1. Examine the connections on the DI water and Wash 1 container. Practice disconnecting the tubing and sensor connections.
2. Examine the liquid levels for the DI water and Wash 1. (Refill the DI water and Wash 1 if needed.)
Empty Waste
1. Examine the connections on the Liquid Waste container. Practice disconnecting the tubing and sensor connections.

Check Calibration Status
1. Access the Reagent Compartment screen and check to see if any current reagent packs need to be calibrated.
a. How can you tell from this screen that a reagent pack needs to be calibrated?
2. If available, load a reagent pack with a new lot number. Check the Reagent Compartment screen again to view its calibration status.
a. How could you determine from this screen if a lot has ever been calibrated before?

Scan Master Curve Calibrator Assigned Value Cards
1. Locate the Master Curve card for the new reagent lot you will be calibrating.
2. Scan the Master Curve into the software, scanning the barcodes on the side that reads “Centaur CP”. Use the handheld barcode reader and scan each one of the barcodes until all are scanned.
If the scan window does not close automatically at the end of the scan, select Cancel and scan again.

3. Calibrate the new lot of reagent if available. If a new lot is not available, choose one with an
expired calibration or one that will be expiring soon.
4. Select the correct calibrator for that assay.
5. Locate Calibrator Assigned Value card in the calibrator box.
6. Scan the Calibrator Assigned value card on the side that reads “Centaur CP” into the software
using the handheld scanner. and scan each one of the barcodes until all are scanned.

Start the Calibration Run
1. Load the rack with calibrators into the sample compartment. Check to make sure that the barcodes were read.
2. Select the assay that you want to calibrate.
3. Under the Calibration side, click on “Calibrate”.
4. Initialize the calibration process.

Review Calibration Data
1. When the calibration run is complete access the Results Current Calibrations tab to see the status.
a. What should the status be if the calibration run has passed?
b. What would you do if the status was Invalid?
c. What would need to be done if the status was Verify?
2. Select the calibration you just performed and open the Details window to view the data.

1. Access the QC Definition tab. View it to see if there are any currently defined controls. Obtain an Expected Value sheet i.e. Bio-Rad control Ranges or Siemens Control Ranges.
2. Define and save at least one new control. As you are going through the procedure, answer the following questions:
a. What happened in the “Control Barcode” field when you typed into the “Lot” field?
b. What happened to the “Mean and “2SD” fields when you enter the High and Low ranges?

3. Select “Close” and click on “Start” to start the run.

1. Close the Sample Compartment window and select the Results button. Select the Date column to point the arrow UP. Your new orders should be at the top of the list.

2. When the LED is neutral (indicating it is safe to remove the rack) open the sample compartment door and remove it.

Shutdown System
1. Make sure samples are finished processing and system status is Ready.
2. Turn analyzer power switch to OFF.
Clean System Interior
1. Prepare Cleaning Solution.
a. Dilute one bottle of Concentrated Cleaning Solution to 2000 mL with DI water.
2. Open the top cover.
3. Clean incubation ring cover:
a. Move sample and reagent probe mechanisms out of the way.
b. Wipe incubation ring cover with gauze saturated with cleaning solution.
c. Rinse incubation ring cover with gauze saturated with water.
d. Dry incubation ring cover with lint-free tissue.
4. Clean exterior of reagent probe:
a. Lift reagent probe to highest point.
b. Check probe for damage.
c. Wipe downward with alcohol wipe.
d. Reposition probe over waste bowl.
5. Clean exterior of waste probe:
a. Wipe probe with alcohol wipe.
6. Clean probe rails:
a. Wipe all probe rails with alcohol wipes.
b. Allow to dry before powering on.
Update Maintenance Log:
1. Update Maintenance Log:
o Select Maintenance.
o Select System Interior.
o Select Perform.
o Enter Comment as needed.
o Select OK.

Perform Automated Weekly Cleaning
1. Make sure system is in the Ready state.
2. Prepare DI Water container:
o Disconnect DI Water container from the system and dispose of water.
o Pour dilute cleaning solution into DI Water container, replace cap, and rock 10 times.
o Let container sit for 10 minutes.
3. Start the automated procedure:
o Select Maintenance.
o Select Automated Weekly Cleaning.
o Select Perform.
4. When procedure 1 displays:
o Connect DI water container with cleaning solution to system.
o Select Accept.
5. When procedure 5 displays:
o Disconnect DI Water container and dispose of cleaning solution.
o Fill DI Water container with 3 liters of DI water and rock at least 5 times to rinse.
o Dispose of water and repeat rinsing at least 3 times.
o Fill DI Water container with fresh DI water and connect to system.
o Select Accept.
6. When procedure 9 displays, select OK.

Exporting data 1. Select Historical.
2. Select Export.
3. Select Selected Records, Today’s Records, All Records, Range or Run ID.
4. Leave File selected.
5. Leave TAB selected. This file can be viewed with Excel, NotePad, or WordPad.
6. Select File Location.
o Find the location to save the exported file – hard drive, floppy drive, USB stick, and CD.
o Type file name and file extension. The software does not automatically add the file extension to the file name. If you forget to add the extension at this point, you can always add it later on another PC.
o Select Save.
o Select OK.
You will see the progress of the save on the bottom left of the Export Results screen.
When the export is complete, the Export Results screen will close.
