ISSX President’s Message
Volume 33 Issue 4, 2013
At the end of December my term comes to a close. It is a good time to reflect on the activities of the Society, challenges we face, and hopefully good decisions made that will have an impact on the future of ISSX.
At the 2013 10th International ISSX Meeting, I thanked Officers and Council members who are also completing their terms in 2013. This included Charles Crespi (Treasurer), Brian Houston, and Ikumi Tamai. During my term I served with Officers and Council members who will continue to serve or were reelected and this group included John Miners (President-Elect), Andrew Parkinson (Treasurer-Elect), Thomas Baillie, Ann Daly, Richard Kim, Paul Ortiz de Monellano, and Hiroshi Yamazaki. Another important group to thank is the ISSX Office professionals at SmithBucklin which include Steve Kemp (Executive Director), Zoë Fuller, Kelly Marks, Alex Zapple, and Sarah Langan. This was a fantastic group of Officers, Council members, and ISSX staff to work with and I
offer my deepest gratitude for the opportunity to serve with them to do the Society’s important work. Finally, I want to offer my warm welcome to newly-elected Officers and Council members: Geoff Tucker (President-Elect), James Halpert (Treasurer-Elect), Lawrence Marnett, and Maria Almira Correia. ISSX needs your experience, creativity, and passion.
With changes in the economy, the pharmaceutical industry, and funding for academia, ISSX had to adapt. It has taken a few years to reach a new steady state. During this time we experienced challenging times with a decline in membership and in the number of attendees at our meetings. However, I am happy to report that all the indicators show that we have stabilized. During this time with the help of Charlie, Andrew, and Steve we have managed the Society’s finances in a way that meets the mission of ISSX and still retains a strong reserve fund for strategic investment in new initiatives and covers ISSX for unanticipated expenses. We used this
time of stability to develop a strategic plan for ISSX that is being implemented over the next 3-5 years. Key themes in the plan include increasing participation and member value, expanding scientific reach, increasing international scope, improving operational excellence, and improving financial sustainability. We have only just started on this plan, but my hope is that John Miners, the incoming President, and the new Council will continue this work.
Another job has been to plan new meetings for the future. Over my term, we added the 20th North American Meeting in Orlando, FL, USA (2015), 13th European Meeting in Glasgow, Scotland, UK (2015), and the 21st North American Meeting in Providence, RI, USA (2017) to the meeting calendar. The Orlando meeting will be chaired by Hartmut Derendorf (University of Florida), the

As mentioned in the preface, “this volume deals primarily with organic chemicals in the aquatic and terrestrial environments, including products from atmospheric transformations” (page xxv). Now in its second edition, this large work is packed with useful and pertinent information concerning the potential fate of chemicals that have made their way, usually unintentionally, into the surrounding biosphere. Accepting that contamination or pollution frequently involves many different chemical entities, although one type may be present at higher concentration than the background plethora, the effects of this “chemical soup” on microorganisms are discussed.
The book is conveniently divided into three sections. Section one (Degradation and Transformation Processes) contains four chapters (Abiotic reactions: An outline, Biotic reactions: An outline of reactions and organisms, Mechanisms,
and Interactions) which provide a broad perspective of chemical and biochemical reactions that a compound may undergo, whether assisted by biological catalysts or not. Perturbations brought about in microbes living in “stressful” situations (physical, chemical) and the potential metabolic consequences of this are also discussed.
Section two (Experimental Procedures), containing two chapters (Experimental procedures and Procedures for elucidation of metabolic pathways), describes practical techniques employed to investigate microbial populations, to establish metabolic pathways, and to determine the structure of metabolites produced.
The final section (Pathways and Mechanisms of Degradation and Transformation) gives details of biochemical reactions involved in the metabolic transformation of a range of substrate classes, with information concerning the enzymology involved. Five chapters make up this larger section (Aliphatic compounds, Carbocyclic aromatic compounds without halogen substitution, Halogenated arenes and carboxylates with chlorine, bromine, or iodine substituents, Heterocyclic aromatic compounds and Miscellaneous compounds).
As is typical with such books, the layout is neat and the print easy to read. Many chemical schemes and copious references after (and within) each chapter add to the work. Good quality paper, firm binding, and an overall solid construction ensure that this book will remain durable for many years. For anyone with an interest in xenobiotics and environmental issues this book certainly is a useful reference source.

Call for Nominations Nominations are now being accepted for the awards to be presented at the 5th Asia Pacific ISSX Meeting. Visit www.issx.org/Awards/Nominations to submit a nomination. The two awards to be presented at this meeting are: Asia Pacific Scientific Achievement Award -‐ Presented to an ISSX member who has made major scientific contributions to the field in the Asia Pacific region. The purpose of this award is to recognize meritorious contributions by senior or mid-‐career scientists that have had a major impact on research in the field. The award will consist of a plaque or medal, an honorarium, and travel support to the 5th Asia Pacific Meeting in Tianjin, China where a lecture will be presented by the awardee. Regional New Investigator Award -‐ Presented to an ISSX member who has made significant contributions to the field during their early career years (normally within 5-‐10 years from the time of receiving his/her highest-‐earned degree). The purpose of this award is to encourage and recognize developing scientists who are active in the field within the Asia Pacific region. The award will consist of a plaque or medal, an honorarium, and travel support to the 5th Asia Pacific ISSX Meeting in Tianjin, China.

Dr. Allan H. Conney, of Rutgers University, one of the most-cited scientists in pharmacology, cancer research, and drug metabolism, died on September 10, 2013 at the age of 83. Born in Chicago, Dr. Conney received his B.S. in pharmacy (1952) and his Ph.D. in oncology (1956) from the University of Wisconsin. Subsequently, he joined Dr. B.B. Brodie’s group at NIH to study enzyme induction and drug metabolism. From 1960 to 1987, he was affiliated with Burroughs-Wellcome and Hoffmann-La Roche. In 1987, Dr. Conney joined Rutgers University and established the Department of Chemical Biology at the Ernest Mario School of Pharmacy. He held the title of William and Myrle Garbe Professor of Cancer and Leukemia Research, State of New Jersey Professor of Pharmacology, and the Director of the Susan Lehman Cullman Laboratory for Cancer Research.
Dr. Conney’s research work spanned over 50 years. He was still working on the day of his passing. He made major contributions to our basic knowledge of drug metabolism and the building of foundations of this area of research. His illustrious research career began when he was a graduate student with Drs. James and Elizabeth Miller at the McArdle Institute. In studies on the investigation of the metabolism of carcinogenic aminoazo dyes and polycyclic aromatic hydrocarbons, he discovered the induction of xenobiotic-metabolizing enzymes in rats, believed to be the first demonstration of cytochrome P450 induction in mammals. In subsequent years at NIH, Burroughs-Wellcome, and Hoffmann-La Roche, he continued the work on enzyme induction and the influence of P450 induction and inhibition on the metabolism of therapeutic agents and carcinogens in animals and humans, long before drug-drug interactions were recognized as an important issue in drug discovery and development. His 1967 review article on “Pharmacological implications of microsomal enzyme induction” (Pharmacological Reviews, 19: 317-366) was one of his most-cited articles in pharmacology in the 1960s and 1970s. During this period, he and his colleagues studied various factors regulating the metabolism of drugs, endogenous steroids and hormones, carcinogens, environmental chemicals, and the cytochrome P450 system. These studies contributed to the discovery of multiple forms of cytochrome P450 enzymes and eventually the purification and characterization of many P450 enzymes. A more detailed account of this work can
be found in his 2003 review article on “Induction of drug-metabolizing enzymes: a path to the discovery of multiple cytochromes P450” (Annu. Rev. Pharmacol. Toxicol. 43:1-30). Another important milestone in Dr. Conney’s research was the collaborative studies with Dr. Don Jerina of NIH on the
metabolism and carcinogenesis of polycyclic aromatic hydrocarbons. The NIH team and the Roche group made the most thorough and systemic studies on the metabolism, mutagenesis, and carcinogenesis of many of the metabolites of a number of polycyclic aromatic hydrocarbons. These studies led to the establishment of the bay region diol epoxide theory to explain the carcinogenesis of this class of compounds.
Since joining Rutgers University in 1987, Dr. Conney had devoted the last 26 years to cancer prevention research. These studies include in vitro and in vivo experiments as well as clinical trials. In recent years, his laboratory discovered that caffeine and exercise inhibit skin carcinogenesis, and the combination of Lipitor and Celebrex inhibit prostate and pancreatic cancer progression. This body of work contributes greatly to our understanding of how cancer prevention can be achieved by simple steps and more acceptable lifestyle changes.
Dr. Conney had a very productive research career. He published more than 500 articles, many of which are very highly-cited. He was elected to the National Academy Sciences in 1982 and served as President of ASPET in 1983-1987. He was honored with many prestigious awards in pharmacology, pharmaceutical sciences, toxicology, and cancer research.
The fields of drug metabolism, pharmacology, and cancer research lost a good friend and a pioneering researcher. He was a brilliant scientist and a wonderful person. He always treated his colleagues with respect and helped many individuals in their career development, particularly young researchers. We will always remember him as a warm, gentle, and scholarly friend.
IN MEMoRIAMRonald W. Estabrook (1926-2013)It is with sadness that ISSX announces the passing of Ronald W. Estabrook, Professor Emeritus, Department of Biochemistry, UT Southwestern Medical Center.
A pioneer in the study on the cytochromes of mitochondria, DMPK, Ronald W. Estabrook was a member of the ISSX Council from 1984 to 1987. He was elected as the fourth President of ISSX, serving in that position from 1988 – 1989. He remained involved in ISSX matters long after his term ended and attended many Council meetings as Past-President and advisor to the Society. He was an ardent photographer and was the unofficial historian of ISSX. His online catalog of photographs from ISSX meetings and other events is extensive.
Dr. Estabrook received his undergraduate degree from the Rensselaer Polytechnic Institute in 1950. He earned his Ph.D. from the Rochester Institute of Technology in 1954. His first breakthrough was as a professor at the University of Pennsylvania, when he and two colleagues discovered the properties of a hemoprotein molecule known as cytochrome P450, demonstrating that P450 plays a crucial role in how the body metabolizes various substances.
To honor his service to ISSX and his scientific achievements, XenoTech permanently endowed the North American Scientific Achievement Award in Honor of Ronald W. Estabrook. Dr. Estabrook himself was one of the first scientists ever honored by ISSX. In 2001 he was presented with the ISSX Award for Distinguished Service, as it was previously named.
Dr. Ronald W. Estabook was born in Albany, NY on January 3, 1926. When he was 17, he left New York to join the Navy, attending officer’s training school at Princeton, and then serving as navigator on a ship sweeping minefields. The young mine sweeper then returned to school and earned a doctorate in biochemistry from the University of Rochester.
He would go on to become an eminent researcher, chairman of biochemistry at UT Southwestern for 14 years, and the first dean of UT Southwestern Graduate School of Biomedical Sciences. He played a major role in turning UT Southwestern into an internationally-respected research center that would attract many of the nation’s brightest and best scientists and students.
In 2006, at the age of 80 years, Dr. Estabrook retired and was named the University of Texas System Ashbel Smith Emeritus Professor Biochemistry. He will be fondly remembered by his many colleagues, students, friends, his wife June, and his children and grandchildren.

Glasgow meeting will be co-chaired by Roland Wolf (University of Dundee) and Ann Daly (Newcastle University), and finally the Providence meeting chair will be Jashvant Unadkat (University of Washington). We have great chairs and meeting organizing committees to develop terrific programs for these upcoming meetings. I look forward to seeing these conferences take shape and attending them myself in a few years.
Another passion during my term has been to support the international mission of ISSX. During my terms as Secretary, President-Elect, and President I have been a strong supporter of the growth of ISSX in the Asia-Pacific region. A key focus was developing sustained relations with the leaders of our affiliate organization, the Chinese Society for the Study of Xenobiotics (CSSX). I helped organize and participated in workshops and conferences with CSSX in Shanghai, Guangzhou, Xi’an, Tianjin, Nanjing, and Kaifeng from 2010-2013. I am proud that I was able to contribute to the vision of ISSX Past-President, Yuichi Sugiyama, to grow the activity of ISSX in China and the AP region. Additionally, we recently started work in India on a small scale to build our presence in a country where there are many scientists in universities and companies interested in xenobiotic research.
In closing my final ISSX Newsletter contribution, I want to say that it has been a privilege to serve the ISSX membership during my term. I will cherish the memories of all the new friendships formed, exciting events, and even the occasional stressful situations I encountered during my term. I look forward to the future meetings of ISSX and I hope we can meet to discuss interesting scientific developments in the fields of xenobiotic research.
