

Drying attachmentsDrying Attachments are available forthe 12 and 24 port manifolds, whichwill direct the flow of air or nitrogen intothe collection vessels to concentrateeluants, prior to further analysis. Dryingattachments can also be connecteddirectly to columns, cartridges, or disks,via adapters to permit drying sorbent beds, prior to finalelution.

Teflon needles and control valvesDisposable Teflon needles andTeflon needles with flow controlvalves are designed to fit throughthe manifold lid via the luerfitting.These needles deliver theeluant directly from the SPE

column or cartridge into the collection vessel in the vacuumchamber.These needles, when used in conjunction with Tefloncolumns and Teflon frits ensure zero extractables from the col-umn, frits, and fluid path. This combination is especially use-ful for critical sample analysis, such as environmental sam-ples.

The liner is designed to contain all liquids used in SPEsample preparation.To use the liner, remove the manifold lidand take out the rack and shelf set.

Place the disposable liner into the glass vacuum chamber,and replace the manifold lid. Proceed with all conditioning and sample preparation steps.Just prior to final elution, the liner, containing the wastesolvents, is removed from the vacuum chamber. There aresmall handles at each end of the waste liner to facilitate itsremoval. Replace the lid, and proceed with the final elution ofthe analyte.Waste solvents should be properly discarded from the liner.The liner can be cleaned and re-used a number of times, priorto discarding.

The Evaporex® has two needle assemblies, each consists of 96 individual stainless steel needles that evenly distribute heated gas into the microplates from both the top and bottom. The upper gas flow path is sealed away from the heating elements in a stainless steel path in order to prevent sample contamination.

Biological samples commonly contain proteins that interferewith downstream applications. The P3 plate uses the CRASHmethod, in which the protein is denatured with acetronitr ileand the flocculant filtered out, allowing 96 samples to behandled at one time. The Protein Precipitation Plate, P3, isbased on the industry standard MicroLute™ format, but wi th-out the chromatographic sorbent. Protein ʻcrashesʼ out ofsolution and precipitates directly in each well when acetonitr ileis added, thus solving all common problems associated withthe CRASH technique of protein clean-up. The novel dual f rit,hydrophobically treated matrix means that there is no ʻwettingoutʼ and leakage of the sample through the plate before theapplication of vacuum.

P3 is now commonly used as the protein precipitation plate ofchoice in many major pharmaceutical companies.Optional drain cap and top cap mats are available for thosewishing to use vortex sample mixing.

*The Protein MicroLute High Efficiency plate is exactly the same as the P3 butwith the frits untreated. This plate is used when sample and acetronitrile arepre-mixed before being pipetted into the plate.

We Offer Affordable, Quality Syringe Filters

Housing Polypropylene housings are highly solvent resistant and exhibit low extractables. They are ideal for filtering samples orsolvents used in chromatography.

Size4mm syringe filters are useful for filtration up to 1mL of low par-ticulate level chromatography of biological samples.17mm syringe filters have a filtration area of 1.3cm2 and a hold-up volume of less than 30uL with an air purge.Ideal for samples up to 12mL.30mm syringe filters have a filtration area of 4.9cm2 and offer thefastest sample filtration.

MembraneThe cellulose acetate and polysulfone membranes are all lowprotein binding, which makes them well suited for filtration ofbiological samples.PTFE membrane in a PP housing can be used to filter solventsand strong acids.Although PTFE is inherently hydrophobic, it can be madehydrophilic by pre-wetting the membrane with methanol andthen rinsing with water.Nylon or polypropylene are ideal for filtration of chromatographysamples.Polypropylene membranes exhibit near zero extractables, arevery resistant to solvents and most chemicals and would be thefirst choice for filtration of chromatography samples.

TCLP

XENOSEP TCLP Filters Xenosep offers two types of TCLP filters suitable for use inEPA Method 1311 depending on the analytes of interest. Bothtypes of TCLP filters are binder-free, borosilicate glassmicrofiber filters with an effective pore size of 0.7 +/- .01μm asspecified in the method.

Volatiles

Use Xenosep Standard-Certified TCLP Filters. These filtersare tested for several important parameters and are certifiedto meet the specification above.

Metals or non-volatiles

Use Xenosep acid-washed TCLP filters. These filters arepre-washed in 1N HNO3 acid and then rinsed with deionizedwater according to EPA Method 1311 to save you time, reducewaste and provide low background blank values. A certificateof analysis demonstrating background TCLP metal values isalso included in each box.

Why are 40 mg/L PAR comparisons important?

EPA Method 1664A requires the PAR concentration as follows:“The weight must be 40 ± 1 mg” (Section 7.10.3 NOTE) and atleast this amount are required for the QCS, OPR, MS, MSD,and IPR samples. (Sections 9.8, 9.7, 9.6.1, 9.3.1, 9.2.2, 7.11and 7.10) Although certain method modifications are allowed,a change in the PAR concentration is not expressly permittedand “shall be considered a major modification subject to appli-cation and approval of alternate test procedures under CFR136.4 and 136.5.” (Sections 1.7 and 1.8)
