Introduction

Many thanks for your interest in our range of UV/VIS/NIR cuvettes and associated products. We hope that the clear layout and classification provide all the information you need in order to find the right product. We’d be happy to receive your suggestions for improvements - please let us know and we’ll try to incorporate them in future versions. The first few pages provide basic information on choosing the right product and how to take care of it. The rest of the catalogue is devoted to the products: a table with the essential technical data and a photo or drawing or both. That’s followed bv a list showing all available configurations of that particular cuvette type with pricing.

We offer two product ranges – here’s why:

Since 1958 NSG Precision Cells, Inc. stood for quality products. In 2014 they rebranded to FireflySci, Inc. At the same time, the Brooklyn based company made a lot of structural changes resulting in much shorter delivery times on the majority of products.

The majority of our customers are in Academia where cost plays a primary role. Based on our similarly named online purchasing platform, the Lab-Club product range evolved – not as comprehensive as FireflySci, but in many case considerably less expensive. In a nutshell, we offer quality products to suit all budgets.

It’s easy to see whether there’s a Lab-Club alternative… The FireflySci part numbers begin with FF, the Lab-Club numbers start with LC. Also, the description tables for the Lab-Club products are in green. Certain cuvette types are available in so many configurations that the list of Lab-Club products may be on the next page. Please note that all the technical data tables are based on the FireflySci products – the data for the Lab-Club products are almost identical. In the very few cases where only Lab-Club products are available, the tables are in green. If, despite the vast range of products, you don’t find exactly what you’re looking for, please let us know. In most cases we are able to quote for a bespoke product.

By the way: cuvette or cell? Both terms are commonly used – we don’t differentiate between the two.

Choosing the right cuvette

1. Absorption or fluorescence measurement – which cuvette do I need?

If you’re using a spectrophotometer, you’ll need a cell with two polished (optically transparent) windows that are parallel to each other and located on opposite sides of the cell. In this case, the light beam enters the cell through the front window and leaves it through the back one in a straight line. For fluorescence measurements the cell needs three or four polished windows.

2. What if I need to measure both, absorption AND fluorescence?

Due to the number of optically polished windows a fluorimeter cell will work in a spectrophotometer - the other way round will not work.

3. Which material should I choose?

Technical data for our optical glass, UV and IR quartz are shown in the section Materials and Transmission Characteristics. In addition to the required wavelength range the price may also play a role. For example, if you only measure in the visible range then an optical glass cell is sufficient. If, however, you occasionally also measure in the IR range, then we recommend an IR quartz cell as it covers both areas.

4. What lightpath do I need?

The lightpath is the distance that the light beam travels between entering and exiting the cuvette chamber. We offer cells ranging from 0.01 to 200 mm. The deciding factor is the amount of sample available. If it’s relatively unlimited then a cell with a larger lightpath is easier to fill and handle. 10 mm cells have become the “industry standard”. However, if you only have a very limited amount of sample, for example DNA/RNA, then you should choose a cell with a correspondingly short lightpath.

5. What volume should the cell have?

In general, the volume is dependent on the lightpath – the longer the lightpath, the larger the volume. However, we also offer low-volume cells with a lightpath from 1 to 100 mm. If you have to work with a predetermined lightpath but only have a limited amount of sample, you’ll still find a suitable cell here. We offer the following cell types:

macro • semi-micro • micro • sub-micro • ultra-micro (Micro Focus Cell – for DNA samples)

6. Rectangular or circular?

90% of all cuvettes sold are rectangular. But if your instrument has a circular holder, for example a polarimeter, we also offer a wide range of circular (cylindrical) cells.

7. What type of cover should I choose?

The simplest cover is the PTFE type that sits loosely on the cuvette and stops contaminants entering it from above. These lids are NOT airtight.

The PTFE stopper offers air-tightness and allows the contents of the cell to be mixed by shaking or turning it upside down.

Screw caps provide the best seal and, hence, are also suitable for anaerobic work.

Graded seal (quartz-to-glass) tubes – please see question 8.

8. What if I need to modify or enhance the cell by adding a further piece of glass apparatus?

In this case you’ll need a cuvette with a graded seal tube. We recommend using Pyrex® for the part to be

connected. Your glass blower can offer further information on the suitability of other types of glass.


A to Z – cuvette terminology explained

Borosilicate glass

Contains large amount of boron trioxide and silicon dioxide. It has a very low thermal expansion coefficient and is, therefore, resistant to thermal shock.

Circular dichroism

Measures differences in the absorption of left-handed polarized light versus right-handed polarized light which arise due to structural asymmetry.

Coefficient of thermal expansion

Refers to how an object’s size is affected by changes in temperature. Specifically, it measures the fractional change in size per degree change in temperature at a constant pressure. Although several types of coefficients have been developed, for example volumetric, area and linear, the one used depends on the particular application and on which dimensions are considered important. For solids (cuvettes) we are primarily concerned with changes to lengths or areas.

Colorimetry

Is a method for analysing, quantifying and describing colour perception. Although close to spectrometry, colorimetry is different in that it emphasises reducing spectra to the physical correlates of colour perception. These are commonly enshrined in the CIE 1931 XYZ colour space values.

Demountable cells

Comprise two or more glass or quartz components that can be taken apart (demounted) and reassembled. These cuvettes are easy to clean and have a very low volume. So when sample size is critical, this type of cell is very convenient for retrieving your sample.

Fused Silica Quartz

Has high thermal shock resistance and is extremely resistant to highly-concentrated acids, with the exception of hydrofluoric acid which has a negative effect on the material even at low concentrations.

Fusing

The FireflySci cuvettes are manufacturer by fusing – the individual glass or quartz components are melted together (fused) at extremely high temperatures. Glass frit is not used in the production of their cuvettes.

Graded Seal

In order to attach glass apparatus to a quartz cuvette or to modify it in any way it’s necessary to have a robust and reliable transition from the one material to the other. Graded Seals have various “middle glasses” with different coefficients of thermal expansion fused into them. In this way, the thermal shock initiated when the glass blower attaches the modification to the glass end of the tube is cushioned and the quartz cell is protected from damage.

Lightpath

Also known as pathlength, this is the distance between the inside of the optically polished windows of a cuvette. It is measured in mm, the FireflySci products range between 0.01 mm and 200 mm depending on cell type.


Matching

Means that the cuvettes are manufactured from materials that have transmission properties within 1% of each other. Accordingly, only the absorption directly through the lightpath of the cuvette is tested – other parameters used in fluorimetry are not addressed. At the point at which all parameters are sustained to a high degree the cuvette is “matched” by the fact that there is little distinction in any of the cells of the same physical configuration, lightpath and material. With more modern machines and stringently controlled manufacturing processes, Matching is not as important nowadays. FireflySci cells are manufactured within extremely tight tolerances that ensure superior matching.

Transmission

Describes how well light passes through the optically polished windows of a cuvette at different wavelengths and is expressed in %. The measurement scale is 0-100%.

Z-dimension (Zd)

The Z-dimension – also known as Zd or centre height – is the height from the very bottom of the cell to the centre of its sample chamber. Particularly if the cuvette’s polished window is very small (in thiscase, usually the entire cuvette is black except for the window) it’s critical that the Zd of the cuvette be the same as the height of the instrument’s light beam. Generally, Zd is either 8.5, 15 or 20 mm, depending on the instrument’s manufacturer.

Only certain cuvettes depend on the correct Z-dimension and you’ll find the relevant information in their data tables. If you’re uncertain which Zd you need for your instrument, please ask us.


Materials and transmission characteristics

FireflySci uses only the highest quality materials for its products. Optical and Pyrex glass are manufactured by Schott (Germany), UV (ES) and IR quartz are from Shin-Etsu, the largest Japanese chemical company. The Lab-Club products also utilize optical and Pyrex glass from Schott and ES quartz from Shin-Etsu. The IR quartz is from Saint-Gobain.

Manufacturing methods

In addition to the use of high quality materials the manufacturing method plays the most important role in the production of excellent cuvettes. Most types of cuvette comprise four or five glass or quartz plates. These are fused together by fire, a process that demands skill and experience. The best method, utilized by FireflySci, is low-pressure thermofusion. Please note that the FireflySci products do not contain glass frit – this “glass cement” is used by some manufacturers of cheap cells and may be permeable. For flow-through cytometry cell uses it’s imperative that the components are fused. Our ES (UV) quartz cells for this application easily withstand the high pressure and high-performance lasers used in this application area. Polishing: after completion, the blank cells need to be polished. As hand-polished cells deliver very unreliable results, polishing at FireflySci is done in special rotating polishing machines. The better the polishing result, the higher the optical transmission of the finished product.


Handling and cleaning cells

General handling

Cells must be kept scrupulously clean. Optical surfaces should not be touched, as oil smudges are difficult to remove. Lens paper is the best thing to clean the cells with, anything else may scratch them.

Cleaning

Cleaning cuvettes carefully after use will ensure that their transmission characteristics remain fairly constant. Improper use or poor cleaning will result in staining or etching of surfaces thus reducing transmission and increasing light scattering. We recommend using a FireflySci cell washer, for example the FF-P65D for two cuvettes illustrated below, as the best and easiest way to clean your cuvettes. Please note that the FireflySci washers do not include the vacuum flask.

Solvents to use

The purity of solvents used for sample and for cleaning is important for the protection of cells. Only distilled water or spectrophotometric grade (highly pure) solvents should be used.

Cleaning agents to use

Depending on the type of sample you are testing, you may decide to clean the cells with a mild agent (such as dilute Hydrochloric acid) as soon as possible after each use. After an acid rinse, distilled water should be used to remove all traces of acid.

For organic materials

If you use mild detergents, we recommend that their pH value is as close to neutral as possible. These detergents should also be free of any suspended material containing lanolin or oils. You may also use dilute Hydrochloric acid. Rinse with the same solvent you originally used with the cell or with distilled water.

Stubborn contaminants

To remove stubborn contaminants, cells can be soaked for a few minutes in mild sulfuric acid. Dichromate cleaning solution is also very effective. Heavy metal salts may be removed by rinsing in concentrated nitric acid or aqua regia (a mixture of hydrochloric acid and nitric acid). Reminder: After an acid rinse, distilled water should be used to remove all traces of acid.

Drying cells

Don’t dry cells with compressed air. Speed up evaporation by using the cell washer as a means of suction.

Warning

Acids should never remain in the cells for longer than one hour. Never allow cells to soak in concentrated acids, alkalines, or other agents which may etch polished optical surfaces. Also, we do not recommend the use of ultra-sonic cleaners.

Cleaning cells with very small path lengths

Use a syringe to introduce the cleaning solvent to the corners and difficult to reach areas of the cell.

Detailed instructions for use

Quick start guide:

1. Check the Z-dimension of your spectrophotometer. The MFC's Z-dimension is 8.5 mm, so if your instrument has 15 or

20 mm Z-dimension, simply use the appropriate spacer.

2. Insert the MFC in the instrument and pull out the UV quartz micro chamber -this is a cuvette with a 1 mm lightpath

and 1.9 µL volume.

3. Inject the sample into the chamber and insert it into the MFC.

4. Run your experiment.

5. It's as easy as that!

Calibration Standards

Advantages of FireflySci standards

The FireflySci calibration standards are certified by the ISO-9001, ISO-17025 and ISO Guide 34 accredited National Research Council of Canada’s Metrology Laboratory. As far as we are aware, FireflySci is the only manufacturer offering certain types of standards. These are:

Combined standard for UV/VIS wavelength and UV photometric accuracy (FF-WAV-7-UV/VIS)

Combined standards for UV/VIS photometric accuracy and stray light performance (FF-FUV Dual Series)

Solid-state standard for VIS/NIR wavelength accuracy (FF-WAV-8-NIR)

Standards for continual UV/VIS/NIR photometric accuracy from 200 to 3000 nm (FF-SPB Series) The metal-on-quartz (nano-deposition technology) standards work with all spectrophotometers, regardless of the optics.

Recalibration: A long-term study of the FF-FUV Series products by the above-mentioned NRC laboratory confirmed that, after nearly 10 years of serial production, not one of the standards went off the manufacturer’s specification. There is no need to have FireflySci standards recalibrated due to age. If, however, recalibration is a requirement in your lab, the FireflySci ISO-17025 certified calibration lab can, of course, provide this service.

Firefly Scientific's equivalent of the NIST SRM 2034 calibration reference standard fulfills the requirements of the European

Pharmacopoeia (EP). Please note that is must be recalibrated annually. If you prefer a product that doesn't require recalibration,

Liquid Potassium Dichromate standards are primarily used by laboratories needing to conform either to the European

Pharmacopoeia (EP) or to cGMP regulations. Our portfolio of these standards ranges from individual standards that need to

be supplemented by the FF-HCL04-Blank through to kits that already include this item.

In many cases, validation has to be carried out every three months or after a repair. Should this repeat calibration NOT be

necessary in your lab, we recommend using our solid state FF-FUV series solid state standards that never need recalibration

due to material aging - with just one filter you can check the photometric accuracy and stray light performance at any point in

the 200-700nm range.

Assessing the photometric and wavelength performance of UV/VIS spectrophotometers is a necessary part of

keeping your laboratory in accordance with GLP, European Pharmacopeia (UP) or United States Pharmacopeia

(USP) standards.

The spectral bandwidth (SBW) refers to the physical slit-width of the spectrophotometer's monochromator, which

has a direct impact on the instrument's resolution. In turn, spectral resolution is the ability to resolve two

neighbouring peaks. Reconciling the resolution is an integral component in getting accurate wavelength and

photometric readings on your spectrophotometer.

Recognizing the need for periodic assessment of SBW and pursuant to EP and USP requirements, FireflySci has

expanded its range of calibration products to include this Toluene in Hexane standard.

Delivery includes:

High-purity solution of 0.02% Toluene in Hexane

High-purity Hexane blank

Suggested use:

1. The EP recommends comparing the peak maximum at 269nm with the peak minimum at 266nm.

2. Determine the resolution by running the Toluene in Hexane against the Hexane blank between 260 and

270nm and calculate the peak minima and maxima.

3. Finally, calculate the peak-to-trough ratios and compare the results with the instruments reference

values. If they are considerable lower, you may wish to contact your instrument manufacturer.

Please note that, in accordance with NIST guidelines, this standard must be recalibrated annually.

Stray light is defined as light that is detected by the spectrophotometer but does not belong to the given bandwidth

of the sample being measured. It often results from light scattering, diffraction or instrument errors and can result in

a variety of distortions and errors in analysis.

measurement is to be performed and then cut off at a specified wavelength.
